1.Study on the expression of PCNA protein in pituitary adenoma

Objective To explore the relationship between the protein expression of PCNA and biological behavior science in benignant pituita ry adenoma.Methods The protein expression of PCNA in 58 patien ts with pituitary adenoma were determined by ABC immunohistochemical method.Results The PCNA index was significantly higher in the patients with recurrent pituitary adenomas than in nonrecurrent ones(P<0.05).There was no significantly difference between bleeding and unbleeding group,cystic and noncystic group,large type and unlarge type group(P>0.05 respectively) .Conclusion The protein expressions of PCNA reflected the proli ferative activties of pituitary adenomas, and could be taken as one of the indic ators to evaluate recurrence and prognosis of the tumor.
3.Construction of CD44 gene overexpression lentiviral vector and its expression in LNCaP cells

Objective To construct an overexpression lentiviral vector of CD44 and obtain an androgen-dependent prostate cancer LNCaP cell line with CD44 overexpression.Methods CD44 gene sequence was obtained by PCR amplification.The synthesized oligonucleotides were cloned in the lentiviral vector LV5 after digested with NotI and NsiI enzyme.Recombinant plasmids were verified by enzyme digestion analysis and sequencing,and were transfected into 293T package cells.Supernatant was collected and concentrated to obtain lentivirus solution at a high titer.The titer of virus was identified by multiple proportion dilution methods.Then the recombinant viruses were transfected into LNCaP cells.Results Fluorescence intensity in infected LNCaP cells was increased,detected by a fluorescence microscope.And QRT PCR and Western blot results showed that the expression of CD44 was significantly increased(P<0.05).Conclusions The overexpression lentiviral vector of CD44 is successfully constructed,and the LNCaP cell line with CD44 overexpression is obtained.

Objective To study the volume of residual urine as related to the surgical indication for BPH. Methods Urodynamic study was carried out in 103 BPH patients who were assigned to group A (no residual urine),group B (residual urine0.05) but significantly different between group C and A or between B and A (P60 ml should be strictly followed.Early surgical intervention should be undertaken if the bladder ontlet obstruction or bladder function deteriorated by BPH causing residual uine.Other causes such as neurogenic should be ruled out before surgery.
10.Effects of Modified Sanxiang Decoction on Gastrointestinal Transmission Function of Patients with Functional Bloating

Objective To study the effects of modified Sanxiang Decoction on gastrointestinal transmission function of patients with functional bloating (FB). Methods Sixty cases of FB patients were randomly divided into treatment group (40 cases) and control group (20 cases). Treatment group was given modified Sanxiang Decoction, while control group was given placebo granules, one bag each time. Twice a day, taking it after mixing it with water. Gastrointestinal symptoms, 48 h GITT, and adverse reactions were recorded. Results There was statistical significance in TCM symptom scores between before and after treatment in the two groups (P<0.05). After treatment, the total score of abdominal distension, belching, anorexia and TCM symptoms of the treatment group were lower than the control group, with statistical significance (P=0.000, 0.000, 0.010, 0.003). The total effective rate was 82%(33/40) in the treatment group, 30%(6/20) in thecontrol group;the treatment group was better than the control group (P<0.05). A total of 30 patients completed gastrointestinal transit test, of which delayed 48 h GITT pass rate accounted for 54% (16/30). Compared with the control group, the 48 h GITT of the treatment group was more obviously improved than that of the control group with statistical significance (P=0.002). There were no significant adverse reactions in both groups. Conclusion Most of FB patients have the problem of abnormal intestinal motility. Modified Sanxiang Decoction can improve the symptoms of patients with FB, promote gastrointestinal motility, and improve intestinal transmission function.
