2.Optimization of Fermentation Medium with Lactose as an Inducer for High Molecular Weight Recombinant Spider Silk Protein

Based on M9 culture medium,the concentration of ingredients of culture medium was optimized for the fermentation of pNSR32/BL21(DE3),the engineering bacterial with spider silk protein,and lactose as an inducer.The condition of optimum culture medium was obtained for the expression of the high molecular weight recombinant spider silk protein by using orthogonal and individual factor experimental design.The result was showed that the optimum culture medium was consisted of 0.3% glycerol,3% yeast,0.75% tryptone,0.05%(NH_4)_2SO_4 and a little inorganic salt_.It was confirmed that the optimum culture medium will benefit the growth of bacterial and expression of recombinant spider silk protein.The production level of propose protein will attain at 20% of the total proteins in the fermentation.
3.Experimental study of recombinant human vascular endostatin for breast cancer in nude mice model

Objective To explore the effect of recombinant human vascular endostatin(Endostar,YH-16) on MCF-7 human breast cancer cells in nude mice. Methods Forty nude mice were randomized into four groups and received injection with MCF-7 cells. When gross tumor volume reached 1.0 cm3 ,mice were injected subcutaneously once every two days as following for 20 days: (1) O. 9% NS(0. 2 mg/kg) in negative control group;(2) YH-16(20 mg/kg) in YH-16 group; (3) DDP (1 mg/kg) in positive control group; (4) DDP (1 mg/kg) plus YH-16(20 mg/kg) in DDP plus YH-16 group. Mice were killed by cervical spine disloca-tion. And the body weight, tumor volume, rate of tumor inhibited, VEGF, apoptosis rate of tumor cells and pul-monary metastasis rate were detected. Results The means (SD) of tumor volume in DDP plus YH-16 group,positive control group, YH- 16 group and negative control group were 0. 686 (0. 229), 1. 258 (0. 101), 1. 888 (0. 215), 3. 366 (0. 284) cm3, respectively (P <0. 05) ; the rates of tumor inhibited in the four groups were 92. 1% ,57.3 %, 36. 5 % and 0, respectively (P < 0. 05) ; pulmonary metastasis rates were 0, 10%, 20% and 90% ,respectively(P <0. 05) ;the mean (SD) of apoptosis rates of tumor ceils were 31.6% (2. 7%) ,28. 1%(2. 7%), 19. 4% (2. 9%), 15.7% (3.2%), respectively (P < 0. 05). The mean of absorbance of VEGF in the four groups were 0. 530 ± 0. 164,0. 759 ± 0. 210,1. 063 ± 0. 295,1. 268 ±0. 145 (RT-PCR) and O. 260 ±0. 082,0. 348 ±0. 085,0. 461 ±0. 099,0. 556 ± 0. 113(Western blot) ,respectively(P < 0. 05). Conclusion YH-16 can inhibit MCF-7 breast cancer cell growth and distant metastasis in nude mice with little adverse reac-tion. YH-16 has synergistic effect combined with DDP.
4.The curriculum reform of the experiment aiming to improve the practical skill level of the undergraduates majoring in medical imaging

With the rapid development of medical imaging equipment, the higher requirement for the practice skills of the medical imaging professionals has been put forward. The traditional “CT and MR diagnostics” experimental teaching has some disadvantages, such as poor quality of images, imbalance of teaching resources, students' lack of practical skills training, no reflection of the practi-cal skills in the course examination and so on. They are not conducive to the cultivation of talents. Aimed to improve the experimental teaching quality of the course and students' practical skills, the reformative teaching method and means have been carried out, such as improving the laboratory facil-ities , composing experimental guidance , encouraging students to learn actively , strengthening the examination of practical skills, cultivating teachers in many different ways and so on. Through the survey, 92.9% of the students expressed satisfaction with the experimental course teaching reform. These measures will also be used in the teaching of all the sub courses of medical imaging diagnostics to promote the medical imaging professional training.
5.Sequestration of remifentanil by the extracorporeal circuit

Objective To assess the possible sequestration of remifentanil by the extracorporeal circuit.Methods Two types of extracorporeal circuits (ECCs) were used-Xijian type I (group A)and AFFINITY(R) NT(group B),while in control group (group C) a glass container was used.The ECC and glass container were filled with priming solution (6%hydroxyethyl starch 1000 ml + lactated Ringer's solution 500 ml).Remifentanil Wag then added to the priming solution with the final concentration of 100 ng/ml.The priming solution was circulated in the closed ECC or stirred the glass container.The concentration of remifentanil in the priming solution was determined at 2,5,10 and 15 min after addition of remifentanil.Results The remifentanil concentration in the priming solution decreased by 75.1%,57.9% and 18.3%at 2 min after addition of the drug in group A,B and C respectively as compared with the baseline value.The remifentanil absorption by ECC was significantly greater in group A than in group B.There was no significant difference in remifentanil concentration at the 4 time points in each group.Conclusion ECC can absorb remifentanil.The remifentanil absorption by Xijian type I is significantly greater than that by AFFINITY(R).
6.Survey of female lower urinary tract symptoms

Objective To investigate the prevalence of lower urinary tract symptoms in women in a community in Fuzhou. Methods We designed a patient-completed questionnaire according to Bristol Female Lower Urinary Tract Symptoms (BFLUTS) questionnaire.A few parts of the questionnaire were adjusted based on living habits and social backgrounds in our country.The health care workers distributed and collected the questionnaires and instructed the women to fill in them one by one. Results A total of 6066 questionnaires were distributed and 5587 were collected. Of the responders 4745 women were more than 18 years. The prevalence of stress incontinence, urgency incontinence,urgency,frequency and nocturia were 9.0% (425/4745),2.4% (113/4745),10.3% (489/4745),1.5% (72/4745) and 9.8% (446/4745),respectively.The prevalence of storage symptoms increased with age,showing a 20.5%(104/507) in 18~27 years’ group,19.1% (332/1740) in 28~37,22.5%(338/1503) in 38~47,32.2%(202/628) in 48~57,41.2%(61/148) in 58~64,58.0% (127/219) in ≥65 years’ group. The overall prevalence was 24.5%.The prevalence of delayed urinary,intermittent stream,increased abdominal urination, incomplete emptying and voiding pain were 7.0%(330 cases),4.1%(194),4.6% (217),4.9% (233) and 4.8%(229),respectively.The prevalence of general voiding dysfunction increased with age,showing a 13.0% (66/507) in 18~27 years’ group,9.3% (162/1740) in 28~37,10.7%(161/1503) in 38~47,13.2%(83/628) in 48~57,16.2% (24/148) in 58~64 and 26.9%(59/219) in ≥65 years’ group.The overall prevalence was 11.7%. Conclusions The prevalence of lower urinary tract symptoms are high in community-dwelling women,and the prevalence of storage symptoms are more common than that of voiding dysfunction in these women.
8.Development and application of the method for the detection of human IL-6 mRNA

Objective:To develop and apply a method for real-time quantifying IL-6 mRNA.Methods:By Ficoll-Hypaque density gradient centrifugation, human peripheral blood mononuclear cells(PBMC) were isolated from whole blood treated with ethylenediaminetetraacetic acid(EDTA). Total RNA extracted from the PBMC in trizol solution was reverse transcribed to cDNA, which used as templates for fluorecent real-time quantitative PCR amplification of IL-6. PCR system consists of IL-6 primer, SBY Green Ⅰ and so on.Results:The method is wide linear range, sensitive, specific, reproducible and simple.Conclusion:The method can accurately quantify IL-6 mRNA.
9.Clinical features of adult-onset Still's disease patients with pulmonary complications

Objective To review the clinical characteristics of adult-onset Still's disease (AOSD) patients with special pulmonary complications.Methods Medical records of 506 AOSD inpatients,who were admitted in Peking Union Medical Hospital from October 1997 through March 2015,were retrospectively reviewed.The clinical features,biochemical parameters,radiology,pulmonary function tests,ultrasound,treatments and prognosis of AOSD patients with special pulmonary complications were analyzed.Results The incidence of special pulmonary complications in AOSD inpatients was 2.2% (11/ 506),including 3 cases of interstitial lung disease and 8 cases of pulmonary artery hypertension.All 11 patients had common manifestations of AOSD,including fever (11/11),arthralgia/arthritis (11/11),rash (9/11),lymphadenopathy and/or splenomegaly (8/11),serositis (10/11),abnormal liver function (9/ 11),neutrophilia (11/11),and hyperferritinemia (11/11).The patients with interstitial lung disease had no or mild symptom,and were diagnosed by HRCT and pulmonary function test.The patients with pulmonary artery hypertension had various symptoms including dyspnea of exertion (8/8),cough (4/8),chest suppression (2/8),and chest pain (2/8).Four patients died from severe infection,including 3 cases of pulmonary artery hypertension and 1 case of interstitial lung disease.Conclusions Interstitial lung disease and pulmonary artery hypertension are life-threatening complications of AOSD patients.Earlier recognition and prompt treatment should be made to improve patient outcomes.
10.POSTNATAL DEVELOPMENT OF S100B AND GFAP IN THE CENTRAL NERVOUS SYSTEM IN RATS

Objective To study the expressions of S100B and glial fibrillary acidic protein(GFAP) in the central nervous system of Sprague-Dawley rats during postnatal development. Methods Twenty-four male SD rats were divided equally into four groups according to different postnatal times:7-day group,14-day group,21-day group and adult group.Immunohistochemistry was used to investigate the expressions of S100B protein and GFAP in the brain and the spinal cord. Results The amount and density of S100B positive astrocytes decreased significantly in frontal cortex,hippocampus,striatum,substantia nigra and spinal cord during postnatal development.It seemed that the second through the third week after birth was a critical period for these changes.The amount and density of GFAP positive AST increased gradually in the brain,but it was the opposite in the spinal cord.Double-labeled immunofluorescence of S100B and GFAP in hippocampus CA1 area showed that S100B positive stains were evenly distributed in the pyramidal,polymorphic and molecular layers from the seventh till the twenty first day after birth,but apparently decreased in each layer especially in the molecular layer in adult hippocampus while the immunoreactivities of GFAP increased.The proportion of double labeled cells also increased with the aging and more of them were found in the pyramidal and polymorphic layers.Conclusion Different patterns of the expressions of S100B and GFAP exist during postnatal development.It may reflect the different roles of these proteins on the glial cell development.And it may also indicate that the expressions of S100B and GFAP are regulated by different mechanisms during the course of development,which reflects the differentiation of subpopulations of astrocytes during ontogenesis.
