Aberrant microRNA(miRNA) expression has been found in most of human cancers.miRNA-related regulation pathways,including the interaction between miRNA and protein-coding gene and the interaction between miRNA and IncRNA,have been shown correlated with the initiation and progression of human cancers.miRNA might be a therapeutic target in the treatment of malignancies.
3.Determination of ginseng saponin Rg_1,Rb_1 and notoginseng saponin R_1 in Qianjin Shenanning Pills by HPLC

AIM:To establish a method of determining effective components in Qianjin Shenanning Pills(Flemingia Philippinensis Merr.et Rolfe,Radix et Rhizoma Ginseng rubra,Radix et Rhizowa Notoginseng,etc). METHODS: The contents of ginseng saponin Rg_1,Rb_1 and notoginseng saponin R_1 were determined by HPLC. RESULTS: The four effective components could be separated and purified by macroporous adsorptive resins of D_(101) and n-butanol,and their contents could be determined. CONCLUSION: The method is simple,feasible and reproducibility is good,it can be used as quality control in medicinal preparation.
4.The Survey of Cryptosporidium Infection Among Young Children in Kindergartens in Anhui Province

Objective To study the cryptosporidiosis infection among young children in 29 kindergartens in the province, and to explore the best way to diagnose the infection. Methods Stool specimens of 1 204 children were collected, oocysts of Cryptosporidium were identified with auarmine O-modified staining, acid-fast staining, safranine T and methylene blue staining, and auarmine O-modeified acid-fast staining. Results The detection rate of Cryptosporidium in four stainings were 2.46%,1.50%,1.98% and 3.46% respectively, and the rate was significantly higher by auarmine O-modified acid-fast staining than other stainings (P
5.Protective effect of propofol combined with Ugustrazin on liver against ischemia-reperfusion injury in rats

Objective To investigate the protective effect of propofol (Pro) in combination with ligustrazin (Lig) on the liver against ischemia-reperfusion (I/R) injury and the mechanisms involved.Methods Fifty male SD rats weighing 240-280 g were randomly divided into 5 groups of 10 animals each: group Ⅰ received sham operation (sham); group Ⅱ I/R; group Ⅲ Pro + I/R; group Ⅳ Lig + I/R and group Ⅴ Pro + Lag + I/R. The animals were anesthetized with intraperitoneal pentobarbital 20 mg?kg-1 . Liver ischemia was produced by clamping the hepatic hilum for 30 min, then the clamp was removed for 60 min reperfusion. In group Ⅲ propofol was infused at 20 mg?kg-1 ?h-1 starting from 20 min before ischemia until hepatic hilum was clamped. In group Ⅳ a bolus of ligustrazin 60 mg?kg-1 was given i.v. 20 min before ischemia. In group Ⅴ propofol was infused and a bolus of ligustrazin was given i.v. as in group Ⅲ and Ⅳ 20 min before hepatic ischemia. Blood samples were taken at the end of 60 min reperfusion for determination of serum activities of ALT, AST and LDH and meanwhile a liver specimen was obtained for determination of MDA content, SOD and XOD activities and for electron microscopic examination. 6.Differential expression of Ezrin in cervical cancer tissues and the relationship between Ezrin and PCNA, MMP-2, MMP-9, HPV

Background and purpose:Previous studies have revealed potential roles for Ezrin in a variety of cancers. Ezrin participates in several functions including cell adhesion, motility and cell survival, and there is increasing evidence that it regulates tumour progression. This study aimed to determine the differential expression of Ezrin between cervical cancer tissues and normal controls and to explore the association of Ezrin expression with clinical pathological characteristics and with the expression of PCNA, MMP-2, MMP-9 and HPV E6 protein. Methods:Retrospective study was performed among 30 cervical cancer tissue samples which were diagnosed in our hospital from Jan. 2002 to Dec. 2006. Ten normal cervixes from uterine myomas were employed as normal controls. Semi-quantitative immunohistochemisty analysis was performed to detect the association of Ezrin expression with clinical pathological characteristics, and with expression of PCNA, MMP-2 and MMP-9, HPV E6 protein in cervical cancer tissue and normal controls. Analyze the association of Ezrin expression with the clinical characteristics. Results:The expression of Ezrin was strong and the distribution was diffuse mainly in the cytoplasm and membrane in cervical cancer tissues, while the normal cervical tissues was weak and mainly in the membrane. However, there was no signiifcant association between the expression of Ezrin and the clinical characteristics including tumor stage, histology and grade. Ezrin expression was positively correlated with the expression of MMP-2, MMP-9 and HPV E6 protein in cervical cancer tissues, except PCNA. Conclusion:Ezrin was over-expressed in cervical cancer tissues and may be involved in the progress of invasion and metastasis in cervical cancer.
7.Potential role of ezrin and its related microRNA in ovarian cancer invasion and metastasis

Objective To screen microRNA (miRNA) that inhibit expression of the metastasisrelated gene ezrin in ovarian cancer cells and explore their correlation to the invasion and metastasis of ovarian cancer. Methods The differential expression of ezrin in two paired high-metastatic and lowmetastatic cell lines were examined by real time reverse transcription (RT)-PCR and western blot. A functional screen with microarray was employed to identify miRNA that were differentially expressed between SKOV3 and SKOV3ip cell lines. Three programs, TARGETSCAN ( http://www. targetscan. org ),MICROCOSM ( http ://www. ebi. ac. uk/enright-srv/microcosm/htdocs/targets/v5/) and PICTAR (http://www. pictar. mdc-berlin. de), were employed to identify all miRNA, which may inhibit the expression of ezrin and were differentially expressed between SKOV3 and SKOV3ip cells. To test the repressive potential of these miRNA, synthetic mimetics were transfected individually into SKOV3ip cells and endogenous ezrin expression levels monitored by western blot and real-time RT-PCR. Results ( 1 ) The mRNA average level of ezrin were (81.74 ± 5.34) -fold higher expression level in SKOV3ip versus SKOV3 cells ( P < 0. 01 ), while (2. 61 ±0. 14)-fold in HO-8910PM versus HO-8910 cells (P <0. 01 ). Elevated protein level of ezrin were observed in SKOV3ip cells compared with that in SKOV3 cells, and the same that in HO-8910PM cells compared with HO-8910 cells. Paired SKOV3 and SKOV3ip cells were employed to study the more significant difference in ezrin expression between them. (2) By a functional screen using miRNA microarray combined with bioinformatics analysis,the miR-183 and miR-22 were indentified as two candidate miRNA,which may have the potential regulatory role in ezrin expression. Real time RT-PCR assays revealed that miR-183 and miR-22 were, respectively, an average of (5.84 ± 0.66)-fold and(6.67 ± 0.67)-fold higher expression level in SKOV3ip versus SKOV3 cells (P <0. 01 ), which were in agreement with the microarray data. A subsequent validation by western blot and real time RT-PCR revealed that over-expression of miR-183 and miR-22 could both lead to an obvious decrease in ezrin protein level,while there were not signicant difference in the level of ezrin mRNA( P >0. 05 ). Conclusion Increased expression of miR-183 and miR-22 may both repress the protein level of ezrin,indicating that miR-183 and miR-22 may bear a potential role in inhibiting ovarian cancer metastasis in a ezrin-mediated way.
8.Effect of differential electroacupuncture frequencies on behavior of acute stress depression model of C57BL/6 mice

Objective To explore the effect of different frequencies interference of electoracupuncture on acute stress of C57BL/6 mice. Methods Thirty male C57BL/6 mice were randomly divided into model group,2Hz electroacupunture group and 100 Hz electroacupunture group.After interfering 7 days by electroacupuncture,all the mice were carried forced swimming tests and tail suspension test,and the immobility time in 5 minutes were taken down. Results The immobility time of forced swimming test and tail suspension test of 2 Hz eletroacupuncture group(32.00±14.73)s,(72.50±20.44)s was shorter than those of 100 Hz electoracupuncture group(145.60±34.62)s,(124.22±21.67)s( P <0.05); and the immobility time of 2 Hz eletroacupuncture group was shorter than that of model group(129.50±33.94)s,(142.23±20.05)s( P <0.05); but the immobility time of 100 Hz electroacupuncture group was no difference with that of model group( P >0.05). Conclusions The effect of 2 Hz eletroacupuncture could improve the behaviors of the acute stress of C57BL/6 mice and had the more effectiveness on anti-depression.
9.Clinical evaluation of delayed open reduction and internal fixation for pilon fracture

Objective To analyze the results of distal tibial pilon fractures treated with delayed open reduction and internal fixation. Methods From September 1997 to May 2001, 26 pilon fractures were stabilized temporarily by application of plaster splint, bandage, calcaneal traction (closed fracture) or unilateral external fixations (open fracture) immediately after injury. There were 19 males and 7 females with an average age of 37.6 years ( 15 to 58 years). Of 26 cases, 6 were open fractures and 20 close fractures, 18 of which were associated with fibular fracture. According to Ruedi-Allgower classification, the fractures were classified as typeⅠin 2, typeⅡin 15, and typeⅢ in 9. The conditions of soft tissue were divided into mild close injury without tension vesicle in 8, and severe injury with tension vesicle or open wound in 18. The definitive internal fixations were performed on an average of 10.2 days (range, 7 to 15 days) after injury as soon as the soft tissue recovered. The final outcomes of treatment were evaluated based on clinical rating and radiographic results. Results All patients were available for follow-up at an average of 3.8 years (range, 2 to 5.5 years) after surgery. All fractures healed at an average of 12.7 weeks (range, 8 to 29 weeks) postoperatively. There were 9 excellent(34.6%), 12 good (46.2%), 3 fair (11.5%), and 2 poor (7.7%) results according the ankle score of Teeny and Wiss. The results were significantly affected by the type of fracture and the severity of soft tissue injury, the excellent and good rate was 88.2% in typeⅠandⅡ, and 66.7% in typeⅢ; the total excellent and good rate was 100% in the patients with mild soft tissue injury, and 72.2% in the patients with severe soft tissue injury. The complications including 2 wound superficial infection and 1 skin necrosis, occurred in 3 patients(11.5%), which were successfully treated by local dressing changes. The incidence of osteoarthritis were found in 16 patients (61.5%) showed by X-ray films, but the radiographic manifestations did not correspond well with the clinical score. Conclusion This two-stage treatment protocol for pilon fracture has some advantages of fewer complications and good function.
10.Safety and effectiveness assessment of neonatal disease screening

Objective To assess the safety and effectiveness of neonatal disease screening so as to provide basis for the establishment of a mechanism for managing neonatal disease screening. Methods Using the ideas of evidence-based medicine, a safety and effectiveness assessment of neonatal disease screening was made by means of literature induction. Results The techniques used in neonatal disease screening were highly safe, but their effectiveness was affected by a variety of links and factors. The differences reported in literature in such aspects as blood-collecting time, testing methods, cut-off values, and indexes for outcome evaluation might mean negative impact on the effectiveness. Conclusion More attention ought to be attached to the understanding and assessment of the safety of screening techniques and more efforts need to be made in conducting systematic studies into the effectiveness of screening techniques.
