2.Effect of ischemic preconditioning on the apoptosis of hepatocytesand expression of HIF-1? and Survivin in rats

Objective:To observe the effect of ischemic preconditioning(IP)on hepatocellular apoptosis caused by ischemia-reperfus ion(IR)injury and expression of apoptosis regulatory genes(Survivin,HIF-1?)protein,to explore the protective mechanisms of IP on IR injury in rat liver.Methods:Ninty SD rats were randomly divided into sham operation(SO)group,ischemia-reperfusion(IR)group,ischemic preconditioning(IP)group.In SO group,the hepatoduodenal ligament of rat was only pulled and separated,and then closed the abdomen;IR group subjected to 30 minutes of ischemia;IP group was achieved by ischemia for 5 minutes,followed by reperfusion for 5 minutes before continuous to ischemia for 30 minutes.Ten rats of each group were killed after reperfusion for 2,6,12,24 hours respectively,then liver tissues were taken out.The rats in SO group were killed after operation for 2 hours.To detect the Apotosis index(A)Iand Survivin,HIF-1? protein levels.Results:AI of hepotocytes significantly increased in IR group and IP group than that in SO group(P
3.Reliability and Validity of Chinese Version of Comprehensive Frailty Assessment Instrument

Objective To translate the English version of Comprehensive Frailty Assessment Instrument (CFAI) into Chinese and evalu-ate its reliability and validity. Methods CFAI was translated into Chinese with the Brislin's translation mode. From May to July, 2015, 200 community aged people in Shijiazhuang, China were assessed with the Chinese version of CFAI and World Health Organization Quality of Life-BREF (WHOQOL-BREF), and 30 of them were assessed with CFAI again after a week. The content validity was rated by six experts of gereology. The total scores of CFAI and WHOQOL-BREF were analyzed with Pearson's correlation, as well as the scores of CFAI test and retest. The CFAI were analyzed with Factor Analysis. The Cronbach's α was tested. Results There were 23 items in 4 dimensions. The content validity index was 0.782. Six factors were extracted by Factor Analysis and the cumulated variance was 64.05%. The total score of CFAI correlated with the score of WHOQOL-BREF (r=-0.764, P<0.001). The Cronbach's α was 0.704~0.897 in dimensions (P<0.001), and the r= 0.604~0.941 (P<0.001) between test and retest. Conclusion The Chinese version of CFAI is reliable and valid, that can be applied to evaluate frailty in community aged people.
4.Expression of TGF-βRⅡ and fibronectin in developing mouse kidney

ObjectiveTo observe the chronological and spatial expressions of transforming growth factor-β receptor typeⅡ(TGF-βRⅡ)and fibronectin (Fn )in developing mouse kidney and investigate the relationship between the expressions and kidney development. Methods Immunohistochemistry and Western blotting were used to observe and measure the expression of TGF-βRⅡ and Fn in embryos and postnatal mouse kidneys. Results The expression of TGF-βRⅡ was noticed in all stages of glomeruli, renal tubes and collecting ducts, and increased gradually with ages; The expression of Fn was noticed too in all stages of glomeruli, renal tubes and collecting ducts, except for the comma-shaped bodies, and also increased gradually with ages. Conclusion TGF-β and Fn might play an important role in the development and maturation of mouse kidney.
5.Antigen-free biological meniscus transplantation for repairing meniscus deficiency in goats

BACKGROUND: The meniscal allograft transplantation has been proved good outcome in clinical, however, the source and matching restrict its application. By means of a patent technique of separating antigen, a new biological meniscus can be obtained.OBJECTIVE: To observe the histological changes of transplanted biological meniscus in goat's knee joint, in addition, to investigate the feasibility of reconstituting meniscus by using biological meniscus. DESIGN, TIME AND SETTING: The animal experiment of goats was performed at the Central Laboratory in the Third Affiliated Hospital of Sun Yat-sen University between June 2005 and June 2006.MATEAIALS: A total of 15 healthy goats were used in the experiment. Fresh porcine meniscus was harvested for the preparing of biological meniscus.METHODS: The medial meniscus of goat was excised, and then the absent part was repaired using biological meniscus. MAIN OUTCOME MEASURES: The animals were sacrificed at months 1, 3, 6 and 12 after operation. And the samples were detected by histology, scanning electron microscopy (SEM) and Indian ink perfusion. RESULTS: The incision of all animals healed well, without infection or death. The transplanted meniscus healed well with the surrounding tissue, and the connection was tight. With time prolonged, the transplanted meniscus slowly soaked, and live cells existed inside the meniscus, but there were no live cells in about 1/4 district at 1 year after the surgery. Indian ink perfusion showed that the surrounding tissue grew inside and set up microcirculation, but only limited to 10% periphery of the meniscus (normal goat approximately 15%). The transplanted meniscus began the necrosis at 6 months after surgery. Under SEM, the surface of the meniscus became crude and cracked in some places, but still maintains the gross shape at 1 year after the surgery.CONCLUSION: There is no obvious rejection after the biological meniscus was transplanted into knee joint of goats. During the process of absorption, autologous tissue of goats can grow into and set up microcirculation in the transplanted meniscus. The transplanted meniscus protects the'knee cartilage notably, only slightly damage in cartilage can be observed.
6.β-Catenin,β-Trcp and CRD-BP in colorectal cancer

Wnt/β-catenin signaling pathway plays an important role in the initiation and progression of colorectal cancer. The key to its carcinogenicity is related to the inhibition of β-catenin degradation, and the subsequent cytoplasmic accumulation and nuclear translocation of β-catenin, which activates the transcription of downstream target genes such as coding region instability determinant-binding protein (CRD-BP). In cancercells ,the expression of CRD-BP mediates the stabilization of β-Trcp, which in normal cells regulates the degradation of β-catenin. The activation of Wnt/β-catenin signaling pathway, overexpression of β-catenin and CRDBP, and the upregulation of β-Trcp by CRD-BP play important roles in the initiation, progression, and metastasis of colorectal cancer.
7.Effect of patient-controlled intravenous analgesia with dexmedetomidine mixed with sufentanil on sleep quality after spinal surgery

Objective To investigate the effect of patient-controlled intravenous analgesia (PCIA) with dexmedetomidine mixed with sufentanil on the sleep quality after spinal surgery.Methods Eighty patients of both sexes, aged 21-72 yr, weighing 48-100 kg, of American Society of Anesthesiologists physical status Ⅰ or Ⅱ , scheduled for elective spinal surgery, were randomly assigned into sufentanil group (group S) and dexmedetomidine mixed with sufentanil group (group DS) with 40 cases in each group.In group S, PCIA solution contained sufentanil 2 μg/kg and tropisetron 6 mg in 100 ml of normal saline.In group DS, PCIA solution contained sufentanil 2 μg/kg, dexmedetomidine 3 μg/kg and tropisetron 6 mg in 100 ml of normal saline.The PCA pump was set up with a 0.5 ml bolus dose, a 15 min lockout interval and background infusion at a rate of 2 ml/h.Sufentanil 5 μg injected intravenously and celecoxib 200 mg given orally were used as rescue analgesics, and numeric rating scale (NRS) score at rest was maintained ≤4 within 48 h after surgery.Ramsay sedation scores were recorded at 1, 2, 6, 12, 24 and 48 h after surgery.Sleep quality was evaluated using the Medical Outcomes Study Sleep Scale (MOS-SS), and the average time of daily sleep and Sleep Problems Index (SPI) were recorded at week 1 before and after surgery.Patient's satisfaction with sleep was assessed on the night of surgery and 1st day after surgery.The requirement for rescue analgesics was recorded.Results There was no significant difference in the requirement for rescue analgesics between group S and group DS (P>0.05).Compared with the value before surgery, the average time of daily sleep was significantly shortened, and SPI was increased at week 1 after surgery in group S (P<0.05 or 0.01), and no significant change was found in group DS (P>0.05).Compared with group S, the Ramsay sedation scores were significantly increased at 1, 2, and 6 h after surgery, the average time of daily sleep was prolonged at week 1 after surgery, the SPI was decreased at week 1 after surgery, and the degree of satisfaction with sleep was increased on 1st day after surgery in group DS (P<0.05 or 0.01).Conclusion PCIA with dexmedetomidine mixed with sufentanil is helpful in improving the sleep quality after spinal surgery in the patients.
8.Cyclic mechanical stretch influences cell adhesion and spreading of immortalized human keratinocytes

BACKGROUND:The mechanical environment of skin tissue and spreading state of epithelial cels are closely related with the wound healing and scar formation process. OBJECTIVE:To analyze the effect of extracelular mechanical stimulation on cel spreading, to test the cel proliferation in order to analyze the effect of spreading form on cel proliferation and other physiological activities. METHODS: Cyclic sine wave mechanical stretching was exerted on immortalized human keratinocyte by using FX-4000 flexible substrate loading system, on the condition of 0.2 Hz and at frequency of 10% amplitudes. The spreading form was compared at 0, 24 and 48 hours, the cel proliferation was analyzed with flow cytometry, and the distribution of vinculin was analyzed with immunofluorescence staining. RESULTS AND CONCLUSION: human keratinocyte would keep the spreading state and could induce more cel proliferation by 24 hours mechanical stretching stimulation. Conversely, after stimulated for 48 hours, the morphology of the human keratinocyte was significantly changed, and the number of human keratinocyte in the division stage was larger than that in the static control group; under tensile stress, the distribution of vinculin was transformed from the surrounding nucleus membrane area to the cel edge. The results indicate that proper mechanical stimulate can increase cel proliferation with keeping cel spreading and adhesion state; the stimulating time of continuous cyclic stretching is the major factor to determine cel spreading morphology and adhesion regions of immortalized human keratinocyte.
9.Determination of oridonin and rosemarinic acid in Donglingcao Tablets by HPLC with changing ultraviolet-visible wavelength

AIM:To establish the method for determining oridonin and rosemarinic acid in Donglingcao Tablets(Rabdosia rubescens(Hemsl.) Hara). METHODS:Oridonin and rosemarinic acid were determined by HPLC simultaneously with changing ultraviolet-visible wavelength.Chromatographic condition was composed of ODS-C_(18) column,methanol-03.% phosphoric acid (40∶60),UV detection wavelength of oridonin at 238 nm and of rosemarinic acid at 329 nm. RESULTS:The average recoveries were 97.8% and 96.7% and related standard deviations(RSD) were 1.9% and 2.3%. CONCLUSION:This method can be used to determine simultaneousely oridonin and rosemarinic acid in Donglingcao Tablets.
10.Clinicopathological features of oncocytic mucoepidermoid carcinoma

Objective To investigate the clinicopathological features,immunophenotype,diagnosis and differential diagnosis of oncocytic mucoepidermoid carcinoma(OMEC).Methods A case of OMEC was studied for clinicopathological and immunohistochemical features,the available literature on this subject was reviewed as well.Results Magnetic resonance imaging revealed a well-defined mass,measured 1 cm×1 cm,within the right submandibular gland.The patient was underwent the total mass and the right submandibular gland resections.On macroscopic examination,the well-defined tumor was gray-yellowish on cut surface with hard consistence and was sized 1 cm×1 cm×0.8 cm.The uncapsulated tumor was mainly solid with focal microcysts on cut surface.Microscopically,the tumor consisted of nests,sheets,and occasional microcysts of oncocytic cells.Focally,intermediate cells,mucilage cell and epidermoid cell were also present.Immunohistochemically,the oncocytic cells were diffusely and intensely positive for CK(AE1/AE3),EMA and P63.By histochemical staining,the oncocytic cells were positive for PTAH staining,and mucilage cell were positive for PAS staining.Conclusion The OMEC is an exceedingly rare variant of mucoepidermoid carcinoma with a good prognosis and should be differentiated from oncocytoma,oncocytic carcinoma,Warthin's tumor with mucinous metaplasia,pleomorphic adenoma,oncocytic myoepithelioma and myoepithelial carcinoma,et al.
