2.Gene therapy of human ovarian carcinoma in vitro

To construct a recombinant adenovirus containing CDglyES fusion gene, which can directly inhibit human ovarian cancer cell and indirectly inhibit vascular endothelial cell growth. Methods:We constructed prAdCDglyES using a homolo-gous recombination method in bacteria. The prAdCDglyES was transfected to 293 packaging cells using liposome, in which rAdCDgly-ES was packaged and amplified. MTT was used to observe the proliferation inhibition effect of rAdCDglyES on human ovarian cancer cells and the growth inhibition effect of expressing products of rAdCDglyES on ECV-304. Results:The titer of rAdCDglyES was 1 × 1013.3 TCID50/L, whereas the inhibition rate on human ovarian cancer cell SKOV-3 was (83.1±6.3)%. This result is significantly different from the control rAd-LacZ, which had an inhibition rate of (24.1 ± 13.2)% (P<0.01). The concentrated culture supernatant from cells transfected with rAdCDglyES can inhibit ECV-304 cell proliferation at a rate of (78.7 ± 1.6)%. This rate is significantly different com-pared with that of the control with the same concentration of culture supernatant from cells transfected with rAd-CD, with an effect on ECV-304 cell shown by an inhibition rate of (23.9 ± 9.7)%(P<0.01). Conclusion:The results showed that the recombinant adenovirus rAdCDglyES could inhibit human ovarian cancer cells directly and indirectly.
3.Changes and correlation analysis between serum homocysteine and plasma fibrinogen in ischemic cerebrovascular disease

Objective To investigate the change of serum homocysteine (Hcy) and plasma fibrinogen (Fg) levels in patients with ischemic cerebrovascular diseases (ICVD) and the correlation between Hcy and Fg.Methods Blood samples were collected from 74 patients with ICVD and 40 controls. The concentrations of Hcy, folate and Fg were determinated by means of fluorescence polarization immunoassay, electrochemiluminescence, and immunoturbidimetry, respectively. In the meantime, plasma lipid was detected by routine method.Results Both Hcy and Fg levels in the patients with ICVD increased significantly as compared with the controls (all P0.05). Multivariate logistic regression analysis indicated that hypertension, high cholesterol, high Fg and high Hcy concentrations played significiant roles in ICVD.Conclusion Both hyperhomocysteinemia and hyperfibrinogenemia are independent risk factors of ICVD.
4.Application of Simulated-patient Teaching in Obstetrics and Gynecology Practice Teaching and Assessment

To study the application of simulated-patient teaching in practice teaching and assessment of obstetrics and gynecology,the two groups of students were taught by simulated-patient teaching and discussed cases teaching respectively,and then they were assessed by using simulated patients for clinical skills.The result shows that simulated-patient teaching is effective in improving students' clinical skills,can act as a worthy tool of clinical teaching and assessment.

7.Analysis of prognosis and therapy strategy in patients with lung cancer aged 80 years and over

Objective To analyze the prognostic factors and trerapy strategy of lung cancer in the patients aged 80 years and over.Methods Totally 107 patients aged ≥ 80 years with lung cancer were retrospectively reviewed.Patients' clinical characteristics and treatment were analyzed.Results Median survival time of the patients was 6.9 months.92.9% (13/14) of small cell lung cancer patients and 34.4% (31/90) of non small cell lung cancer patients were treated.Life cycle of patients who accepted effective treatments and supportive treatments were 16.5 months and 8.7 months,respectively (P=0.008).In the early stage of tumors,survival time of patients undergoing surgery was 36.7 months,15.5 months in patients without surgery (P=0.023),while in the late stage,survival time of patients receiving combined chemotherapy was 13.4 months,4.6 months in patients receiving single agent chemotherapy(P=0.002).In small cell lung cancer,survival time of patients who received radiotherapy was 12.8 months,6.4 months in patients who did not receive radiotherapy (P=0.049).Performance status (PS),clinical stage,early surgery,late chemotherapy and radiotherapy(x2=38.236,18.831,5.187,9.827,4.186,P<0.05),but not sex and pathology type affected the prognosis.PS score (P=0.003)and clinical stage(P=0.046) were the independent influencing factors.Conclusions Performance status and clinical stage are the independent influencing factors of lung cancer in the patients aged over 80 years.Patients may improve survival if receiving surgery,chemotherapy and/or radiotherapy when they have good PS,otherwise patients may choose best supportive care.

The ratio of plasma aldosterone concentration to plasma renin activity (ARR) is a practical parameter in screening for primary aldosteronism (PA).However,variations of the cutoff value of ARR in different studies have been reported due to plenty of influential factors that may affect the secretions of renin and aldosterone. Lack of standardization of assays for ARR also makes direct comparisons among different studies difficult.The associated influential factors on ARR were introduced in this review.
10.Effect of epigallocatechin gallate against exercise-induced fatigue in mice.

OBJECTIVETo investigate the effects of epigallocatechin gallate (EGCG)against exercise-induced fatigue in mice.
METHODSTotal 120 mice were randomly divided into three groups and tested separately. For each test, there were 30 mice subdivided into high dose (50 mg/kg . d EGCG) and low dose (10 mg/kg . d EGCG) groups as well as saline control group(1 ml/kg . d) with 10 in each. Burden swimming, running wheel endurance, stick climbing and hypoxia tolerance exercise were used to establish fatigue mice training model in three groups. And intraperitoneal injection with different doses of EGCG per day for consecutively 28 days and the mice in the control group were treated with normal saline. After the last each test, the blood lactic acid (BLA), blood urea nitrogen (BUN), blood lactate dehydrogenase (LDH), muscle glycogen (MG) and liver glycogen (LG) of each group of mice were determined.
RESULTSEGCG treatment groups(B and C)revealed a prolonged the mice survival time of burden swimming test, hypoxia tolerance, running wheel time and the ability of stick climbing(P < 0.05 or P <0.01), and increased LDH activity and MG and LG contents, reduced contents of BLA and BUN. High dose group had an obviously increase effect than lower dose group(P <0.05).
CONCLUSIONEGCG has significant effects against exercise-induced fatigue in mice.
