1.Advances of Src kinase family and paclitaxel resistance

Src family kinase (SFK) highly expresses in many types of cancers,broadly adjusting their malignant behaviors.Paclitaxel is a widely used chemical agent.However,because of constant resistance,the effect of paclitaxel has been greatly attenuated.The present review summaries the recent research progress of the structure and adjustment of SFK and the molecular mechanism of paclitaxel resistance,as well as the regulation of SFK on paclitaxel resistance,in order to provide new references and evidences upon the paclitaxel-based tumor therapy.
2.Expression and clinical significance of Mel-18 mRNA in gastric carcinoma

Background and purpose: Mel-18 is one of the mammalian polycomb group members. A number of related researches have implied that Mel-18 may play a role in human tumorigenesis.In this study, we measured the expression of Mel-18 in gastric carcinoma cells in vivo to explore the expression and clinical significance of Mel-18 in gastric carcinoma. Methods: Real time RT-PCR was used to detect the expression of Mel-18 in cancer tissue and corresponding normal tissue in 52 cases of gastric carcinoma. The association between Mel-18 expression and the clinicopathological parameters of the tumors was analyzed. Results: The analysis revealed that there was significantly decreased expression of Mel-18 in 18 (34.62%)carcinoma tissues in comparison with para-cancer normal tissue. There was no correlation between Mel-18 expression and clinicopathological parameters, such as age, gender, tumor size and histological differentiation (P>0.05).The decrease of Mel-18 expression was significantly negatively correlated with lymph node metastasis and clinical stage (P<0.05). The expression levels of Mel-18 evaluated by the ratios of gene expression were 1.357,0.453,0.183 and 0.170 in stage Ⅰ, Ⅱ,Ⅲ and Ⅳ gastric carcinoma, respectively. They were 0.634 and 0.210 in patients without lymph node metastasis and in patients with lymph node metastasis, respectively. Expression of Mel-18,lymph node metastasis, and clinical stage were significant covariates, respectively (P<0.05). Conclusion: Our results showed that Mel-18 might play a crucial role in tumorigenesis and progression of gastric carcinoma. It is possible that Mel-18 could be used as one of the biomarkers for predicting the prognosis of gastric carcinoma.
3.Impact of drug intervention on cardiomyopathy in STZ-induced type 1 diabetic rats

Objective To investigate the pathological changes of myocardium in diabetic rats and the effect of aspirin, perindopril, prednisone on the pathological changes of myocardium in type 1 diabetic rats. Methods Thirty SD rats were randomly divided into the following groups: ( 1 ) diabetes mellitus (DM)group ( n = 20): treated with intraperitoneal injection of streptozocin (STZ) 60 mg/kg at one time, diluted in sodium citrate buffer. (2) normal control group (n = 10): intraperitoneal injection of sodium citrate buffer. The blood glucose and the weight were measured regularly for 8 weeks. The 20 diabetic model rats were randomly divided into four groups (n = 5 individually), from the 4th day, the rats were given normal saline dissolved aspirin, perindopril, prednisone gavage respectively, and the normal control group and diabetic group were given an equal volume of saline daily gavage. After 8 weeks, the pathological changes in the cardiac muscle tissue were observed under light microscope and electron microscope. Results Three days after the injection of STZ, the blood glucose in the DM group was higher than 16. 7 mmol/L,20 rats were all modeling successfully. The model rats suffered from polydipsia, polyphagia, polyuria and weight loss significantly. HE staining showed diabetic myocardial cell swell, hypertrophy, degeneration and significant nuclear enlargement; the myocardial fiber fractured,dissolved, contraction wavy and disorganized fibre infiltrated by inflammatory cells. Under electron microscope, diabetic myocardial muscle fibers arranged irregularly, fractured and dissolved; the glycogen between the muscle fiber accumulated, mitochondria swelled, most of the ridges in mitochondria were not clear, cell plate widened. Compared with the diabetic group, the above-mentioned pathological changes reduced in the drug intervention group,and the changes in perindopril group was most significant. Conclusion Injection of STZ 60 mg/kg could successfully induce diabetic in rats and held stable blood glucose level. After 8 weeks, diabetes could cause servere damages in myocardium in the rats, but aspirin, perindopril, prednisone could reduce the cardiomyopathy in diabetic rats,in which perindopril had more positive effect.
4.Targeted therapy of metastatic renal cell carcinoma

Surgery is the only curative treatment modality for renal cell carcinoma (RCC). However, approximately 30% of patients who undergo successful nephrectomy for RCC will develop locoregional or metastatic recurrence. Effective treatment for recurrent or metastatic RCC is limited. It is known that conventional radiation therapy and chemotherapy are relatively ineffective for RCC patients with distant metastases. Although immune therapy with high dose interleukin can provide disease control for a portion of patients with advanced RCC, its therapeutic effect usually is not sustainable. In addition, substantial adverse effects and complications have limited the use of high dose interleukin treatment. Advances in the understanding at the molecular level of cancer have led to much progress in the development of anti-cancer agents, including drugs of targeted cancer therapy. Targeted therapy is not only effective in cancer treatment, but also has reduced adverse effects compared with conventional chemotherapy and immune therapy. Much progress in the treatment of advanced RCC by targeted therapy has been achieved in recent years. In this review, we will illustrate the roles, mechanisms and effects of several targeted therapy agents, including the two newly FDA-approved drugs, sorafenib and sunitinib, in the treatment of advanced renal cell carcinoma.
5.Killing effect of laser irradiated hexyl 5-aminolevulinate on human erythroleukemia cell line K562 and drug-resistance cell line K562/ADM

Objective To investigate the photodynamic therapy(PDT)of human erythroleukemia cell line K562 and drug-resistance cell line K562/ADM using hexyl 5-aminolevulinate(He-ALA)in vitro.Methods Four groups were set:PDT group(photosensitizer with light irradiation),laser group(light irradiation only),dark-cytoxocity group(photosensitizer only)and normal control group(neither photosensitizer nor light irradiation).K562 cells and K562/ADM cells cultured were incubated with various concentrations of He-ALA(0.025,0.1,0.4 and 1.6mmol/L)for 4 hours,then illuminated with different light doses(4.5,9,18 and 36 J/cm2)using a laser with a wave length of 630nm.After 12 hours incubation,Wright's staining method was used to observe the cell's morphological changes,the survival rates of cells were analyzed by CCK8 assay,and the changes in colony-forming abilities of cells were analyzed by methylcellulose colony-forming assay.Results He-ALA or light irradiation alone had no evident cytotoxicity,while the application of He-ALA plus light irradiation effectively killed the leukemia cells.With the increase of the concentrations of He-ALA and the dosages of irradiation,the viability of cells decreased.Under the same conditions,the survival rate of K562 was significantly lower than that of K562/ADM(P

[Objective]To analyze factors related with surviving rate and to evaluate effectiveness of the adjuvant chemotherapy in the treatment of osteosareoma.[Method]Eighty-four patients aging from 9 to 47 years(averaged,21 years)were analysed respectively:52 of them were male and 32 were female.The tumors were located at the femur in 42,the tibia in 29,the humerus in 13.There were 22 patients classified as stage ⅡA and 62 patients as ⅡB.The pathological study,of subtype of osteosarcoma revealed that 47 were osteoblastic,11 chondroblastic,19 fibroblastic and 7 other subtypes.There were 46 patients who received the chemotherapy;38 patients without chemotherapy,49 of the 84 patients treated surgically had limb salvage procedures,35 had amputations.Multivariate analsis was done by using the proportional hazards model of Cox,categoric data were analyzed by using the chi-square statistic.[Result]All cases were followed up from 6 to 74 months(with an average of 25.5 months).Cox model analysis showed that age,sex,site,and subtype were not significant prognostic variables in this group of patients;the significant affecting prognosis in patients was Enncking staging and chemotherapy.Chi-square showed significant difference in the higher metastasis rates of lung in the group without chemotherapy than in those with chemotherapy group(P
8.Green computing-based construction of hospital information system

Energy conservation has become a problem needing to be immediately solved with the extensive con-struction of hospital information system. As a high computing technology, green computing uses resources economically and effectively and is characterized by energy conservation and environment friendship. Its concept and characteristics, and the status quo of hospital information system construction were described. A green computing framework model was established for hospital information system construction, which plays an active role in implementing the idea of green environment protection, low carbon and energy conservation, and sustainable development, thus laying a foundation for studying the mechanisms underlying energy conservation in hospital information system construction and for assessing and optimizing its strategic system.
10.Value of vestibular evoked myogenic potential on the diagnosis of inferior vestibular neuritis

OBJECTIVE To invstigate the diagnosis role of VEMP for inferior vestibular neuritis. METHODS 18 patients with inferior vestibular neuritis were studied. Especially, the oVEMP and cVEMP were recorded in all cases. RESULTS All 18 patients with VN (vertigo symptom in 15 cases, and balance disorders in 3 cases) presented normal in PTA, caloric test and Ovemp test. By contrast, the cVEMPs was abnormal in all patients with VN, including flat/no amplitude of cVEMP in 14 cases, and lower amplitude in 4 cases. After three months, 10 patients recovered, with nomal amplitude of cVEMP in 9 cases and lower amplitude in 1 case. And after half-a-year-follow-up, 18 patients went back to normal, with lower amplitude in only 2 cases. CONCLUSION The VEMP examination is valuable in the diagnosis and prognosis of inferior vestibular neuritis.
