1.Purification of epidermal melanocytesin culture
Tianjin Medical Journal 2017;45(1):5-8
Objective To compare the influence of different coating materials and cultural conditions on the purification and growth of human epidermal melanocytes. Methods The full-thick foreskin, epidermis and cell suspension obtained from human foreskin were cultured in the plates, which were precoated with matrigel or laminin respectively. When having reached 80%-90%confluence, the cells were treated with 0.05%trypsin-EDTA for 4 minutes and resuspended in M254 medium, which were supplemented with G418 and 5-BrdU, respectively. The purity of melanocytes was observed by an immunofluorescence staining with melanocyte markers. Results During the primary culture, the cell suspension generated more cells at faster speed compared with those of skin explants and epidermal specimen. Moreover, the epidermis released cells earlier and proliferated quickly over skin explants. The melanocytes in the plates coated with laminin other than with matrigel displayed faster and better growth. The unwanted keratinocytes and fibroblasts were removed by using differentiation trypsinition combined with supplement of G418 or 5-BrdU. Conclusion Using a plate coated with laminin to culture cell suspension from human foreskin, and via a differentiation trypsinization combined with supplement of small doses of G418 to subculture the cells, is advantageous to the melanocyte purification, without affecting their growth.
2.Advances in chemotherapy of small cell lung cancer
Chinese Journal of Clinical Oncology 2016;43(10):418-423
Small cell lung cancer (SCLC) accounts for about 13%of lung cancer. SCLC exhibits an early metastatic potential and sensitivi-ty to first-line cytotoxic chemotherapy compared with non-small cell lung cancer. Current treatments include surgery, radiotherapy, and chemotherapy. Platinum plus etoposide is viewed as the standard first-line chemotherapy. However, recurrence rate of cancer af-ter platinum plus etoposide chemotherapy is high. Topotecan monotherapy is the standard second-line chemotherapy. Meanwhile, novel targeted immunotherapy showed poor effects. Hence, new breakthroughs in the treatment of SCLC are urgently needed.

5.Analysis of Risk Factors for Chronic Renal Insufficiency in the Patients with Chronic Heart Failure
Journal of China Medical University 2015;44(8):725-729
Objective To elucidate the related risk factors for the cardiorenal syndrome(CRS),and to provide a reference for the prevention and treatment of CRS. Methods A retrospective study was conducted on 269 patients with chronic heart failure. The subjects were divided into two groups in accordance with estimated glomerular filtration rate(eGFR):patient group(CRS group),eGRF<60 mL/(min ! 1.73 m2)and control group,eGRF≥60 mL/(min!1.73 m2). The patients'general information as well as the related clinical lab indices and accessory examination indi-ces were collected. The risk factors for chronic heart failure and renal insufficiency were analyzed with multivariate logistic regression analysis. Re?sults The prevalence of chronic heart failure patients suffering from CRS was 26.39%. The single factor analysis showed that there were statistically significant differences between the two groups in terms of age, body weight, history of hypertension, and history of diabetes(all P<0.05). Compared to the control group, uric acid, cystatin C,serum urea nitrogen, brain natriuretic peptide, glycosylated hemoglobin,high-sensitivity C-reactive pro-tein,and left ventricular end-diastolic diameter in the CRS group were significantly higher;white blood cell count, hemoglobin, albumin, and left ventricular ejection fraction were significantly lower(all P<0.05). There was no statistically significant difference(P>0.05)between the two groups in regard to white blood cell count,glutamic-pyruvic transaminase, glutamic oxalacetic transaminase, cholesterol, triglycerides, high density lipoprotein, low density lipoprotein, troponin I,serum free triiodothyronine, serum free thyroxine,or thyroid stimulating hormone. Multivariate logis-tic regression analysis showed that older age,low body weight, history of hypertension, history of diabetes, hypoalbuminemia, superb sensitivity C-re-active protein levels, high uric acid, high cystatin C hyperlipidemia, and low ejection fraction were independently associated with the occurrence of CRS. Conclusion Older age,low body weight, history of hypertension, history of diabetes,hypoalbuminemia,high-sensitivity C-reactive protein levels, high uric acid, high cystatin C hyperlipidemia, and low ejection fraction are the high risk factors for CRS in heart failure patients and are inde-pendently associated with the occurrence of CRS. Early identification and control of these risk factors are important for the prevention and treatment of CRS.

OBJECTIVETo summarize the clinical manifestation and diagnosis of ganglioneuroma in spine and investigate the clinical effect of surgical treatment.
METHODSThe clinical data of 6 patients underwent a surgery for ganglioneuroma in spine from January 2008 to January 2015 were retrospectively analyzed. There were 4 males and 2 females, aged from 2 to 63 years old with an average of 34.6 years. The courses of disease were from 3 days to 17 years. Five patients complicated with superficial hypesthesia in correlative level of tumor, and the muscle strength under tumor plane had decreased at different levels, with the strength of grade II-IV. Two cases complicated with hypermyotonia and positive bilateral Hoffmann's and Babinski sign. Five cases were sporadic lesion in correlative spinal canal and one case complicated with the giant occupying lesion in thoracic cavity.
RESULTSSix operations had been performed including 5 en bloc and 1 subtotal resection. Postoperative pathological results showed tumor cells scattered or fasciculated inserted into Schwann cells in the stroma. In 2 patients complicated with radiculalgia before operation, 1 case was relieved and 1 was invariant after operation. All 4 patients with preoperative dyscinesia in the limbs obtained improvement after operation. All the patients were followed up from 0.3 to 6.8 years with an average of 2.5 years. At the final follow-up, according to ASIA grade, 5 cases were good and 1 case was invariant. During the follow-up, only 1 patient experienced chemoradiation because of merging ganglioneuroblastoma and receiving subtotal resection. No recurrence in other 5 cases.
CONCLUSIONGanglioneuroma is a benign and rare tumors in spine. Clinically, radicular pain and sensory-motor disorders are the main manifestations. Its diagnosis depends on pathological examination. Prognosis of surgical treatment is good.
