1.Investigation of Water Sources Contaminated by Volatile Organic Compounds in Jiangsu Province

Objective To investigate the water source contamination in Jiangsu Province. Methods Using blowing and arresting device and GC-MS, 90 water samples collected from 15 city drinking water sources in Jiangsu Province were analyzed. Results The main detected compounds were 1, 2-dichloroethane, chloroform, benzene, trichloroethene, toluene, tetrachloroethene, chlorobenzene, m-xylene, p-xylene, o-xylene, 1, 4-dichlorobenzene, 1, 2-dichlorobenzene, 1, 2, 4-trichlorobenzene and hexachlorobutadiene, while 1, 1-dichloroethene, trans-1, 2-dichloroethene, cis-1, 2-dichloroethene, 1, 1, 1-trichloroethane, carbon tetrachloride, bromodichloromethane, dibromochloromethane, bromoform, isopropylbenzene, 1, 2, 3-trichlorobenzene and styrene were not found. The highest concentration of 1, 2-dichloroethane was 27.79 ?g/L, which was close to the limit of surface water. It should be noticed that the detected concentrations of chlorobenzene in water source of district 3 and water source of district 4 were a little higher. Compared with the others, in water source of district 1, water source of district 9 and water source district of 14 a higher concentration of toluene and xylene were detected and the concentration of benzene in water source of district 1, water source of district 2, water source of district 3 and water source district 4 was higher. Only in water source of district 13, none of 25 volatile organic compounds was detected. Conclusion Some drinking water sources have been contaminated by volatile organic compounds in Jiangsu Province.
2.Sodium Phenylacetate Enhances HLA Molecule Expression on the Surface of Tumor Cells

Sodium phenylacetate can induce differentiation of tumor cells and has been approved as a drug for the treatment of adults with cancer. In order to explore the immunological mechanism of its antitumor effect, the influence of sodium phenylacetate on HLA class I and II molecule expression in various human tumor cell lines, including breast adenocarcinoma (MCF-7.MUA-453), gastrocarcinoma(MKN-28,MKN-45), ovarian cancer(3AO) and cervical cancer(Hela), was studied with ELISA. The result showed that HLA class II molecule was absent from the surface of MCF-7 cells, but they could be induced after 7 days of continued treatment with sodium phenylacetate. Sodium phenylacetate was found to increase HLA class I molecule expression on the surface of MCF-7 cells, HLA class 1 and II molecule expression on the surface of MDA-453、 MKN-28、 MKN-45、 Hela and 3AO cells. The effect of sodium phenylacetate on HLA class I molecule expression in tumor cells is dose-and time-dependent.
3.Endothelial nitric oxide synthase gene polymorphism and acute myocardial infarction.

Objective To investigate the association of the Glu298-Asp(894G→T)mutation at exon 7 of the endothelial nitric oxide synthase gene and acute myocardial infarction.Methods By using the designed primers based on flanking sequences of the Glu298-Asp mutation at exon7 of eNOS, 894G→T fragments were amplified by nested PCR from genomic DNA of healthy control and AMI subjects, digested by restriction enzyme after amplification and detected by agarose gel electrophoresis for 894G→T genotyping. To count the genotype and allele frequencies, the significance of difference in the genotype and allele between two groups were assessed by statistical analysis.Results Three genotypes containing GG, GT, TT were identified on the 894 site of the eNOS gene exon 7 in both AMI and controls. In AMI group, there were 25 patients having 9 homozygotes, the other 16 having heterozygotes. In controls, there were 13 cases with G894T mutation,all having heterozygotes. 4.Recent Advances in Research of Antifreeze Proteins

Many overwintering organisms produce antifreeze proteins (AFPs) that can be adsorbed onto the surface of ice crystals and modify their growth. These proteins show great diversity in structures, and they have been found in a variety of organisms. AFPs from insects have higher thermal hysteresis activity than other organisms. Recent studies revealed the structures of AFPs and put forward different ice-binding models. No mechanism, however, can apply to all antifreeze proteins and the molecular interaction between AFPs and ice are not accurately resolved. AFPs can be applied extensively to agriculture, aquaculture and low temperature storage of organs, tissues, as well as cells. To confer transgenic plant cold resistance application of AFPs is essential, while the expression and regulation of antifreeze gene need to be elucidated.
5.Changes of protein kinase C ? in renal tissue of diabetic rats

Objective To study the expression and transcription of PKC? in renal tissue of streptozotocin-induced diabetic rats. Methods Hyperglycemia was induced with streptozotocin(55 mg/kg) in Sprague-Dawley rats. After 5 weeks, the expression of PKC? protein and mRNA was measured by immunohistochemistry, reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis. Results In diabetic animals, the expression of PKC? was greatly enhanced especially in the proximal tubules and the glomeruler mesangial areas with upregulating of the membrane-associated PKC?. RT-PCR analysis showed that mRNA level of PKC? increased by 1.67 folds in diabetic rats as compared with the normal ones. Conclusion The expression of PKC? protein and mRNA is upregulated in early diabetic nephropathy, which suggests an interaction between PKC? and the pathogenesis of diabetic nephropathy.
6.Therapeutic application of S-1 in advanced squamous cell carcinoma of head and neck

In the therapeutic application of head and neck squamous cell carcinoma ( HNSCC),S-1 is used to combine with other chemotherapeutics or even with radiotherapy to constitute chemoradiotherapy regimens.Adverse effects are well tolerated.Regimens containing S-1 are wildly used to control distant metastasis as well as local recurrence.The studies of treatment also show good application value.

8.Clinical study on the alteration of Th17 cells in patients with chronic myeloid leukemia

Objective To investigate the role of Th17 cells in chronic myeloid leukemia (CML) pathogenesis. Methods 33 CML patients [15 newly diagnosed (ND)- and 18 chronic-phase (CP)- CML patients] and 15 healthy controls were enrolled. The percentage of Th17 cells in the peripheral blood (PB) of CML and controls were evaluated by flow cytometry. RORC mRNA expressions were examined by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR). The levels of IL-17 in PB of CML and controls were measured by enzyme-linked immunoadsorbent assay (ELISA). The relationships between Th17 cells and clinical characteristics in CML were analyzed. Results The percentage of Th17 cells in PB of ND-CML patients [(0.71±0.41) %] was significantly lower than that of CP-CML patients [(4.08±0.74) %, P<0.05] and healthy controls [(3.18±1.32) %, P<0.05]. The mRNA level of RORC in PB of ND-CML patients (0.043 3±0.040 5) was significantly decreased compared with that in CP-CML patients (0.086 1±0.052 3, P<0.05) and healthy controls (0.091 0±0.058 4, P<0.05). The levels of IL-17 in PB of ND-CML patients [(1.43±0.22) pg/ml] and CP-CML patients [(1.36±0.19) pg/ml] were slightly higher than that of healthy controls [(1.23±0.14) pg/ml, P< 0.05]. The percentage of Th17 cells had significantly negative correlations with white blood cell counts in PB or bcr-abl (IS). Conclusion Th17 cells may play an important role in CML pathogenesis, which has potential implication for immunotherapy of this malignancy.
9.Efficacy and safety of bronchoalveolar lavage through flexible fiberoptic bronchoscope in treatment of neonatal atelectasis

Objective To evaluate the clinical efficacy and safety of flexible fiberoptic bronchoscopy (FFB) and bronchoalveolar lavage (BAL) in treatment of neonatal atelectasis.Methods Eligible patients, who were diagnosed as neonatal pulmonary atelectasis and admitted consecutively to the neonatal intensive care unit of the Children's Hospital of Hebei Province from January 2013 to January 2015, were included in the study.They were randomly assigned to FFB group (n=30) and control group (n=28).Newborns in the FFB group received BAL under FFB, while those controls received tracheal irrigation after intubation.The duration of lung recruitment, oxygen exposure and antibiotic administration, hospital stay, culture results of respiratory secretions, prognosis and total expenses during hospitalization were compared between the two groups.Complications of FFB were also recorded.Chi-square test and t-test were performed for statistical analysis.Results (1) In the FFB group, atelectasis occurred in the upper fight lobe (n=26), upper lobes of both sides (n=1), lower right lobe (n=2) and lower left lobe (n=1), while in the control group, atelectasis occurred in the upper right lobe (n=26), lower left lobe (n=1) and middle right lobe (n=2) (x2=0.094, P > 0.05).(2) The positive rate of bacteria culture results showed no difference between bronchoalveolar lavage fluid in FFB group and tracheal secretions in the controls [43%(13/30) vs 32%(9/28), x2=0.770, P > 0.05].(3) The duration of lung recruitment, antibiotic administration and hospital stay of the FFB group were all shorter than those of the control group [(4.7±3.4) vs (7.4±6.6) d, (14.0±4.5) vs (20.3±10.9) d, (15.1±4.7) vs (21.8±12.3) d, t=-5.718, 8.604 and 7.733, all P < 0.05].(4) Among babies in the FFB group, nine experienced fever and returned to normal after physical cooling;three showed more shadow in chest X-ray with aggravated dyspnea during a short period, and relieved 12 hours later;two had minimal hemorrhage from tracheal mucous membrane;one showed crying hoarse.Serious complications, such as pneumothorax, massive bleeding or cardiac arrest, did not occurred.No death or refuse of treatment was reported.Conclusion FFB and BAL is much more effective than tracheal irrigation after intubation in treatment of neonatal atelectasis without any severe complications.
10.Action of blocking renin-angiotensin system on protein kinase C?Ⅱ isoform in renal tissue of diabetic rats

Objective To investigate the expression and translocation of protein kinase C (PKC) ?Ⅱ isoform in renal tissue of diabetic rats and the effects of blocking renin-angiotensin system (RAS) on them. Methods Diabetic rats induced with streptozotocin were randomized to 4 groups. (1) Diabetic control without treatment. (2) Treatment with irbesartan (40 mg?kg -1 ?d -1 ). (3)Treatment with fosinopril (40 mg?kg -1 ?d -1 ) and (4) Treatment with a combination of irbesartan and fosinopril (20 mg?kg -1 ?d -1 each). Six normoglycemic rats served as normal control.
