1.Accurate clinical examination guarantees the whole management process of hepatitis C patients in the era of direct-acting antiviral agents

Hepatitis C virus (HCV) infection is an important public health issue around the world.Direct-acting antiviral agents (DAAs) have become an important method to reduce HCV infection.DAAs can improve the clinical outcome of antiviral therapy,shorten the course of treatment,and reduce complications and have a wide range of application;therefore,they have great advantages over conventional therapies.Accurate clinical diagnosis has increasing importance in the era of DAAs and guarantees the whole management process of chronic hepatitis C patients from the aspects of disease diagnosis,therapeutic regimens,outcome evaluation,and long-term prognosis monitoring.
2.Homology Study of Resistant Gram-negative Bacilli Isolating from Hospitalized Patients,Hospital Environment and Hands in Intensive Care Unit

OBJECTIVE To study the homology of resistant Gram-negative bacteria isolating from the hospitalized patients,hospital environment and hands of medical staffs in intensive care unit(ICU).METHODS To collect and identify 58 clinical resistant Gram-negative bacteria isolates and environment isolates from intensive care units,the same type of Gram-negative bacilli were studied on the homology using plasmid profile,restriction endonuclease profile and enterobacter repetitive intergenic consensus PCR(ERIC-PCR).RESULTS The homology existed between the resistant bacteria of clinical isolates and environment specimens.CONCLUSIONS We found that the multidrug-resistant Gram-negative bacteria derive from a common origin and cause an outbreak in ICU.So medical staff must emphasize the concept of hand hygiene and reinforce the measure of cleaning and sterilization,in order to control nosocomial infection.

4.Cervical Loop Electrosurgical Excision Procedure in the Treatment of Cervical Intraepithelial Neoplasia

Objective:To evaluate the efficacy of loop electrosurgical excision procedure(LEEP) in the treatment of different grade of cervical intraepithelial neoplasia(CIN). Methods:898 cases with CIN treated by LEEP were retrospectively analyzed. Operation time, blood loss during the operation, changes of histopathologic results and high-rick HPV before and after the operation were studied. The diagnosis and treatment results by LEEP were evaluated. Results: The average operation time was 5.1 minutes. The average blood loss during the operation was 5.6 ml. The concordance rate of pathologic results under colposcope before operation and after LEEP was 77.73% (698/898) .96 cases had increased CIN grade after LEEP(10.69%). The HPV-DNA loading dose after operation declined significantly (P< 0.01). Conclusions: The detection rate of invasive cervical cancer by LEEP is higher than multiple punch biopsy under electronic colposcope. LEEP is an effective method for the diagnosis and treatment of CIN, and it can eliminate high-rick HPV at the same time of ablating the cervical lesion.
5.The method and evaluation of CAD/CAM system introduced in the prosthodontics experimental courses

ObjectiveTo investigate the effectiveness of introducing CAD/CAM system into Prosthodontics experimental teachin.MethodsStudents from Chongqing Medical University with major of Dental Department in Grade 2007 was selected.To make all students master the basic knowledge of CAD/CAM system,problem-based learning was used to teach the undergraduate grasping the CAD/CAM system of the affiliated hospital.Results 90 % of the students showed strong interest in learning knowledge of the CAD/CAM system.98 % of the students could master CAD/CAM system knowledge.ConclusionCAD/CAM system knowledge is suitable for development of prosthodontics.It is necessary that the basic knowledge of CAD/CAM was introduced into prosthodontics experimental courses.
7.Role of endoplasmic reticulum stress in high fattyacid induced injury in cardiomyocytes

Aim To explore the role of endoplasmic reticulum stress (EPR) in high fatty acid induced injury in cardiomyocytes.Methods Cardiomyocytes were exposed to different concentrations of palmitic acid (0, 0.1, 0.2, 0.4 mmol·L-1) for 24 h and different time points of high palmitic acid (0.2 mmol·L-1) (12, 24, 48 h).Cell viability was measured by CCK8, and the protein expressions of EPR such as GRP78, CHOP, PERKphos, IRE1phos, ATF6 and apoptosis related proteins such as Bcl-2 and Bax were determined by Western blot.Results When the cells were stimulated with palmitic acid (0.1~0.4 mmol·L-1) for 24 h, the cell proliferation rates of 0.2 and 0.4 mmol·L-1 groups were significantly decreased.Cardiomyocytes exposured to high palmitic acid (0.2 mmol·L-1) for 24 h showed an increase in theexpression of EPR related proteins (GRP78, CHOP, PERKphos, IRE1phos and ATF6) and Bax(P<0.05), while Bcl-2 expression was significantly reduced.Pretreatmented with EPR inhibitor pravastatin(10 mol·L-1) significantly increased high palmitic acid induced Bcl-2 expression (P<0.05) and significantly decreased high palmitic acid induced Bax expression (P<0.05).Conclusion Activated EPR might play an important role in treatment of high fatty acid induced myocardial injury.
8.Video-urodynamic Characteristics and Management in Infra-sacral Cord Injury: 169 Cases Report

Objective To discuss the video-urodynamic characteristics and management in infra-sacral cord injured patients. Methods The video-urodynamic finding and managements in 169 patients was reviewed. They were followed up for 2 years. Results Detrusor areflexia was found in 76.33% (129/169) patients, among them there were 27.22% (46/169) patients appeared hyper-compliance, and 7.10% (12/169) patients appeared reflux. The video-urodynamic characteristics showed over activity and low-compliance in 12.43% (21/169) patients, including reflux in 5 cases. Different management was chosen according to the video-urodynamic examination. No special complication was observed after 2 years follow-up. Conclusion The majority patients with infra-sacral cord injury appeared detrusor areflexia and hyper-compliance. Special management according to the video-urodynamic characteristics is important for protecting upper urinary tract and preventing urinary system infection.
9.Expression of translationally controlled tumor protein in squamous cell carcinoma tissue and cell lines A431 and SCL-1

Objective To detect the expression of translationally controlled tumor protein (TCTP) in squamous cell carcinoma (SCC) tissue and cell lines A431 and SCL-1,and to evaluate the effect of TCTP on apoptosis in and proliferation of SCC cells.Methods An immunohistochemical method was used to measure the expression of TCTP in tissue specimens from 65 patients with SCC.Western blot was performed to detect the expression of TCTP in A431 and SCL-1 cells.Three small interference RNAs (siRNAs) targeting the TPT1 gene were designed,synthesized,and transfected into A431 cells.Then,reverse transcription (RT)-PCR and Western blot were conducted to measure the expression of TPT1 mRNA and TCTP,respectively,methyl thiazolyl tetrazolium (MTT) assay and flow cytometry were carried out to detect cell proliferation and apoptosis,respectively.Results TCTP was overexpressed in SCC tissue specimens,and the expression level was positively correlated with the histologic grading of SCC (P < 0.05).Western blot showed that TCTP was expressed in both A431 and SCL-1 cells,and the expression was relatively high in A431 cells.The transfection efficiency of siRNAs varied from 90% to 95%.A decrease in the expression of TPT1 mRNA and TCTP was induced by the siRNAs in A375 cells (all P < 0.05).Conclusion The downregulation of TCTP expression may increase the apoptosis in and suppress the proliferation of A431 cells.

Scientific innovation teams are key weight indicators and promoters in science and technology of a hospital.The First Affiliated Hospital of Anhui Medical University(the Hospital)draws funding support from the Natural Science Foundation,to empower its innovative teams with programs and funding.These efforts have effectively propelled its innovations spearheaded by the dermatology team to give rise to outstanding outcomes and power teams.The application for talents support from NSF,the project approval for the hospital funding is affected to some extent.With the help from NSF and the innovative teams,scientific teams of the hospital have harvested a general upgrading and accelerated development of disciplines,elevating the scientific level of hospital in general.
