ObjectiveTo investigate the effects of lentivirus-mediated heat shock protein 70 (HSP70) gene on calcium homeostasis in PC12 cells undergone ischemia and hypoxia, and the mechanism involved.Methods PC12 cells at logarithmic phase were collected, and were divided into recombination lentivirus infection group (infected by lentivirus containing HSP70 and fluorescent gene), lentivirus control group (infected by lentivirus containing fluorescin without HSP70 gene) and non-infection group. HSP70 gene and protein expressions in PC12 cells were determined by reverse transcription-polymerase chain reaction (RT-PCR) and Western Blot. After being challenged with ischemia and hypoxia for 4 hours, the viability of cells was detected by methyl thiazolyl tetrazolium (MTT), the levels of lactic acid dehydrogenase (LDH) in cell supernatant were determined by LDH measurement test kit. The concentration of intracelluer calcium ([Ca2+]i) was assayed by flow cytometer. The activities of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and total ATPase were measured by ATPase test kits.Results The expressions of exogenous HSP70 gene and protein were found by RT-PCR and Western Blot in the recombination lentivirus infection group. After being challenged with ischemia and hypoxia, the viability of cells in the recombination lentivirus infection group was increased significantly as compared with the lentivirus control group and non-infection group (A value: 0.575±0.020 vs. 0.395±0.014, 0.363±0.045,t1= 17.996,t2= 10.600, bothP< 0.001), the levels of LDH in culture medium and the concentration of [Ca2+]i were decreased significantly [LDH (U/L): 743.46±23.68 vs. 935.43±34.77, 962.89±26.68,t1= 11.179, t2= 15.044, bothP< 0.001; [Ca2+]i relative fluorescence: (31.60±2.43)% vs. (41.48±3.33)%, (40.40±3.05)%, t1= 5.853,t2= 5.502, bothP< 0.001], and the activities of Na+-K+-ATPase, Ca2+-Mg2+-ATPase and total ATPase were increased significantly [Na+-K+-ATPase (μmol·mg-1·h-1): 8.608±0.307 vs. 6.728±0.173, 6.450±0.091,t1=9.237,P1= 0.001,t2= 11.675,P2< 0.001; Ca2+-Mg2+-ATPase (μmol·mg-1·h-1): 10.523±0.036 vs. 7.910±0.209, 8.064±0.195,t1= 9.718,P1= 0.001,t2= 11.535,P2<0.001; total ATPase (μmol·mg-1·h-1): 17.041±0.324 vs. 14.150±0.182, 13.983±0.085,t1= 16.113,t2= 17.602, bothP<0.001]. There was no statistical difference in above indexes between lentivirus control group and non-infection group.Conclusion HSP70 can maintain the PC12 cells calcium homeostasis, which may be one of the important mechanisms of anti-apoptosis.

Objective To study the effects of aged and exercise training on hydroxylysylpyridinoline (HP) crosslinking concentrations of extracellular matrix in right ventricular (RV),left ventricular septum (LVS) and left ventricular free-wall (LVFW) in Fischer 344 rats.Methods We evaluated HP crosslinking concentrations of RV,LVS and LVFW extracellular matrix in young adult versus old Fischer 344 rats using high-pressure liquid chromatography (HPLC).Rats were assigned to either a sedentary control group or an exercise training group.Results HP crosslinking concentrations in LVS [(0.23±0.01) vs.(0.43±0.03)mol HP/mol Col,P＜0.05] and LVFW [(0.22±0.05) vs.(0.51±0.03) mol HP/mol Col,P＜0.05] in old sedentary control rats were elevated compared with adult sedentary rats.In adult exercise training rats,training had no effect on HP crosslinking concentration in any of the three regions of the heart.As compared with old sedentary rats,HP crosslinking concentrations in LVS [(0.22±0.06) mol HP/mol Col vs.(0.43±0.03)mol HP/mol Col,P＜0.05] and LVFW[(0.21±0.07)mol HP/mol Col vs.(0.51±0.03) mol HP/mol Col,P＜0.05] were decreased in old exercise training rats.Conclusions HP crosslinking concentration are increased with aging in LVS and LVFW of rats,and exercise can prevente this age associated change.

Objective To study the relationship of left-ventricular function change with circulation levels of CD34+ cells,toll like receptor 2 (TLR2),TLR4 and high sensitivity C-reactive protein (hs-CRP)in elderly isolated systolic hypertension.Methods According to New York Heart Association (NYHA)class,left ventricular ejection fraction (LVEF) and systolic blood pressure (SBP),the healthy subjects and the patients were divided into 5 groups:control 30 cases,class Ⅰ 25 cases,class Ⅱ 29 cases,class Ⅲ 24 cases and class Ⅳ 20 cases.The expressions of circulating CD34 + cells,TLR2 and TLR4 in the patients and healthy subjects were measured by flow cytometry.The level of hs CRP was measured by nephelometry immunoassay.Results In class Ⅳ versus class Ⅰ,the levels of LVEF[(26.4±2.1)％vs.(40.5±3.6)％]and CD346 [(0.5±0.2) × 109 vs.(2.5±0.4) ×109 /L]were decreased,while SBP[(211.1±8.4)mm Hg vs.(151.1±5.9)mm Hg],TLR2 [(8.0±1.0) vs.(3.2±1.1)MFI],TLR4 [(6.4±1.3)MFT vs.(2.4±1.1)MFI]and hs-CRP [(14.7±0.3)mg/L vs.(4.9±0.6)mg/L]were increased (all P＜0.05).Conclusions The lower levels of CD34+ cells,higher TLR2,TLR4 and hs CRP levels are correlated with increasing SBP,decreasing LVEF and higher NYHA class.

Objective To assess whether mTOR was activated in peripheral T lymphocytes of acute coronary syndrome (ACS)patients and to investigate the relationship among mTOR,regulatory T cells and cytokins. Methods Patients with acute coronary syndrome (n=32) were selected in ACS group. Meanwhile, patients who complainted of chest pain but were proven to be normal in ECG and in coronary arteriography, were excluded as ACS patients but were diagnosed as Chest Pain Syndrome (CPS) and selected in CPS group (n=28). The expression levels of mTOR, phospho-p70S6KT389 (indicative of mTORC1 activity) and phospho-AKTS473(indicative of mTORC2 activity) were investigated in T cells, which were isolated from peripheral blood of patients in ACS group or CPS group, using Western blot. The proportion of CD4+CD25+Foxp3+Tregs over CD4+cells were evaluated by FACS. And T cell subset related cytokines such as IFN-γ, IL-4, IL-17 and TGF-β1 were exam?ined by ELISA. Then we investigated the relationship among mTOR,regulatory T cells and cytokins by Spearman analysis. Results mTOR and p-p70S6KT389 expression were significantly enhanced in ACS group as compared with those in patients from CPS group (P<0.01). Higher levels of IFN-γ, IL-17 but lower level of TGF-β1 cytokines as well as decreased propor?tion of Tregs were observed in ACS group than those in CPS group (P<0.05). There was no significant difference in the level of IL-4 and p-AKTS473 between the two groups (P>0.05). By correlation analysis, p-p70S6KT389 expression level positively correlated with IFN-γ, IL-17(rs=0.91,092,P<0.01)and negatively correlated with Tregs and TGF-β1(rs=-0.85,-0.80, re?spectively, P<0.01). Conclusion mTORC1 pathway was activated in peripheral T lymphocytes of ACS patients, and Tregs insufficiency and cytokins imbalance both contribute to the activation of mTORC1 pathway and pathological process of ACS.

Objective To clone and analyze the full-length cDNA of mouse integrin ?4, and repair the mutation sensible locei that caused the change of amino acids. Methods The cDNA of ?4 gene was amplified by RT-PCR using the total RNA extracted from mouse small intestine peyer’s patch. The PCR product was inserted into pMD19-T vector and then transformed into E. coli JM109. The positive recombinant clone was analyzed by restriction endonuclease and DNA sequencing. The mutation of ?4 cDNA that caused the change of amino acids was repaired. Results The cDNA of mouse ?4 had a length of 3 099 bp, and encoded a product of 1 032 amino acids. There were 12 bases pairs mutation of ?4 gene and the 6 base pairs causing the change of amino acids was repaired. Conclusion The cDNA of mouse ?4 is cloned successfully.

Objective To understand the relationship between Demodex and bacteria infection in rosacea (brandy nose), and to find effective means for the treatment. MethodsCellophane tape was used to detect Demodex on the nasolabial grooves and the face; sebum and tissue on face was scraped and cultured to examine bacteria under microscope. The hospital_made anti_rosacea lotion was used on the affected part two times a day for 7 days. Results It was found that 193 (74^2%) of 260 cases with rosacea were infected by Demodex and 209 (80^4%) of the patients were infected by bacteria. The overall effective rate of the treatment for rosacea was 73^5%. Conclusion Bacteria infection in rosacea is an important factor inducing rosacea. The curative effect of the anti_rosacea lotion is good.

The association of polymorphisms of calcium channel?1 subunit ( Cav1.1 ) gene ( - 1551T/C at exon 11 ) with thyrotoxic periodic paralysis (TPP) was investigated by PCR-RFLP.The distributions and frequencies of - 1551TC + CC genotype and C allele in TPP group were significantly higher than those in hyperthyroidism (HT) and normal control (CON) groups.There was no statistic difference between HT and CON groups.Cavl.1 gene - 1551TC + CC genotype and allele may contribute to the development of TPP in male HAN population from North China.
