To investigate the inhibitory effects of siRNA expression vector on proto-oncogene Pokemon expression in SW480 cells and to provide experimental basis for further research about the biological function of Pokemon.siRNA expression vectors were constructed to express a short hairpin RNA against Pokemon.The recombinants were transfected into SW480 cells with liposome.Cellular morphology and transfection efficiency were observed.The expression of Pokemon were checked by real-time fluorescence quantitative PCR and Western blot.MTT assay was used to detect the effect of siRNA on the growth of SW480 cells and Fluorescence Activated Cell Sorter was applied to study cell apoptosis of SW480 cells.The transfection efficiency was about 36% and the cellular morphology changed greatly at 48h after transfection.siRNA expression vectors could specifically reduce the expression of Pokemon mRNA and protein in SW480 cells.Compared with negative control,the inhibition ratio of Pokemon mRNA expression was 34.2% and 67.7%,in 24th hour and 48th hour,the inhibition ratio of Pokemon protein was 48.3% and 73.6%,in 48th and 72th hour,respectively.Transfection with Pokemon siRNA expression vectors could inhibit cell growth and induce cell apoptosis of SW480 cells.siRNA expression vectors were successfully established that could effectively inhibit the expression of Pokemon in SW480 cells.Pokemon gene silencing could decrease growth and increase apoptosis of SW480 cells.

Recent years,progress has been made on the basic researches and clinical applications of ocular surface reconstruction with autologous or allogeneic limbal stem cells,oral mucosa epithelium and ex vivo cultured limbal stem cells.However,there are several issues,including the successful treatment for severe ocular damage,longterm follow-up and evaluation of clinical outcome,and the in vivo tracking of donor stem cells,remained to have definitive conclusions.Future studies should address the questions and challenges based on the basic research of limbal stem cell deficiency and standardized evaluation of clinical outcome.

This article applied 1H-NMR-based metabolic technology combined with pattern recognition method to study metabolic changes of brain frontal cortex among spontaneously hypertensive rats ( SHR ) and evaluate Pingan prescription treatment effect . It was aimed to explore the valuable information between hypertension and brain damage in order to provide a scientific basis for the mechanism of Pingan prescription . Rats were ran-domly divided into four groups , which were the normal control group , hypertension model group , captopril group and the Pinggan prescription group . Medications were administered continuously for 14 days . Then , the
frontal lobe tissues of rats in each group were removed . The 1H-NMR based metabolic technology combined with pattern recognition method were used in the detection and analysis of frontal lobe tissues of rats from four group in order to find characteristic metabolites . The results of the principal components analysis ( PCA ) and partial least squares discriminant analysis (PLS-DA) showed that captopril group and Pinggan prescription group were significantly different from the hypertension model group . And the classification tendency of rat sam-ples from the Pinggan prescription group and the captopril group were obvious , which suggested that Pinggan prescription can obviously improve the rat's overall metabolism. The four metabolites were identified as glucose, galactose , dopamine and succinic acid . It was concluded that hypertension damaging frontal tissues may mainly express in energy metabolism and nerve damage . And Pinggan prescription can effectively reduce blood pressure and relieve metabolic abnormalities due to disease through the influence on its metabolites .

[ABSTRACT]OBJECTIVETo assess the efficacy of postauricular injection of shuxuening combined with lidocaine in treating intermediate and high-frequency sudden hearing loss.METHODS48 patients with intermediate and high-frequency sudden hearing loss were randomly divided into postaurical injection group (n=24) and control group (n=24). The control group was given hormone, microcirculation improvement, nutrition of nerve and other conventional treatment. The postauricular injection group received 3 doses of postauricular inject of 0.5ml shuxuening and 0.3ml lidocaine tertian injection in addition to conventional treatments.RESULTSTwo weeks after administration, the total effective rates of postauricular injection group 87.5%were significantly higher than that of the control group 62.5%. Pure tone average improved (20.74±15.19)dB on average in the postauricular group, and (12.44±9.12)dB in the control group. The difference was statistically significant (P<0.05). CONCLUSIONPostauricular injection of shuxuening combined with lidocaine treatment for intermediate and high-frequency sudden hearing loss has better curative effect than that of the conventional treatment.

Abstract: Objective　To analyze the drug resistance of Staphylococcus aureus in blood samples of children and adults from 50 hospitals in Shandong Province, and to understand the drug sensitivity characteristics of Staphylococcus aureus bloodstream infection (BSI), so as to provide reference for clinical experience. Methods　The distribution and drug resistance of Staphylococcus aureus isolated from blood samples from 50 hospitals in Shandong province from 2017 to 2020 were analyzed based on the Cooperative Research Network of Pediatric Bacterial and Fungal Resistance Monitoring in Shandong Province. Meanwhile, the drug sensitivity characteristics of Staphylococcus aureus were divided into children group (<14 years old) and adult group (≥14 years old). The data were analyzed by Whonet 5.6 and SPSS 22.0 with reference to CLSI 2021 M100 document standard judgment results. Results　A total of 3 661 Staphylococcus aureus strains were collected from 50 medical institutions in Shandong Province, including 675 in 2017, 870 in 2018, 1 080 in 2019, and 1 036 in 2020. The drug resistance rates of multiple antibiotics in blood culture methicillin-resistant Staphylococcus aureus (MRSA) group and methicillin-sensitive Staphylococcus aureus (MSSA) group were significantly different (P<0.05). There were significant differences in antibiotic resistance rates between adult group and children group (P<0.05). The overall detection rate of MRSA was 27.5%, and no staphylococcus aureus strains resistant to vancomycin, linezolid and tigecycline were found. Conclusion　The detection rate of MRSA strains decreased continuously and increased by 2020. The detection rate of MRSA in adult group was lower than that in children group, suggesting that we should pay attention to the monitoring of bacterial resistance in children group, to the management of multiple resistant bacteria and rational use of antibacterial drugs.

Objective: It was to investigate the content alteration of the tannins in cibot rhizome and its processed samples. Method: The casein method was used to determine the content of tannins. Results: The content of tannins decreased after the medicinal material was processed. Conclusion: Processing may decrease the content of tannins in cibot rhizome. The raw was better than the other processed samples if the tannins were used as active constituents.

Objective To observe the influences of NaAsO2 on H3K36me3 modifications,mRNA transcription of O6-methylguanine-DNA methyltransferase gene (MGMT) in HaCaT cells,and to explore the relationship between the transcription of MGMT gene regulated by H3K36me3 and DNA damage induced by arsenic,in order to provide new ideas and scientific basis for prevention and intervention of arsenism.Methods HaCaT cells were treated with 1.25,2.50,5.00 and 10.00 μmol/L NaAsO2 for 24 h,and were also treated with 10.00 μmol/L NaAsO2 for 6,12 and 24 h.HaCaT cells that treated with 0.00 pmol/L NaAsO2 and 0 h were used as blank control group.The degree of DNA damage in peripheral blood cells was detected by single cell gel electrophoresis.The level of H3K36me3 modifications was detected using Western blotting.Quantitative real-time polymerase chain reaction was used to detect the mRNA levels of MGMT gene.Quantitative chromatin immuno-precipitation was used to detect the level of H3K36me3 modifications in the coding regions (ChIP1 and ChIP2) of MGMT gene.Results ①Among the groups of HaCaT cells treated with 2.50,5.00 and 10.00 μmol/L NaAsO2,the levels of tail DNA％ (11.83 ± 1.15,16.85 ± 2.52,24.23 ± 2.75) and olive tail moment (OTM,10.90 ± 1.13,16.19 ± 2.26,23.83 ± 2.79)were significantly increased compared with those of the control group (0.00 μmol/L,2.40 ± 0.51,2.26 ± 0.40,all P ＜ 0.05).After treated with 10.00 μmoFL NaAsO2 for 12 and 24 h,compared with the control group (0 h,3.66 ± 1.02,3.38 ± 1.00),the degrees of tail DNA％ (15.51 ± 1.92,24.18 ± 2.42) and OTM (13.58 ± 2.04,23.14 ± 2.11)were significantly increased (all P ＜ 0.05).②Compared with the control group (0.00 μmol/L,100.00 ± 0.00),the levels of H3K36me3 modifications (60.59 ± 9.75,57.82 ± 11.28,39.45 ± 7.09) were lower at the dosages of 2.50,5.00 and 10.00 μmol/L NaAsO2 (all P ＜ 0.05).Compared with the control group (0 h,100.00 ± 0.00),the levels of H3K36me3 modifications (48.47 ± 9.67,47.75 ± 6.98) were lower after treated with 10.00 μ mol/L NaAsO2 for 12 and 24 h (all P ＜ 0.05).③The levels of H3K36me3 modifications in HaCaT cells exposed to different doses of NaAsO2 were negatively associated with the tail DNA％ and OTM (r =-0.897,-0.903,all P ＜ 0.05).④Compared with the control group (0.00 μmol/L,100.00 ± 0.00),the mRNA levels of MGMT gene were lower at the dosages of 2.50,5.00 and 10.00 pmol/L NaAsO2 (78.20 ± 3.50,61.40 ± 2.60,49.15 ± 4.70,all P ＜ 0.05).⑤There was no observed H3K36me3 enrichmem regularity in the gene encoding ChIP1 and ChIP2 regions of MGMT gene in all doses of NaAsO2 groups (all P ＞ 0.05).Conclusions H3K36me3 may be involved in the regulation of arsenicinduced DNA damage in HaCaT cell.Amenic could inhibit the mRNA transcription of MGMT gene in HaCaT cells,but the transcription of MGMT gene regulate by H3K36me3 is not closely related to DNA damage induced by arsenic.

Objective To investigate the clinical effect of loading dose of phenobarbital in neonatal hypoxic-ischemic encephalopathy(HIE).Methods Three hundred and fifty-six patients with neonatal HIE were randomly divided into treatment group and control group by systematic sampling,with 178 patients in each group.Two groups were given comprehensive therapy,with or without convalsions,early application of loading dose of phenobarbital in 20 mg/kg intramuscular injection,12 h beck to a maintenance dose 5 mg/(kg·d)in treatment group,while a conventional dose of phenobarbital in 5 mg/(kg·d)was used in control group after convulsion happened.Clinical effect of short-term and long-term were observed.Results The rates of eelampsia,choloplania and total effect was 11.2％(20/178),7.9％(14/178)and 87.1％(155/178)in treatment group,and 68.0％(121/178),50.6％(90/178)and 77.5％(138/178)in control group,there were significant differences between two groups(x2 =6.403,78.459 and 8.308,P ＜ 0.05).DQ score in treatment group was higher than that in control group(P＜ 0.05).The patients in two groups were followed up to age 18 months,the deformity rate in treatment group[7.9％(14/178)]was lower than that in control group[28.1％(50/178)],there was significant difference between two groups(x2 =29.085,P ＜ 0.05).Conclusion Loading dose of phenobarbital is very effective to treat neonatal HIE.

ObjectiveTo study exogenous surfactants disaturated phosphatidyl choline (DSPC) kinetics in full term infants with respiratory distress syndrome(RDS) who were treated with different dose of poractant alfa.MethodsOne hundred and twenty-two full term infants with RDS undergoing mechanical ventilation were divided into group A (80 cases) and group B (42 cases) by simple random method,who received 100 mg/kg or 200 mg/kg poractant alfa mixed with DSPC.Clinical manifestation and respiratory parameters were recorded,and DSPC half-life period and pool size and endogenous DSPC synthesis rate were calculated.ResultsFifty-six infants (70.0％) received a second dose after(25 ± 11) h and 18 infants (22.5％) received a third dose after(41 ± 11 ) h in group A.Twelve infants ( 28.6％ ) received a second dose after (33 ± 8) h and 2 infants (4.8％) received a third dose in group B.DSPC half-life period in group B was longer than that in group A[the fast:(32 ± 19) h vs.(15 ± 15) h;the second:(43 ±32) h vs.(21 ± 13) h],there was significant difference (P＜ 0.01 or ＜ 0.05).DSPC synthesis rate and pool size before the first and second doses had no difference between the two groups.But the oxygenation index at the first and the second dose in group B was less than that in group A,there were significant differences (P ＜ 0.01 or ＜ 0.05).ConclusionPoractant alfa given to full term infants with RDS at a dose of 200 mg/kg results in a longerDSPC half-life period,fewer retreatments,and better oxygenation index values.
