1.Efficient isolation of chondrocytes from epiphyseal cartilage and their biological characteristics during culture

Objective To establish the methods of efficiently isolating chondrocytes from epiphyseal cartilage and observe the basic biological characteristics of the cultured ones. Methods The improved isolation method through three-step enzymatic digestion using 1 g/L trypsase in 1 g/L ethylenediaminetetraacetic acid, 1 g/L hyaluronidase and 2 g/L type Ⅰ collagenase was established to isolate the primary passage chondrocytes from epiphyseal cartilage of young rabbits and its efficiency evaluated. The cellular biological characteristics of the chondrocytes were observed under the inverted and light microscopes. Results (1) All chondrocytes were isolated from the matrix after it was completely digested by three above-mentioned enzymes. The harvested chondrocytes significantly and positively correlated with the wet weight of cartilage. The number of chondrocytes from 1 g of wet cartilage was 32.3?106 with a viability of 97.6% on average. (2) The chondrocytes of the primary and the first passages showed triangle or multi-angle shapes (oval during growth and fusion) with the positive immunohistochemical stain of collagen type Ⅱ and the strong heterochromia to toluidine blue. The chondrocytes after the third passage became spindle-shaped with the negative stain of collagen type Ⅱ and the weak heterochromia to toluidine blue. Conclusions (1) The chondrocyte-isolating method through three-step enzymatic digestion has advantages of high efficiency in harvesting primary chondrocytes, high cellular viability and simple manipulation. (2) The chondrocytes differentiate and lose their special phenotypes gradually with passage.
2.Cost-effectiveness Analysis of Three Angiotension-converting Enzyme Inhibitors in Treatment of Essential Hypertension

OBJECTIVE:To evaluate the economic effectiveness of three angiotension-converting enzyme inhibitor(ACEI) in treating patients with essential hypertension.METHODS:186 patients with essential hypertension were randomly divided into three groups receiving enalapril,fosinopril and perindopril respectively.The results were evaluated with pharmacoeconomic analysis.RESULTS:In treatment of hypertension,enalapril was minimal in cost.CONCLUSION:Therapeutic effect will be improved and cost decreased when pharmacoeconomic cost-effectiveness analysis is used.
3.Microendoscopic discectomy for enormous central type lumbar disc herniation:report of 23 cases

ObjectiveTo study the feasibility and effect of microendoscopic discectomy on the central type of enormous intervertebral disc herniation.MethodsFrom 2003 to 2006,23 patients(18 males,5 females) were diagnosed with enormous central type lumbar disc herniation and treated by microendoscopic discectomy.Their clinical data were restropectively analyzed,and postoperative outcomes in hip function were evaluated by Macnab scoring system.ResultsBeing followed up for 3-12 months,21(86.9%) patients were rated as good or excellent for their outcome,and the other two as fair.ConclusionMicroendoscopic discectomy through interlaminar approach achieves satisfied outcome for enormous central type lumbar disc herniation.
4.Research of automatic human face recognition correlative with face pose

Engenfaces is a simple and effective face recognition method based on the relativity of the training set and testing set of face images.However,the correct recognition rate decreases by eigenfaces sharply when the face pose varies.Aiming at improving correct recognition rate in this case,a face recognition approach correlative with face poses is presented.The basic idea is toensure that testing face with particular pose should be matched in the face database whose poses are the same as that of the testing face.Experimental results show the new method gains higher correct recognition rates than traditional eigenfaces method.
5.Management of lumbar intervertebral disc herniation with lateral recess stenosis by microendoscopic discectomy: a report of 79 cases

Objective To review the preliminary experience on managing the patients suffering from lumbar disc herniation with lateral recess stenosis by microedoscopic discectomy.Methods A total of 79 patients affected with lumbar disc herniation with lateral recess stenosis were treated by MEDⅡ in our hospital from Jan 2002 to Sep 2004.Results All patients were discharged within 7 d postoperatively and no complications occurred.The average duration of follow-up was 12.3 months and the successful clinical outcome was 99%.Conclusion Microendoscopic discectomy is a minimum invasive procedure.The successful clinical outcome could be achieved in properly selected patients.The microendoscopic discectomy is an effective procedure in managing lumbar intervertebral disc herniation with lateral recess stenosis.
6.Protective effect of Liensinine on oxidative damage of erythrocyte ghost by oxygen free radicals

Aim To study the influence of Liensinine(LIE) on the oxidative damage of erythrocyte ghost. Method The erythrocyte ghost oxidative injury caused by super oxide anion (O?_2) from the auto-oxidation of pyrogallic acid was used as model. The effect of LIE on oxidated erythrocyte ghost was observed by the measurement of the malonidial aldehyde (MDA)-the degradation product of lipid peroxidation, blocking degree, speed of auto-oxidation and membrane fluidity. Result O?_2 caused lipid peroxidation of erythrocyte ghost, increased the content of MDA and the speed of auto-oxidation, decreased membrane fluidity, and alleviated blocking degree. But LIE(80, 100, 120 mg?L -1 )could decrease the content of MDA and speed of auto-oxidation markedly, improve the membrane fluidity and blocking degree significantly, and inhibit the lipid peroxidation. Conclusion LIE has protective action on the oxidative damage of erythrocyte ghost initiated by O?_2
7.Chondrogenesis of articular chondr ocytes following in vitro biphasic s eeding into three-dimensional scaffold

Objective To establish the technique of biphasic seeding of articular chondrocyte s in vitro for cartilage engineering in u sing a self designed biological gel a nd the three-dimensional scaffold a nd observe the efficiency of cartilage regeneration in vitro-tissue engin eered articular cartilage from cell scaffold complex.Methods The articular chondrocytes were iso lated enzymatically from the epiphyseal cartilage of young rabbits,and were then plated i nto the tissue culture flasks and were cultivated.The first passage ar-ticular chondrocytes were collecte d and mixed fully with the self-made l iquid biological gel-matrix at appr oxi-mately 2.5?10 7 cells /ml to form cell-gel fluid.The cell-gel fluid was dropped into the p orous CPPf /PLLA(calcium polyphosphate fiber /poly -L -lactic acid)scaffold,and a cell-gel-scaffold c omplex was formed after being solidified.The complexes were cultivated for 4weeks.The changes of complexes in morphology and synthesis of collagen typeⅡandⅠand aggregates were investigated under the gross and the phase and light microscope.The GAG sulfate content in complexes was quantitatively mea sured by the modified dimethyl-methylene blue method.Results1)After feeded,the porous CPPf /PLLA s caffolds were completely filled with the liquid chondrocyte-gel and the chondrocyte-gel-scaffold comp lexes were well formed by solidification.During the cultivation,the complex es could keep its original shape and m aintain the stable homogeneous three-dimensional distribution of chondrocytes in themselves without cell falling.In the same time,the com-plexes were gradually increasing th e consistency with the elasticity an d lubrication surface.2)The chondro-genesis began in the periphery area a nd extended to the central area of the complexes with the passage of cultivation period.After the 2nd we ek,the complexes were gradually reorganized into the mature engineered cartilage with typical cartilaginous histological structure,with ric h typeⅡcollagen and the strong typical het-erochromia to toluidine blue,but wi th gradually fading negative immunological stain of collagen typeⅠ.Meantime,the scaffold was graduall y de-graded in the complexes.The average content of GAG sulfate in the enginee red cartilages at 4th week was(15.70?2.00)mg /g(wet weight ),and was over 30%of the natural articular cartila ge.Conclusion The technique of chondrocytes biphasic seeding for three-dimensional scaffold has advantages of simple manipulation,fix ing cell stably in the scaffold,main-taining the original shape of the com plexes and the stable homogeneous th ree-dimensional distribution of chondrocytes in the scaffold,and ad vancing the regeneration and matura tion of tissue-engineered articula r cartilage.[

Cardiovascular and cerebrovascular diseases is the most fatal diseases in the world.The prevention and treatment of cardiovascular and cerebrovascular diseases are both basic and clinical focus.Aspirin has been used as a prevention medicine in cardiovascular and cerebrovascular diseases for over a century, which is the longest one in history.Aspirin use is estimated at 100 billion tablets annually as an analgesic, antipyretic and antiplatelet drugs.However, there are still many new findings about aspirin, such as aspirin resistance and aspirin hydrolase.This paper reviews the current research advances and future directions of aspirin in cardiovascular and cerebravascwlar diseases, mainly focuses on aspirin resistance and personalized medication.

10.Selective hepatic vascular exclusion for hepatectomy

Objective To explore the safety and efficacy of hepatic resection with selective hepatic vascular exclusion(SHVE). Methods SHVE was used in 246 consecutive patients undergoing major or complex liver resection in our center. Preoperative demographic and clinical data,details of the surgical procedure, pathologic diagnosis, postoperative course and complications were collected and analy zed. Results From January 2000 to July 2007, liver resections were performed IJnder SHVE in 246 patients; total SHVE, right partial SHVE and left partial SHVE in 145, 54 and 47 patients, respectively. SHVE was converted to total hepatic vascular exclusion(THVE)in 3 patients to repair the wall of inferior vena cava(IVC). Hemodynamic tolerance to SHVE was excellent, only with a slight increase in systemic and pulmonary vascular resistance during clamping.There were no deaths. and the morbidity was 24.8%.The mean leyth of hospital stay was 9.6 days(range 8-18) .Conclusion Our study showed that SHVE is a safe and effective procedurei and it is applicable to liver tumors near but not invade the inferior vena cava.
