1.EFFECTS OF ANGIOTENSIN Ⅱ ON PAI-1 GENE EXPRESSION IN HUMAN GLOMERULAR MESANGIAL CELLS AND ITS MECHANISM

To explore the effects of angiotensin Ⅱ on plasminogen activator inhibitor 1 (PAI 1) gene expression in human glomerular mesangial cells.Glomerular mesangial cells (GMCs) were cultured from a healthy adult human kidney (unsuitable for renal transplantation) and then treated with angiotensin Ⅱ at concentration of 10 -9 ,10 -8 , 10 -7 and 10 -6 mol/L, respectively. The mRNA expressions of angiotensinⅡAT1 and AT2 receptor, and PAI I were examined with RT PCR and Northern blot analysis, respectively. Plasminogen activator activity in the supernatants of the GMCs was measured with fibrin plate method. After the GMCs were stimulated by angiotensin Ⅱ, the mRNA level of the AT1 receptor was markedly enhanced, but the AT2 receptor mRNA level was still undetectable as that in normal condition. When GMCs were treated with angiotensin Ⅱ for 48 hours, PAI 1 mRNA expression was increased in a dose dependent manner. The plasminogen activator activity was significantly reduced in the supernatants of the angiotensinⅡ treated GMCs. the results suggested that angiotensinⅡ promote PAI 1 gene expression of the GMCs possibly through its AT1 receptor.

5.The effect of Leptin on the proliferation of human circulating T lymphocytes

Objective:To investigate the effect of human Leptin on the proliferation of human circulating T lymphocytes in vitro and the influence of Leptin on the action of PHA on the proliferation of human circulating T lymphocytes.Methods:PBMC were isolated from hepartinized venous blood of normal donors by density-gradient sedimentation over Ficoll-Hypaque and cultured in the 1640 complete medium.T lymphocytes were then obtained by depleting the monocyte and B cell populations.Cells were then cultured in the 1640 complete medium for 56 h in the presence of PHA.The proliferative effect was assessed by means of incorporation of thymidine.TdR was added and cultured for 16 hours.Cells were then processed in the harvester to measure the incorporation of [()~3H] thymidine.Radioactivity was determined by liquid scintillation counter.Results:The results showed that Leptin alone did not affect the proliferation of T lymphocytes,but it could enhance the effect of PHA on the proliferation of T lymphocytes at the concentrations of PHA from 2-8 ?g/ml.A dose-response studies of T lymphocyte proliferation showed that the maximal effect of Leptin were observed at 10 nmol/L.Leptin did not affect the proliferation of T lymphocytes when the concentrations of PHA were over 8 ?g/ml.Conclusion:The studies demonstrated that Leptin alone did not affect the proliferation of lymphocytes,but it could enhance the action of PHA on cell growth in a dose-dependent manner.
6.Effect of SHEN-YI,a Chinese herbal formula on ratio of Th1/Th2 cells within immune organs of MRL/lpr lupus mice

Objective:To define the effects of SHEN YI,a Chinese herbal formula on ratio of Th1/Th2 cells within immune organs of MRL/lpr mice Methods:MRL/lpr mice were administered daily with the Chinese herbal formula from the age of 12 weeks and both the treatment and control groups were killed at the age of 28 weeks The percentage of CD4 +CD30 -(Th1),CD4 +CD30 +(Th2)cells within thymus and spleen were determined in vitro by FACScan with or without PHA P stimulation The CD4 + T cells selected from splenocytes were stimulated with PHA P, then, RT PCR was performed to detect IL 4/IFN ? mRNA expressed by the selected cells In addition, the ratio of the two cytokines expressed by the CD4 + T cells from the control and treatment groups was calculated respectively Results:CD4 +CD30 + T cells nearly can't be detected within Thymus Without PHA P stimulation, there was no difference in percentage of CD4 +CD30 + T cells between the control and treatment groups After stimulation with PHA P, FACScan showed an increment in the percentage of CD4 +CD30 + T cells both in the control and treatment groups, however, the extent of expansion of CD4 +CD30 + T cells percentage in treatment group was lower than it in the control one RT PCR showed a reduction in the ratio of IL 4/IFN ? mRNA in the treatment group compared to it in the control one Conclusion:The Chinese herbal formula can inhibit Th cells to differentiate into Th2 cells in the spleen under the stimulation of PHA P
7.Analysis on Monitoring and Measuring Data of Plasma Concentration of Antiepileptic Drugs of 432 Cases in Our Hospital

60 years old). 43 cases of antiepileptic drugs combination accounted for 10% and the therapeutic plasma concentration of 27 cases deviated from normal range(62.8%). CONCLUSION:Results of plasma concentration monitoring provide an important basis for clinical drug use. Monitoring data and other clinical index can promote rational use of antiepileptic drugs.

Objective To explore the differences of the joint function and the patient's satisfaction between two different methods to reconstruct limb length for unilateral Crowe Ⅳ developmental dysplasia of the hip (DDH).Methods The clinical controlled study were used.21 cases with unilateral DDH Crowe Ⅳ treated with total hip arthroplasty were divided into 2 groups randomly.The patients in the first group were reconstructed the limb with the compensatory length and those in the second group were did with the real length.The patients were followed up 10 years.Two independent sample t-test was used to compare these two operations.Harris scores and SF-36 scores were used in the test.Survival analysis with Kaplan-Meier method was involved in the survival of the prosthesis and then test with Log-rank test.X-ray films of different period were compared to confirm the prosthetic loosening.Results Seventeen patients were followed up 8-10 years.There were no significant differences both in Harris score and most of the SF-36 scores at the median follow-up of 10 years.The item mental healthy of SF-36 expressed significant difference.Five patients required revision.There were 10 cases suffered with polyethylene wear,6 cases with severe osteoporosis in greater trochanter,and 3 cases with osteolytic reaction.Conclusion There are no significant differences between the two surgical method in the survival rate of prosthesis and the joint function in the median follow up.The satisfaction of the control groups is lower than the trial groups both at the early stage after the operation and the median follow up.The revision rate between the two groups is similar but the reason is different.

10.Cytokines Combined with Striatal Conditioned Medium Promote Directional Differentiation of Mesenchymal Stem Cells into Dopaminergic Neurons in Vitro

Objective To explore the possibility of basic fibroblast growth factor(bFGF)and epidermal growth factor(EGF)combined with striatal conditioned medium promoting the directional differentiation of rat bone marrow mesenchymal stem cells(BMMSCs)into dopaminergie neurons.Methods 1.Separation and culture of BMMSCs:BMMSCs were harvested from healthy adult Wistar rats for serial subcultivation.2.Preparation of Striatal conditioned medium:newborn Wistar rats within 24 hours were selected,and their brain tissues were removed to prepare striatal conditioned medium.3.Induced differentiation of BMMSCs:the 5th passage BMMSCs were collected and pre-induced in low glucose-Dulbecco's modified eagle medium(L-DMEM)containing bFGF and EGF.Twenty-four hours later,pre-induction liquor was replaced with striatal conditioned medium for further induced differentiation.4.Result assessment:the morphological changes of stem cells were observed under inverted phase microscope.The expression of neuron specific enolage(NSE)and tyrosine hydmxylage(TH)were identified by immunocytochemical technique.Results The cell body of rat BMMSCs contracted into round and spindle shape after induction by bFGF and EGF combined with striatal conditioned medium.Partial neuron-like cells with prominence could be found.Immunocytochemieal detection showed that the percentages of NSE and TH positive cells were(72.70±14.81)% and(34.50±15.93)%,respectively.Conclusion BMMSCs can be induced directionally into dopaminergiC neurons by bFGF and EGF combined with striatal conditioned medium in vitro.
