4.Recombinant adenovirus vector-delivered report gene expression in rats kidney through different ways
Basic & Clinical Medicine 2006;0(09):-
Objective To investigate expression of recombinant adenovirus(rAV) vector-mediated report gene in kidney, and to seek for the best passway of rAV vector-mediated gene transfer in vivo. Methods Thirty Sprague-Dawley(SD) rats were divided into 5 groups randomly. rAV-GFP(1013viral particales?L-1) was injected through different ways. 2, 7 and 14 d after injection, GFP expression in left kidney, right kidney, liver and lung was evaluated by fluorescence microscope and computer software. Results GFP gene expressed in kidey and reached peak 7 d after injection expression. Conclusion rAV vector-mediated report gene can be transducted in kidney. The rAV-mediated transgene expression in kidney is a potential strategy in the treatment of renal diseases.

Objective To evaluate the predictive value of diffusion weighted imaging ( DWI) for progres-sion free survival(PFS)in patients with cervix cancer after neoadjuvant chemotherapy (NACT).Methods Thirty two consecutive patients with pathologically confirmed cervical cancer underwent MRI including DWI before and after NACT.Pathologic results and MR images were reviewed .Univariate analysis was applied by Kaplan -Meier method. The Cox proportional hazard model was used to evaluate prognostic variables for multivariate analysis .Re-ceiver operating characteristic ( ROC) curves were used to find optimal cutoff values of independent prognostic fac -tors for disease progression. Results Of 32 patients,10 had disease progression during follow -up period. In uni-variate analysis,FIGO stage, tumor size, the depth of tumor invasion, lymph node metastasis and ADC changes be-tween before and after neoadjuvant chemotherapy (ΔADC) were significantly associated with the disease progres-sion.In multivariate analysis , FIGO stage and ΔADC were the independent prognostic factors for PFS .Optimal cutoff values that determined by ROC curves analyses were FIGO stage ⅡandΔADC 0.31.Area under the ROC curve(AUC)of FIGO stage and ΔADC were 0.841(sensitivity 90.0%,specificity 68.2%)and 0.864(sensitivity 80.0%,specificity 81.8%),respectively.Conclusion In patients with cervix cancer after NACT ,FIGO stage andΔADC are significant predictive values for PFS .

Objective To establish a RP-HPLC method for determination of the concentrations of dextromethorphan and its metabolites dextrorphan in human urine. Methods Phenacetine was used as internal standard, and the urine sample was hydrolyzed by enzyme, alkalified and extracted with hexane-butanol(91).The separation was carried out on DiamonsilTM C18 column(250 mm?4.6 mm,5 ?m) with mobile phase of acetonitrile-1% triethylamine buffer solution(pH adjusted to 2.2 with H3PO4).Gradient elution was done for 0-15 min(20%-35% A).The flow rate was 1.0 ml/min.The detection wavelength was set at 280 nm and the column temperature was 40℃.Results The linear ranges of dextromethorphan and dextrorphan were 0.05-2.0 ?g/ml(r=0.999 9,n=5) and 0.5-20.0 ?g/ml(r=0.999 9,n=5),respectively,and their lowest detecting concentrations were 0.04 ?g/ml and 0.4 ?g/ml,respectively.The intra-day and inter-day precision were both less than 10%.The low, middle and high extraction recoveries were between 94%-108%.Conclusion Our method is accurate and sensitive, and is suitable for the CYP2D6 phenotype analysis and pharmacokinetic studies of dextromethorphan and its metabolites in human.

Objective Through the investigation of 80 cancer operation patients' families,we would like to investigate further more the effect of their anxiety conditions for the patients' mental state and find out the corresponding strategy. Methods 80 cancer patients' families in our department were chosen to fill the anxiety scales between the 2nd to 5th day after their admission and some corresponding directions were given. Results Compared with the norm, the cancer patints' families' anxiety scores had significant difference(P
8.Effect of propofol on hypoxic pulmonary vasoconstriction in rats
Chinese Journal of Anesthesiology 1995;0(12):-
Objective To investigate the effect of propofol on hypoxic pulmonary vasoconstriction(HPV)and assess the underlying mechanism in rats. Methods Twenty male SD rats weighing 300-400 g were.anesthetizedwith intraperitoneal phenobarbital.Heart and lungs were removed after thoracotomy.Pulmonary.arterial rings 4 mmin length and 1.0-1.4 mm in diameter were prepared and suspended in Earl solution maintained at 37℃ with a pHof 7.40 and aerated with 20% O_2-5% CO_2-25% N_2.The rings were stimulated with phenylephrine(PE)10~(-6)mol?L~(-1) with different preloads(300,500,700,900,1 100 mg).The isometric tension of the arterial rings wasmeasured. The optimal preload was determined to be 900 mg which allowed best contractility.Hypoxia was inducedby aerating the solution with 95% N_2-5% CO_2 and the flow rate was adjusted to maintain PO_2 of the solution at40-20 mm Hg and pH at 7.40.HPV of the rings were recorded. Then propofol was added to achieve a finalconcentration of 1,3,10,30,100 ?mol?L~(-1) and HPV was again induced and the changes in HPV wererecorded. In addition the effects of propofol(10,30 ?mol?L~(-1))on vasoconstriction produced by KCI and PE werealso measured. Results The lower doses of propofol(10 ?mol?L~(-1))significantly inhibited HPV(P

Objective To show the condition that the NOSIE (Nurses' Observation Scale for Inpatient Evaluation) used in schizophrenia patients. Method To evaluate the male and female schizophrenia patients (paranoid type) by NOSIE. Results The total cases were 100 patients, male and female were both 50 cases. The result of evaluation showed that there was significant difference in 5 factors such as social competence, irritability, retardation, psychosis and depressive between male and female patients. As well, it matched with clinical information. The result of the consistency test for survey indicated that its Pearson coefficient of correlation for all of 7 factors in NOSIE were 0.617~0.843. Conclusion NOSICE can provided some objective data to clinical works of nurses. But, in other hand, its identity is lower.

Objective The purpose of this study was to access the efficacy of Venlafaxine and Reboxitine on treatment of depressive patients who had not responded to selective serotonin reuptake inhibitors for eight weeks. Methods 117 cases depressive patients who had not responded to selective serotonin reuptake inhibitors for eight weeks were divided into three Venlafaxine group(n =41),Reboxitine group(n =39),Original medicine group(n =37) by randomly and were treated by corresponding drugs for 8 weeks. The therapeutic response and safety were evaluated by scale of HAMD and TESS at before and after treatment the end of 1,2,4,8 week. Results The score of HAMD in Venlafaxine and Reboxitine groups were significantly lower than Original medicine group at the end of 4,8 weeks after treatment and the score of HAMD in Venlafaxine group were significantly lower than Reboxitine group at the end of 8 weeks after treatment. The score of HAMD in Venlafaxine and Original medicine groups were marked descendanter than before treatment from after treatment second weekend(P <0.01),while reboxitine group did not marked descent until the 4th weekend after treatment. The reduce score of factors in cognition, retardition and desperation of Venlafaxine and Reboxitine groups were significantly higher than Original medicine group at the end of treatment. The curative effect of Venlafaxine group were supper than Original medicine group. The side reaction of three groups were not difference. Conclusions There are obviously curative effect using Venlafaxine to treat the depressive patients who had not responded to selective serotonin reuptake inhibitors. The time of initial effect in Venlafaxine were rapider than Reboxitine group.
