Objective:To investigate the effect of P53 up-regulate modulator of apoptosis(PUMA) on the apoptosis of pancreatic carcinoma BxPC-3 cells and the possible mechanism. Methods: BxPC-3 cells were infected with recombinant adenovirus containing PUMA gene(Ad-PUMA) at 100 MOI for 0-96 h.Apoptosis of BxPC-3 cells was examined by FCM.Expressions of PUMA,Bcl-2,Bax,Cytochrome C and Caspase-3 proteins in BxPC-3 cells were detected by Western blotting.Bax expression in the cytoplasm and mitochondrion and Bax oligomer expression expression in BxPC-3 cells were determined by Western blotting. Results: Apoptosis rates of BxPC-3 cells were significantly increased with the time of Ad-PUMA infection, and peaked after 48 h.Ad-PUMA infection increased the expressions of PUMA,Cytochrome C and Caspase-3 proteins in BxPC-3 cells, and decreased the expression of Bcl-2 protein.Apoptosis rate of BxPC-3 cells after Ad-PUMA infection was correlated with PUMA expression.Ad-PUMA did not affect the expression of total Bax protein in BxPC-3 cells, but Bax expression in cytoplasm was dramatically decreased after infection, and Bax expression in mitochondrion was markedly increased.Furthermore,Ad-PUMA infection induced Bax oligomerization in BxPC-3 cells. Conclusion: PUMA can promote apoptosis of pancreatic carcinoma cells through mitochondrion pathway.

BACKGROUND:The occurrence and severity of denture stomatitis are closely related to denture cleaning methods and wearing time. Therefore, it is of great significance to explore the effects of denture cleansers on denture surface roughness and color stability.OBJECTIVE:To investigate the effects of four different denture cleansers on the surface roughness and color stability of heat-curing denture base resins and elastic denture base materials by means of soaking test.METHODS: Samples of heat-curing denture base resins (n=25) and elastic denture base materials (n=25) were polished and soaked in NaClO, Efferdent, Polident partial, Polident overnight and water, respectively. The soak process was repeated 90 times. Then the color aberration and surface roughness were tested using color meter and roughmeter before soaking and after 30, 60, 90 times of soaking.RESULTS AND CONCLUSION:These four kinds of denture cleansers have greater influence on the elastic denture base materials than on the heat-curing denture base resins. With the increasing of soaking time, the color aberration and surface roughness of the two kinds of denture materials were increased gradualy after treatment with different denture cleansers, and the values were ranked as follows: NaClO > Efferdent > Polident overnight > Polident partial and water. To conclude, denture cleansers can induce the changes in denture surface roughness and color stability of heat-curing denture base resins and elastic denture base materials. NaClO exerts the strongest effect, followed by Efferdent and Polident overnight, and Polident partial exhibits the weakest effect.

Objective:To investigate the effects of Mor-platin, a novel mitochondrial platinum complex, on proliferation and migration of human hepatoma carcinoma HepG2 cells. Methods:Cell counting kit-8 (CCK-8) assay was used to analyze cell proliferation of Mor-platin and classic anticancer drugs, particularly cisplatin, in HepG2 cells. A laser confocal microscope was used to observe whether Mor-platin can target mitochondria. The morphological changes in cellular mitochondria after treatment with Mor-platin were ob-served on a transmission electron microscope. Cell apoptosis was measured by flow cytometry, and cell invasion was evaluated by three-dimensional tumor spheroid model. Results:Mor-platin can inhibit cell proliferation and is dose dependent. The half inhibitory concentration (IC50) of Mor-platin is lower than that of cisplatin. Laser confocal images showed that Mor-platin can target cell mito-chondria and enrich cell mitochondria. Transmission electron microscopy images showed that cell mitochondrial morphology changed after Mor-platin treatment. Furthermore, cell mitochondrial membrane is incomplete and mitochondrial cristae are reduced. Cell apoptosis caused by Mor-platin is dose dependent. The three-dimensional tumor spheroid model showed that the cell areas of the group subjected to Mor-platin treatment are smaller than those of the control group. Conclusion:Mor-platin can target cell mitochon-dria, change the cell mitochondrial morphology, inhibit cell proliferation, and thus promote cell apoptosis. It also showed better anti-cancer effects than cisplatin. Furthermore, Mor-platin can inhibit three-dimensional tumor spheroid invasion. These results suggest that Mor-platin is a potential antitumor drug.

A retrospective study was conducted based on the clinical data of 42 patients of portal hypertension and splenomegaly who underwent laparoscopic splenectomy. The patients were divided into two groups including pedicle priority group and conventional group by different operative method. The operation time and intraoperative blood loss in the pedicle priority group were significantly lower than those in the conventional group (both P < 0.05),and there was no statistically significant difference on the conversion rate of laparotomy, active time postoperation, exhaust time, postoperative hospitalization stay and the incidence of complications (all P > 0.05).Priority processing for splenic pedicle has obvious advantages in laparoscopic splenectomy for portal hypertension and splenomegaly, and it could reduce the difficulty of operation, shorten the operation time and reduce bleeding.

Objective To investigate the effects of a novel |3 titanium fixation with high strength and low elastic modulus on bone healing of osteoporotic fracture and its effects on stress shielding. Methods Forty female Sprague-Dawley rats were oophorectomized before establishment of femur fracture model. The fracture of left femur was stabilized by titanium alloy pin with high elastic modulus (110 Gpa,high elasticity modulus group) and the fracture of right femur by titanium alloy pin with low elastic modu-lus (30 Gpa, low elasticity modulus group).Six and 12 weeks later, the bone volume (BV) , total vol-ume (TV) and bone volume fraction (BVF) were detected by Micro-CT, and the maximum load was de-tected by biomechanical test. The effects of two titanium alloy pins on formation of bony calluses and me-chanical properties of bones were compared. Results Compared with the low elasticity modulus group, more collagen and bony callus, thicker bone trabeculae and more regular lamellar bone were found in the high elasticity modulus group. In low elasticity modulus group, TV (ROI1) was increased obvious-ly at week 6, with no statistical differences in BV (ROI 1), BV (ROI 2) and maximum load between two groups (P < 0.05).TV and BV in region ROI 1 in the low elasticity modulus group were significantly higher than that in high elasticity modulus group (P < 0.05) , while BV in region ROI 2 in low elasticity modulus group was significantly higher than that in high elasticity modulus group at week 12 postoperative-ly (P<0.05).Biomechnical test indicated that the maximum load in low elasticity modulus group was higher than that in high elasticity modulus group. Conclusions The novel B titanium alloy with high strength and low elastic modulus helps reduce effects of stress shielding, diminish bone resorption, in-crease the fibrous callus formation at the initial phase and improve the quality of bone healing at the later phase.

Objective To investigate the therapeutic effect of VSD combined with external fixator in the treatment of open tibial and fibular fractures in Tibet plateau. Methods From August 2014 to August 2015, totally 16 cases of open tibial and fibular fractures were treated in our department, including 12 males and 4 females with an average 39. 4 years-old. There were 4 cases of proximal and mid tibial fractures while 12 cases were distal fractures. Debridement, vacuum sealing drainage combined with external fixator for stabilization of fractures was performed in all patients after general condition was improved. A vacuum sealing drainage change or second stage wound closure or soft tissue coverage was undergone after 7 days. Bone union time by X-ray and complications were recorded. Results All wounds were healed by second stage. Twelve patients months were followed up ( by calling back to the hospital ) for an average 18 months ( 12 to 24 months ) , 4 cases were lost to follow-up. X-ray demonstrated that bone union was obtained at a mean 5. 5th month (3 to 7 months). There were 9 cases (75%) obtained a first phase of fracture healing, while 3 cases ( 25%) of delayed healing. Pin-track infection was occurred in two patients and was cured by conservative treatment. No complications of deep infection, graft skin or flap necrosis, malunion, nonunion or osteomyelitis was seen during follow-up. Conclusions Vacuum sealing drainage combined with external fixator for treatment of open tibial and fibular fracture in Tibet can rapidly and effectively stabilize the fracture, and simultaneously safely and effectively seal the wound, reduce the wound healing time, promote fracture healing and lower the complication rates.

[ ABSTRACT] AIM:To investigate the effects of microRNA-378*( miR-378*) on the survival and apoptosis of human mesenchymal stem cells ( hMSCs ) .METHODS: The expression of miR-378* was determined by microRNA arrays and quantitative real-time PCR ( qRT-PCR) .H2 O2 was used to induce hMSCs apoptosis.By transfection of miR-378*mimic or inhibitor, we up-regulated or down-regulated miR-378* expression in hMSCs.The effect of miR-378*and connective tissue growth factor ( CTGF) on hMSC survival and apoptosis were detected by MTT, LDH, caspase-3/7 and TUNEL assays.RESULTS:The expression of miR-378*was up-regulated in the old hMSCs compared with the young hMSCs.H2 O2 increased the expression of miR-378*, decreased the expression of CTGF.Up-regulation of miR-378*re-sulted in increasing apoptosis and decreasing survival of hMSCs.Conversely, down-regulation of miR-378*resulted in de-creasing cell apoptosis and increasing survival. The regulation of miR-378*on hMSC apoptosis and survival was attenuated by inhibiting the expression of miR-378* and CTGF together.Direct repression of CTGF expression inhibited the hMSC survival and increased apoptosis.CONCLUSION:miR-378*enhances apoptosis of hMSCs by repressing the expression of CTGF.
