1.Optimization of Extraction Technology of Hyperici Perforate Herba, Coptidis Rhizoma, Schisandrae Chinensis Fructus, and Ziziphi Spinosae Semen forJinzao Yuanzhi Capsule by Orthogonal Test

Objective To optimize the extraction conditions of Hyperici Perforate Herba, Coptidis Rhizoma, Schisandrae Chinensis Fructus, and Ziziphi Spinosae Semen forJinzao Yuanzhi Capsule.Methods Hyperin extraction rate was set as index, and HPLC was used for determination. The extraction conditions of ethanol concentration, ethanol quantity, extraction time and other factors were studied with orthogonal test and comparative test.Results Ethanol concentration and extracting times were remarkable factors in the test. The optimum extraction technology forJinzao Yuanzhi Capsule was to reflux and extract the materials with 8 times of 70% ethanol for 2 times, reflux extraction for 1.5 hours, filtration. Then 6 times of 70% ethanol was added for 1.5 hours’ reflux, filtering, and merging extracted liquid.Conclusion The optimum extraction technology is reasonable and feasible.

Objective To explore the effects of curdione on cognitive function and expression of inflammatory factors in the hippocampus after partial hepatectomy in aged mice.Methods The animals were divided into control group, sham group, operation group, and high-, medium-and low-dose curdione groups.The model of partial hepatectomy in aged mice was established according to the method reported in literature.The levels of SOD, MDA, CAT and GSH-Px in the hippocampus were detected by enzyme-linked immunosorbent assay (ELISA), while the expressions of NF-κB, IL-1β, IL-6 and TNF-α were measured by Western blot.Results The escape latency was significantly increased (P<0.01), and the number of platform-site crossovers was significantly decreased in operation group (P<0.05).Compared with those in operation group, curdione significantly decreased escape latency and increased platform-site crossovers (P<0.05, P<0.05).The levels of SOD, CAT and GSH-Px in the hippocampus were significantly decreased (P<0.05) and the content of MDA was significantly increased after partial hepatectomy (P<0.01), and these were reversed by curdione (P<0.05, P<0.05, respectively).The expressions of NF-κB, IL-1β, IL-6 and TNF-α protein were significantly inhibited by curdione (P<0.05).Conclusion Curdione can significantly improve cognitive dysfunction after partial hepatectomy in aged mice and the mechanism may be related to its inhibition of oxidative stress, inhibition of NF-κB activation and reduction of inflammatory cytokines IL-1β, IL-6 and TNF-α in the hippocampus.
8. Pharmacodynamics of Danshen co-microemulsion in treatment of experimental acute myocardial ischemia induced by pituitrin in rats

Objective To study the effects of Danshen co-microemulsion (Co-ME) in treatment of acute myocardial ischemia induced by pituitrin in rats. Methods The experimental model of myocardial ischemia was produced by injecting pituitrin into the sublingual vein of rats. The serum level of malonaldehyde (MDA) and the activities of phosphocreatine kinase (PCK), lactate dehydrogenase (LDH), and erythrocuprein (SOD) were examined in Co-ME, salvianolic acid microemulsion, tanshinone microemulsion, tanshinones suspension and blank microemulsion groups 30 min after injection of pituitrin. Results Co-ME, tanshinone microemulsion, salvianolic acid microemulsion and tanshinone suspension all improved myocardial eschemia to some extent; besides, they also decreased PCK, LDH activities and MDA content, and increased SOD activity. The therapeutic effect of Co-ME group was superior to those for the tanshinone microemulsion and salvianolic acid microemulsion groups, and significantly better than that of tanshinone suspension group (P<0. 05). Conclusion Microemulsion system can greatly promote the absorbance and bioavailability of tanshinone, displaying a therapeutic effect for acute myocardial ischemia. Salvianolic acids combined with tanshinones can promote the therapeutic effect.

Characterized by dihydropyrimidinuria, dihydropyrimidinase (DHP) deficiency refers to a rare disorder of pyrimidine degradation, with high phenotypic heterogeneity.The disease-causing gene is DPYS, and less than 40 cases were reported worldwide.Urinary gas chromatography/mass spectrometer (GC/MS) can screen clinically suspected patients, and gene sequencing is the main means of the diagnosis of the disease.This article reviews the pathogenesis, clinical manifestation, genotype and recent research progress of the disease.
