Objective To study the effect of hypoxia on proliferation of cultured bovine retinal pigment epithelium (RPE) cells and expression of the antiapoptotic protein bcl-2. Methods The bovine RPE cells were cultured under normal and hypoxic chamber respectively. After 24 hours, the proliferation of RPE cells was evaluated by[3-(4,5-dimethylthiazole-2yl)-2,5-diphenyl tetrazolium bromide, MTT]-test. Objective To study in clinic the potential antifatigue effect of red-light irradiation onto the soldiers exhausted by a high intensity of exercise with detection of myocardial enzymes, coagulation indices and other blood routines before and after the bio-light therapy. Methods 200 soldiers selected were assigned averagely and randomly into two groups, test group and control group, with 100 for each group. All the volunteers underwent a strong intensity and exhaustion exercise. Then the red-light irradiation of 30J/cm2?30 min once a day performed 2 weeks was given to the soldiers in test group, and the soldiers in control group received similar treatment except that daylight lamp was employed instead of red-light irradiation. The blood samples were then collected from all the volunteers before the exercise and 7d and 14d after the exercise for detecting and comparing the indexes of aspartate transaminase (AST), lactate dehydrogenase (LDH), creatine kinase (CK), creatine kinase MB isoforms (CK-MB), Hydroxybutyric dehydrogenase (HBDH), white blood cell (WBC), hemoglobin (HB), red blood count (RBC), platelet (PLT), prothrombin time (PT), activated partial thromboplastin time (APTT), plasma fibrinogen (FIB), thrombin time (TT) and calcium (Ca2+). Results The findings showed an obvious increase in the indices of HB, RBC and Ca2+ of the soldiers in the test group, the indices were significantly higher than that of the soldiers in the control group. However, no significant difference was found between the two groups in such indices as AST, ADH, CK, CK-MB, HBDH, WBC, PLT, PT, APTT, FIB and TT. Conclusion As one of the harmless biotherapies, red light irradiation is conveniently and easily applied for restoring the exhausted soldiers to their normal physical ability. Without side-effects, the red light irradiation may improve the antifatigue and endurance capacity after an individual underwent a strong intensity and exhaustion exercise. It′s valuable and acceptable for enhancement of soldiers′ combat capabilities.

Objective The study is aimed to investigate the effects of radiation with red light on ATP contents in animals' liver,skeletal muscle and blood,and further to surmise the effects of the radiation on surmounting the exercise-induced fatigue.Methods 18 French white rabbits,weighed 2.0?0.5 kg,were randomly assigned to 3 groups with 6 animals for each:the animals in group A acted as control with no treatment by red light radiation;in groups B and C the animals received radiation with red light once a day and 3 hours for each time,the treatment for group B continued for one week and for group C continued for 2 weeks.Once the treatment of red light radiation ended,the venous blood were collected from the ear edge and the animals were executed,the liver tissue and skeletal muscle samples were collected,and then the contents of nicotinamide adenine dinucleotide(NAD),adenosine monophosphate(AMP),adenosine diphosphate(ADP)and adenosine triphosphate(ATP)in the samples were detected.The same collection and detection of samples were also done for the control animals.Results Compared to control group,the levels of NAD,AMP,ADP and ATP in blood samples,liver tissues and skeletal muscles in both groups B and C were significantly higher after red light radiation for certain period.Conclusion Red light radiation is one of the widely-used biotherapies with such advantages as harmless,acceptable,effectiveness on relieving pains,tension and nervousness,for its capability of inducing beneficial bio-reactions in the subcutaneous,muscular and skeletal muscles with possible improvement of exercise-induced fatigue and endurance.Since ATP serves as a key factor in supplying energy for the body and supporting basic cellular respiration,and red light radiation may increase the ATP contents in blood,tissue and muscular,so the radiation may improve the ability of anti-fatigue.

ObjectiveTo investigate the diagnostic value of N-terminal pro-brain natriuretic peptide (NT-proBNP) and MB isoenzyme of creatine kinase (CK-MB) for heart failure (HF) in pneumonia children.MethodsThe NT-proBNP and CK-MB were assayed in 132 pneumonia children with HF, 138 pneumonia children without HF and 62 healthy children were recruited into this study. A receiver operating characteristics (ROC) curve and a logistic regression model were employed to assess the diagnostic accuracy of NT-proBNP and CK-MB for HF in pneumonia children.ResultsPneumonia children with HF had higher blood NT-proBNP and CK-MB than those in pneumonia children without HF and healthy controls (P<0.01 for both). Pneumonia children with HF had higher blood NT-proBNP and CK-MB than the pneumonia children without HF. The area under curves (AUCs) of NT-proBNP and CK-MB for HF were 0.85 and 0.72, respectively. The AUC for their combinational usage was 0.87.ConclusionBoth NT-proBNP and CK-MB are effective markers as diagnostic adjuncts for HF in pneumonia children. Combination of NT-proBNP and CK-MB can improve the diagnostic accuracy for HF in pneumonia children.

Objective To determine the efficacy of glucose-loweringtreatment by proton magnetic resonance spectroscopy (1H-MRS).Methods Bilateral frontal lobes of 45 male patients with type 2 diabetes mellitus (T2DM) underwent 1 H-MRS separately bythe time it was first diagnosed.Among them,30 were treated with insulin,15 were rejected.The bilateral frontal lobes of all patients were re-examined with 1 H-MRS after 6 months.The ratios of metabolites in the frontal lobe were recorded and compared,and observing whether the lactate(Lac) peak appeared or not.Results Significantly higher values of the N-acetylaspartate (NAA)/creatine (Cr)ratio and NAA/choline (Cho) ratio after 6 months (2.414 2± 0.206 6 and 2.244 2±0.341 5) were found in bilateral frontal lobes compared with pre-therapy (1.905 3±0.135 7 and 1.906 0±0.322 1)(P＜0.05).Significantly lower values of the NAA/Cr ratio and NAA/Cho ratio in the refuse treatment group(1.504 2 ± 0.262 4 and 1.510 0± 0.220 8) were found in bilateral frontal lobes compared with pre therapy (1.960 0±0.175 0 and 1.880 8±0.297 0)(P＜0.05).Lac peak was not detected in the all experiments.Conclusion 1 H-MRS provides valuable information in the assessment and evaluation of clinical efficacy and prognosis of T2DM by detecting the changes of brain metabolism.

Objective To explore the value of multivoxel 1H-MRS in the differential diagnosis of corpus callosum lesions.Methods 17 patients with corpus callosum lesions confirmed by clinical or pathological method underwent MRS examination.These patients included 2 cases of lymphoma,4 cases of Marchiafava-Bignami disease,3 cases of glioma, 7 cases of infarction,and 1 case of multiple sclerosis.MRS features of lesions were analyzed.Results 2 cases with corpus callosum lymphoma revealed significantly increased Cho peak, decreased Cr and NAA peak,towering Lip peak.Among 4 cases with Marchiafava-Bignami diseases, 2 cases revealed increased Cho/Cr and decreased NAA/Cr,1 case showed normal,1 case revealed inverted Lac peak.For 3 cases with corpus callosum glioma,NAA peak decreased or disappeared,and Cho peak increased in varying degrees.7 cases with corpus callosum infarction revealed significant inverted Lac peak, and NAA peak decreased in varying degrees.1 case of multiple sclerosis revealed increased Cho peak,decreased NAA peak,and inverted Lac peak during acute period with enhancement,while these features returned normal during inactive period.Conclusion Multivoxel 1H-MRS plays an important role in the differential diagnosis of corpus callosum lesions.

Objective To detect the effects of siRNA targeting CDX2 gene expression on of BCR-ABL, caspase and Bax expressions, and the mechanisms thereof. Methods According to the earlier experiments, siRNA specifically targeting CDX2 gene (CDX2-siRNA) and the negative control sequence (CDX2-siRNA-NC) were selected, and then were transfected into K562 cells by Roche X-tremeGENE HP DNA Transfection Reagent. The flow cytometry analysis was used to detect the effects of siRNA on cell apoptosis. The expressions of BCR-ABL, caspase-9, Bax mRNA and protein were tested by RT-PCR and Western blot assay. Results MTT and flow cytometry analysis showed that after the silence of CDX2 gene expression, the proliferation of K562 cells was prohibited and the apoptotic rate of K562 cells was distinctly increased compared with that of normal cell group, but the negative control group had no significant change. According to the RT-PCR and Western blot assay, in comparison with the normal cell group and the negative control group, the expression levels of BCR-ABL mRNA and protein were obviously decreased, and the difference was statistic significance. On the other hand, the expressions of caspase-9 and Bax mRNA and protein were significantly higher than those of other two groups (P<0.05). Conclusion CDX2-siRNA can promote apoptosis of K562 cells obviously, and the mechanism is related with the down-regulation of BCR-ABL and the up-regulation of caspase-9 and Bax.

Objective To explore the characters of non-professional drivers' stress. Methods Two hundred and sixty-four non-professional drivers in Chengdu city were randomly investigated by Driver Stress Inventory(DSI). One hundred and five professional drivers were as control group. Results ①Compared with professional drivers,non-professional drivers scored higher in thrill seeking, cognitive fatigue and aggression (non-professional drivers :4.63±1.49,5.12±1.56,5.65±1.42; professional drivers: 3.60 ± 2.04,3.97 ± 2.02,4.56 ±1.84, P ＜ 0.01 ) ,and lower in hazard monitoring, driving anxiety and physiological fatigue (non-professional drivers:7.04±1.48,6.44±1.50,5.78±1.75; professional drivers:7.89 ± 1.45,6.99 ± 1.28,6.45 ± 2.37, P ＜0.01 ). ②Non-professional drivers' stress had significant differences in gender, age, marital state and driving experiences (P＜0.05). The male drivers scored higher in thrill seeking, aggression and physiological fatigue (male:4.79±1.53,6.78±1.46,5.94 ± 1.80; female:4.32±1.37,5.80±1.37,5.50 ± 1.61, P ＜ 0.05 ). Compared with other age driver groups, the 20 ～29 driver group scored higher in cognitive fatigue (5.44 ± 1.38 ), and lower in hazard monitoring(6.77±1.44) ,driving anxiety (6.03 ±1.46). Compared with unmarried drivers, married non-professional drivers scored higher in hazard monitoring and driving anxiety ,and lower in cognitive fatigue( married drivers:7.23±1.43,6.65±1.48,4.88±1.6; unmarried drivers :6.77±1.47,6.06±1.43,5.59±1.36, P ＜0.05 ). The non-professional drivers who drove more than 20 thousand kilometers scored higher in driving anxiety (6.89±1.41). Conclusion The non-professional drivers' stress isn' t optimistic, and influenced by their gender,age, marital state and driving experiences. Stress management should pay attention to them.
