AIM:To investigate the expression and clinical significance of PAK4 in the cell lines and tissues of non-small cell lung cancer (NSCLC).METHODS:PAK4 expression in human bronchial epithelial (HBE) cells, NSCLC cell lines, NSCLC tissues and adjacent non-tumor tissues were assessed by immunohistochemistry, real-time PCR and Western blot.Prognostic value of PAK4 expression was evaluated by Kaplan-Meier analysis and Cox regression.RE-SULTS:PAK4 was over-expressed in the NSCLC cell lines at both mRNA and protein levels compared with HBE cells ( P<0.05).PAK4 was over-expressed in the NSCLC tissues at both mRNA and protein levels compared with adjacent non-tumor tissues (P<0.05).PAK4 was over-expressed in the metastatic NSCLC tissues compared with the primary NSCLC tissues (P<0.05).Higher PAK4 staining scores were positively correlated with differentiation, lymph node metastasis, distant metastasis, and clinical stage.Kaplan-Meier analysis and log-rank test showed that overall survival was significantly different between the patients with up-regulated PAK4 and the patients with down-regulated PAK4 ( P<0.05) .PAK4 over-expression was associated with NSCLC progression.CONCLUSION:Increased PAK4 expression was associated with tumor invasion, metastasis and prognosis in the patients with NSCLC.PAK4 is an important prognostic marker and potential ther-apeutic target in NSCLC.

Objective　 To study the postprandial lipid metabolism in patients with coronary heart disease.　Methods　11 CHD cases as CHD group and 15 normal as control group were selected to undergo oral fat load test. Blood samples were obtained before test and 2，4,6 and 8 hours after test. Serum TG, TC, LDL-C,HDL-C, LPO and LDL-LPO-C were calculated.　Results　1.Preprandial TG, LDL-C, LPO and LDL-LPO-C in CHD group were significantly higher than that of control group. 2. Postprandial TG, LPO, LDL-LPO-C in CHD group were significantly higher than that of control group. Postprandial HDL-C in CHD group were significantly lower than control group. 3. Postprandial TG (8h), LDL-LPO-C in CHD group were significantly higher than in the preprandial. Postprandial LDL-LPO-C in control group was significantly higher than preprandial. Conclusions 1. Postprandial high-lipemia and dyslipemia was obvious in CHD group. 2. Postprandial high-lipemia was closely related to the development of CHD. 3. Postprandial LDL-LPO-C in CHD group increased significantly and LDL-C sensitivity to oxidative intensified with postprandial time. 4. Postprandial LPO in CHD group increased significantly.

Objective To study the effects of metallothionein(MT) egg-milk power on excluding lead in vivo in saturnic rats.Methods A total of 60 Wistar rats were randomly divided into four groups: control(n=20),black control(n=10) and lead-treated groups(n=30).After the rats in lead-treated group were treated with 0.2% lead acetate daily for three weeks,they were randomly divided into two groups: lead-treated group(n=10) and MT egg-milk power treated group(n=20).The rats in the last group were treated with MT egg-milk power for three weeks.At the end of the experiment,samples of whole blood,liver,kidney,brain,femur,faeces and urine were collected for lead detection by atomic absorption spectroscopy.Results As compared with controls,the lead content of samples were significantly elevated in lead-treated rats(P

Abstract: Superior mesenteric veinous thrombosis (SMVT) is a rare complication that often occurs in conjunction with intra-abdominal diseases such as diverticulitis, appendicitis, inflammatory bowel disease, etc. Its clinical symptoms are non-specific and include fevers, abdominal pain; it has no specific symptoms, and the diagnosis depends on clinical laboratory tests and imaging studies. The occurrence of superior mesenteric veinous thrombophlebitis is related to septic phlebitis caused by the sloughing of the embolus containing bacteria into the portal vein with blood flow. Due to the nonspecific clinical features of this disease, diagnosing it based on clinical characteristics and microbiological aspects is a challenge. A case of superior mesenteric veinous septic thrombophlebitis caused by Bacteroides fragilis infection is reported and to provide a reference for the diagnosis and treatment of such diseases. The patient was a 34-year-old man who was admitted the hospital with intermittent abdominal pain and fever. Computed tomography (CT) showed partial thrombosis of the superior mesenteric vein, colonoscopy revealed diverticulitis in the ileoceca, and the blood culture grew Bacteroides fragilis. The patient was treated with anti-infection (ceftazidime 2.0 g q12h intravenous infusion for 11 days; metronidazole 0.5 g, q8h intravenous infusion for 3 days) and anticoagulation (rivaroxaban 20 mg qd orally for 8 days. On the 11th day of hospitalization, the patient's condition improved, and he was discharged. In this case, for patients with fever and abdominal pain, superior mesenteric venous thrombophlebitis should be included in the differential diagnosis. Through auxiliary examination, blood sample culture and other technologies, clear diagnosis should be made in time to improve patient outcomes.

The cell cycle was analyzed by flow cytometric analysis. The morphological changes were observed by fluorescence staining and cellular genome electrophoresis. ResultsAfter sealing c-myc gene with ASODN,the growth of cells was repressed and the effect was time-dependent and dose-dependent ( P = 0. 02 ). The ability of proliferation decreased, the expression of c-myc gene was inhibited on transcription and translation levels; 5-FU can induce apoptosis of hepatoma cells HEPG-2 dramatically with the dose of 10 μ mol/L, when treated by both c-myc ASODN and 5-FU, HEPG-2 cells was induced apoptosis in a cooperative style ( P =0. 01 ).ConclusionsThe liposome mediated c-myc (APSODN) and 5-FU can inhibit the proliferation of HEPG-2 cells by inhibiting the expression of c-myc gene and can induce apoptosis of hepatoma cells HEPG-2 in a cooperative style. c-myc ( APSODN ) can increase the sensitivity of 5-FU to hepatoma cells and decrease the effective concentration of 5-FU.

AIM:To investigate the effect of oleuropein on interleukin-1β( IL-1β)-induced SD rat articular chondrocytes .METHODS:The SD rat articular chondrocytes were isolated by 2 step enzyme digestions .The chondrocytes were cultured in vitro.Inverted microscopic observation was performed during the culture .Alcian blue staining and type II collagen immunohistochemical staining were used to identify the chondrocytes .The effects of oleuropein on the viability of chondrocytes were determined by CCK-8 assay.The cells in 3rd passage were pretreated with oleuropein at 10, 50 or 100 μmol/L and subsequently stimulated with IL-1βat 10 μg/L for 24 h.Production of prostaglandin E 2 ( PGE2 ) and ni-tric oxide (NO) were evaluated by the Griess reaction and an enzyme linked immunosorbent assay (ELISA).The mRNA expression of matrix metalloproteinase ( MMP)-1 and MMP-13 was measured by real-time PCR.The protein levels of in-ducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) and nuclear factor-kappa B (NF-κB) were detected by Western blotting .RESULTS:The cell viability of chondrocytes was not significantly impaired by treating with oleuropein at concentration of 10, 50 or 100μmol/L for 24 h compared with control group .Pretreatment with oleuropein significantly in-hibited the production of PGE 2 and NO induced by IL-1β.Oleuropein also significantly decreased the IL-1β-stimulated MMP-1 and MMP-13 mRNA expression in articular chondrocytes .Pretreatment with oleuropein inhibited the IL-1β-media-ted activation of NF-κB by suppressing the degradation of its inhibitory protein IκBαin the cytoplasm .CONCLUSION:Oleuropein inhibits IL-1β-induced inflammatory gene expression by suppressing NF-κB activation at the transcriptional le-vel, suggesting a new mechanism for the anti-inflammatory effects of oleuropein as a novel agent on treating with osteoarthri-tis.

Objective To investigate the validity of selecting subclones of human breast cancer cells with high-and low-metastatic potentiality,based on their different ability of penetrating the matrigel in vitro,and analyze their biological characteristics.Methods Continuous in vitro invasion assays in matrigel were applied to screen two subclones of human breast cancer cell line MDA-MB-435s,characterized by high-and low-metastatic potentiality.Cell growth curve assay,invasion assay,adhesion assay and angiopoiesis-promoting assay were conducted to explore the different biological properties of these two subclone cells.Results Two subclones of MDA-MB-435s,of high-and low-metastatic potentiality respectively,were obtained through continuous in vitro invasion assay,manifesting significant difference in growth rate,invasion ability,adhesion ability and angiopoiesis-promoting ability.Conclusion It is feasible to obtain subclones of human breast cancer cells of high-and low-metastatic potentiality by utilizing their difference in penetrating the matrigel in vitro.

Objective To revise the Chinese version of the National Institutes of Health-Chronic Prostatitis Symptom Index (CHN-NIH-CPSD), and evaluate its feasibility, reliability, validity and responsiveness. Methods The NIH-CPSI was translated into Chinese according to a standard methodology including forward-backward-forward technique. The CHN-NIH-CPSI was pre-tested in consecutive samples of 162 native-speaking Chinese chronic prostatitis(CP)patients. Ninety-five of 162 filled the index again on the same day and after 4-week therapy. Ninety-seven healthy men were included as evaluated. Results The recovery of the questionnaires was 100% and all the patients filled the index completely. The mean time to complete the questionnaire for the patient group was 5.2±2.4 (range 2 - 12) min. The split-half reliability was 0.82. For the overall index and each subscale, the test-retest reliability was 0.98, 0. 98, 0. 98, 0. 97, respectively(P＜0.01);and the Cronbach's α coefficient was 0. 61,0. 71, 0. 59, 0. 75, respectively. The confirmatory factor analysis showed good construct validity with a goodness of fit index of 0. 85 and a x2 of 124.67(P＜0. 01). Of all 162 patients, the scores of the overall index and each subscale were 23. 33±5.91. 8. 80±4.26, 5.30±2.82, 9. 23±1.90, respectively;and those of healthy controls were 1. 95±1.97, 0. 37±1.03, 0. 15±0.58, 1.42± 1.20,respectively. Of the 95 patients, the original scores were 23. 53±5.60, 9.21 ±4.04, 5.10±2.75,9.21 ±2.05, comparing with 19.47±6.36, 7.79±3.95, 3. 58±1.88, 8.11±2.50, the 4 weeks later scores. The group t-test and paired t-test showed good responsiveness. Conclusions The CHN-NIH-CPSI has high feasibility, reliability, validity and responsiveness for testing the patients with CP. It is suitable for Chinese-speaking patients and helpful for cross-cultural comparisons of men with CP in clinical and research settings.
