1.Percutaneous transluminal stenting for stenosis of internal carotid artery: a report of 83 cases

ObjectiveTo evaluate the methodology of percutaneous transluminal stenting for stenosis of internal carotid artery(ICA).Methods83 patients with ICA stenosis greater than 85% received antiplatelate therapy perioperatively, and underwent cerebral vascular angiography as well as cervical Doppler sonography. Four kinds of stents were used with or without predilation.ResultsThere were not procedure related morbidities. All procedures were successful based on angiography. Cerebral blood flow (CBF) restored to normal or significantly improved. Symptoms disappeared in 56 cases and ameliorated in 27 cases. Follow up by angiography and sonography at 3 to 24 months showed no distortion of stents and restenosis. ConclusionsPercutaneous transluminal stenting combined with perioperative medical therapy is a safe therapeutic method for stenosis of internal carotid artery.
2.Effect of 3-n-butylphthalide pretreatment on expression of the HSP70 after brain ischemia/reperfusion.

OBJECTIVETo explore the effect of 3-n-butylphthalide pretreatment on the delayed neuronal death(DND) and the expreesion of heat shock protein70 (HSP70) in rat hippocampus after ischemia/ reperfusion.
METHODSAll rats were randomly divided into sham group (n = 36), total cerebral ischemia (TCI) group (n = 36), butylphthalide (NBP) group (n = 6), NBP + TCI group( n = 36), quercetin + NBP + TCI group (n = 6), dimethyl sulfoxide (DMSO) + NBP + TCI group (n = 6). The model of total cerebral ischemia/reperfusion was established by blocking vertebral arteries and carotid arteries. In sham group, TCI group and NBP group, the animals were further divided into instantly, 6 h, 12 h, 1 d, 3 d, 5 d groups according to the time interval after sham operation or TCI. Histological changes of the hippocampus were evaluated using thionin staining under light microscope by determining the delayed neuronal death (DND) and the expression of HSP70 was assayed using immunohistochemistry.
RESULTSNBP pretreatment could reduce delayed neuronal death in CA1 of hippocampus induced by TCI-reperfusion injury in rats, and up-regulated the expression of HSP70 in CA1 hippocampus of brain ischemic/reperfusion for 5 days. Quercetin blocked the acquirement of the brain ischemic tolerance induced by NBP preconditioning.
CONCLUSION3-n-butylphthalide (NBP) prevents the neurons from ischemia/reperfusion injury through upregulating the expression of HSP70.

3.Establishment of a multidrug resistant human salivary gland adenoid cystic carcinoma cell line

Objective:To establish a multidrug resistant human salivary gland adenoid cystic carcinoma cell line.Methods:Human salivary gland adenoid cystic carcinoma cell line ACC-2 was treated by 24-hour-exposure to high dose of Bleomycin(BLM)(20 ?g/ml).Drug sensitivity was evaluated by MTT assay.Cell counting was employed to make the growth curve and to calculate the cell doubling time.Flow cytometry(FCM) was used to study the cell cycle distribution and apoptosis.The colony formation ability was also observed.Results:Multidrug resistant cell line of human salivary gland adenoid cystic carcinoma was established and named ACC-2/BLM.After 10 times repeated exposure to BLM,the resistance index(RI) to BLM,5-Fluorouracil(5-FU),Cisplatin(CDDP),Cyclophosphamide(CTX),Vincristine(VCR) were 7.299,1.03,2.15,1.114 and 5.96 respectively.Compared with ACC-2,the proliferation potential of ACC-2/BLM cells was decreased.The ACC-2 apoptosis cells were much more than ACC-2/BLM cells after 9-day-treatment by BLM at 60 ?g/ml.Conclusion:ACC-2/BLM cell line has multidrug resistant characteristics.
4.Investigation on the influencing factors of free education for rural order-oriented medical students

Objective:To understand the influencing factors of free education for rural order-oriented medical students and to explore the willingness of students to serve at the grass-roots, so as to provide reference for further optimizing the free education of medical students.Methods:A self-designed questionnaire survey was performed among 196 medical undergraduates majoring in free clinical medicine in one medical university of Shandong Province in March 2018 and 198 undergraduates majoring in general clinical medicine enrolled in the same year. The results of the survey were analyzed by SAS 9.2, and chi-square test, rank sum test and logistic regression were used to make statistic analysis.Results:The factors such as score of the national university entrance exam (wald χ2 = 52.309, P < 0.001), household registration location (wald χ2 = 9.304, P = 0.002), whether the only child in the family (wald χ2 = 12.608, P < 0.001), average monthly income of the family (wald χ2 = 5.557, P = 0.018), decision makers (wald χ2 = 15.206, P = 0.002) and willingness to serve at the grass-roots medical institutions (wald χ2 = 67.832, P < 0.001) had influence on medical students' choice of free education for order directional medical students. There were 113 (57.65%) of the free medical students intended to work at the grass-roots, while only 16 (8.08%) of the general medical students who were willing to work at the grass-roots. Conclusion:The implementation of free education for order-oriented medical students policy can satisfy the wishes of some people who have the willingness to serve at the grass-roots, but their family income and college entrance examination scores are not high. However, there are still some problems, which need the joint efforts of the government health departments and the training colleges and universities, and the corresponding follow-up support policies should be issued by the state at the same time.
6.Cellular Apoptosis of C6 Mouse Glioma Cells Induced by hING4 Mediated by Adenovirus

The known members of inhibitor of growth (ING) gene family are considered as candidate tumor suppressor genes. ING4, a novel member of ING family, is recently reported to regulate brain tumour angiogenesis through transcriptional repression of NF-?B-responsive genes, induce G2/M arrest by the increased p21 expression in a p53-dependent manner, suppress the loss of contact inhibition and represses activation of the hypoxia inducible factor, which plays an important role in the progression of tumorigenesis. However, seldom studies about ING4 inducing tumor cells apoptosis were reported.The C6 cells (mouse glioma cells) were infected respectively with the blank adenovirus carrying GFP (Ad) and the recombinated Ad-hING4-His, then RT-PCR assay was used to detect the transcriptions of hING4, as well Western-blotting assay was ued to detect the expressions of hING4. The effects of hING4 expression upon C6 cells were observed, and the growth curve was drawed and tumor control rates were calculated. The C6 cells, which were affected by blank Ad and Ad-hING4-His, were respectively observed by LSCM (laser scan confocal microscope) and transmission electron microscope (TEM), detected by flow cytometry; and the genomic DNA of both groups were extracted and electrophoresised in agarose gel to examinate the DNA fragments. The results showed hING4 can significantly inhibit the growth of C6 cells by promoting the cell’s apoptosis, which probably is the first one to prove this property of ING4.The experimental and theoretical foundation for gene therapy for gliomas with ING4 in the future was established.
7.Bioinformatics Analysis on Molecular Network and Bio-function ofBu-Fei-Yi- Shen Decoction

This article was aimed to study the molecular network and bio-function ofBu-Fei-Yi-Shen (BFYS) decoction for chronic obstructive pulmonary disease (COPD) by bioinformatics analysis, in order to provide new ideas for research on pharmacological mechanism of Chinese medicine compound prescription. Components of herbs in BFYS decoction were searched in the databases. Targeted proteins of each component were found from PubChem. Comparison analyses were performed on molecular network, bio-function and canonical pathways by Ingenuity Pathway Analysis (IPA). The results showed that there were 239 target proteins of BFYS decoction. There were 9 molecular networks of BFYS decoction. The top 3 networks' functions were Cellular Development, Energy Production, and Cancer. The top 3 bio-function of BFYS decoction were Cellular Growth and Proliferation, Cell Death and Survival, and Inflammatory Response. The top 3 canonical pathways of BFYS decoction were Cell Cycle:G1/S Checkpoint Regulation, Chronic Myeloid Leukemia Signaling, and Cyclins and Cell Cycle Regulation. It was concluded that the search of target proteins for herbal compounds and bioinformatics analysis by IPA can be used to reveal the molecular network and bio-function of BFYS decoction.
8.The clinical significance of plasma resistin levels in patients with acute coronary syndrome

Objective To explore the association between plasma resistin levels and acute coronary syndrome. Methods Four hundred patients were divided into coronary heart disease (CHD) group(310)and control group(90)according to the coronary Angiography (CAG). And CHD group was divided into ACS subgroup(n=217)and SAP subgroup(n=93)according to the clinical information. 85 cases in CHD group were underwent 64-slice spiral computed tomography coronary artery imaging. The severity and extent of coronary lesions were analyzed by CAG and graded by means of Gensini coronary score system. Resistin level in plasma of all patients was determined by enzyme linked immunosorbent assay. Results Resistin levels in CHD group[(889.1±248.2)pg/ml] were significant higher compared with the control group[(261.6±111.9)pg/ml] (P<0.05), and resistin levels in ACS subgroup[(1260.0±368.0)pg/ml] were much higher than that in SAP subgroup[(518.3±128.4)pg/ml] (P<0.05). Conclusions The resistin levels of patients with acute coronary syndrome increased significantly and might be associated with the vulnerable plaque. Resistin levels and 64 slice spiral computed tomography coronary artery imaging can be used to detect the vulnerable plaque in CHD patients.
9.Endovascular mechanical recanalization of subclavian artery total occlusion

Objective To discuss the skills and effects of several endovascular mechanical techniques for the recanalization of subclavian artery total occlusion. Methods Endovascular mechanical recanalization of subclavian artery total occlusion was performed in 32 patients with symptomatic subclavian artery total occlusion. The re-open rate and the therapeutic results were observed and analyzed. Results Several endovascular mechanical techniques, including percutaneous transluminal angioplasty, were employed in treating 32 patients with subclavian artery total occlusion. After the procedure, the ischemic 8ymptoms of posterior circulation and/or upper extremity were markedly relieved. Conclusion It is safe and feasible using appropriate endovascular mechanical technique for re-canalizing the occluded subclavian artery.
10.Assessment and direction of Objective Structured Clinical Examination on clinical practice in Department of Obstetrics and Gynecology

Objective To analyze the results of the Objective Structured Clinical Examination(OSCE) of medical interns in Department of Gynecology and Obstetrics,and to assess the role of OSCE in clinical teaching and practice. MethodsAtolal of 104 medical interns of five-year education program in 2002 and seven-year education program in 2003 in School of Medicine had taken part in OSCE of gynecology and obstetrics.The OSCE consisted of six stations: gynecologic examination,obstetric examination,oral test,interrogation of standardized-patients(SP),drawing partogram,and non-stress test(NST) explanation.After gathering the data of each station,the software of SPSS 11.0 was used to make statistical analysis. Results The median scores were as follows: 92.0 for gynecologic examination,91.0 for obstetric examination,83.5 for oral test,80.0 for interrogation of SP,80.0 for drawing partogram,and 70.0 for NST explanation.There was no significant impact of familiarity to OSCE on the results of the test.The results showed that medical interns did good job in basic performance,but lack of clinical practice skills in some complicated practice of gynecology and obstetrics.Conclusion The grading system and stations setup of OSCE in Department of Gynecology and Obstetrics in Shanghai Sixth People's Hospital show objectivity and equity.The OSCE can be widely applied in the after-department examination and even graduation examination.
