3.Application and development of gene chip in the study of ischemia-reperfusion injury

Ischemia-reperfusion injury is a pathophysiological process caused by a number of gene expression changes,with the development of artery occlusion-reperfusion and surgery involved in vascular,such as thrombolytic therapy,cardiopulmonary bypass heart surgery and organ transplants,etc.it has been more and more important.Gene chip technology provides a powerful means for the systematic study of the mechanisms of incidence and prevention of ischemia-reperfusion injury,for its unique advantage in high throughput,high-efficiency,large-scale research.Since gaining importance in recent years,it has been widely used in various organs and tissues,and has achieved some progress.In this paper,the application of gene chip technology in the incidence and prevention of ischemia-reperfusion injury of various organs and tissues in recent years is given a certain summary.
5.Epigenetic modification in human leukemia.

Epigenetic modification, which involve DNA methylation, RNA-associated silencing and histone modification, is implicated in cell proliferation, differentiation, survival, apoptosis and malignant transformation. Some leukemogenesis has been shown to be aberrance of epigenetic modification. This paper discussed the potential causes of some of leukemias correlating with the methylation of cell cycle regulation genes, small interference RNA and modification abnormality of histone after translation. The study on epigenetic modification abnormality of leukemia cells provides a new strategy for treatment of leukemia.

7.Effects on the opposite structures of the minimally invasive transforaminal lumbar interbody fusion and unilateral fixation

ObjectiveTo investigate the changes of the opposite structures with the minimally invasive transforaminal lumbar interbody fusion and unilateral fixation.MethodsFrom January 2009 to January 2010,a total of 34 patients with low back pain underwent the minimally invasive transforaminal lumbar interbody fusion and unilateral internal fixation were retrospectively analyzed.Visual analogue scale (VAS) and Japanese Orthopaedic Association(JOA) low back pain scores were used to assess the outcome of the surgery.All patients were followed up for at least 6 months after surgery.The opposite structures of the segments with fusion and unilateral fixation were investigated with CT scans.CT imaging of the patients with opposite low back pain after surgery were analyzed for exploring possible causes of the pain.ResultsThere were significant differences between the pre- and post-operation in VAS and JOA scores(P<0.01).Four patients met pain in the opposite lumbosacral area after surgery.The CT scans showed that the rebuilding of the lordosis and disc height of the opposite side were more difficult.Rotation could happen in the progress of surgery.For the 4 patients with opposite low back pain after surgery,the insufficiency of the rebuilding of lordosis and disc height of the opposite side,and the rotation between the vertebras were more obvious,and changes were observed in the opposite facet joint on the post-operational CT scan of one patient.Conclusion The minimally invasive transforaminal lumbar interbody fusion and unilateral fixation are effective for patients with low back pain.It's important to pay more attention to the rebuilding of the lordosis and disc height of the opposite side,and the rotation of the vertebras in the surgery.
8.Prokaryotic expression of P gene from rabies virus and application of the indirect ELISA assay in the detection of its gene products

The complete length of P gene from rabies virus was amplified by RT-PCR using a pair of specific primers designed according to the relevant sequences from GenBank. The PCR product was cloned into cloning expression vestor pGM-T to obtain the cloning expressed plasmid pGM-T-P. After double-digestion by NotI and EcoRI, the product was transferred into prokaryotic expression vetor pET-32a(+)to obtain the prokaryotically expressed plasmid pET-32a-P. The target gene was then expressed in the E.coli BL21(DE3) cell with IPTG induction. The highest expression of target protein was analysed by SDS-PAGE, and the good immunoreactivity to rabies virus antibodies was proved by Western-blot analysis. By using purified protein, the indirect ELISA assay for the detection of rabies virus antibodies in canine serum was applied after management of the optional working condition.

10.Advances of GOLPH2 in tumors

As a novel Golgi protein,GOLPH2 expression is associated with maintaining the structural integrity of the Golgi apparatus.As an oncoprotein,GOLPH2 expression is associated with malignant biological behavior of cancer cells.Important roles of GOLPH2 in the tumorigenesis and metastasis make it become a new target for tumor diagnosis and treatment.
