Objective To make use of the characteristics of presenting and processing tumor antigen of Dendritic cells pulsed with recombinant human Hsp70 and Mage-3 peptide to enhance killing capability of breast neoplasms. Methods Autologous dendritic cells were isolated from PBMC and then stimuiated in vitro with GM-CSF and IL-4.Dendritic cells were loaded with Mage-3 peptide;at the same time Hsp70L1 was added. The specific groupings each induce generation of tumor specific cytotoxic assay with ELISA. Results DCs pulsed with Mage-3 peptide enhanced the growth expansion of CTL, and promoted the secretion of cytokines such as TNF-αand IL-6. DCs pulsed with recombinant human Hsp70 and Mage-3 peptide had cytotoxicity against human breast cancer cells MCF-7.Conclusions DCs pulsed with recombinant human Hsp70 and Mage -3 peptide have higher biological activities. Modified DCs can stimulate the proliferation of lymphoeytes,present effectively tumor antigen to stimulate generation of speeific CTLs. Dendritic cells pulsed with recombinant human Hsp70 and Mage-3 peptide have high ability to kill breast cancer cells.

Objective:This work aims to use the characteristics of dendritic cells (DCs) pulsed with recombinant human HSP70, which can present and process tumor antigens, to enhance the killing activity of cytotoxic t lymphocytes (CTLs) against breast neoplasms. Methods:Autologous DCs were isolated from peripheral blood mononuclear cells and then stimulated in vitro with granulocyte macrophage-colony stimulating factor and interleukin-4. The DCs were loaded with A549 tumor cell freeze-thaw lysate, and rhHSP70 was added as an immune adjuvant. The specific groups were subjected to tumor-specific cytotoxic assay, enzyme-linked immunosorbent assay, and fluores-cence-activated cell sorting. Results:DCs pulsed with A549 tumor cell lysate enhanced the growth expansion of CTLs, upregulated CD40 and CD80 populations in CTLs, and augmented Th1 cytokines. In addition, the cytotoxicity of specific CTLs against A549 was highly enhanced. The above indications became more obvious after the addition of rhHSP70. Conclusion:DCs pulsed with freeze-thaw cell lysates derived from breast cancer can enhance growth expansion of lymphocytes. They may serve as an effective tumor antigen to stimulate the proliferation of specific CTLs, which are very effective in activating specific T-cell responses against breast cancer cells in vitro. The improved anti-tumor immunity response by DC-based vaccines may be related to the maturation of the DCs promoted by rhHSP70.

Objective:To explore the directed forgetting ability of elder people to disease/death-related words under the conditions of the neutral and negative emotion.Methods:Thirty-six elder people and thirty-seven young people were included in this study.The directed forgetting effects to disease/death-related words were compared by using item-method directed forgetting paradigm under the neutral and negative emotional conditions.Results:Under the neutral and negative emotion conditions,higher recognition of to-be-remembered (TBR) than to-be-forgotten (TBF) words in elder group and younger group (Ps ＜ 0.05).The elder group showed the weaker effect of directed forgetting than younger group[neutral emotion:(0.4 ± 0.1) vs.(0.6 ± 0.1);negative emotion:(0.3 ± 0.1) vs.(0.7 ± 0.04),Ps ＜ 0.05].Under the negative emotion condition,elder people showed stronger effect of directed forgetting for sensitive words compared with control words[(0.4 ± 0.1) vs.(0.1 ± 0.1),P ＜ 0.001].Conclusion:The results suggest that the directed forgetting ability of elderly is weaker than that of young individuals,the ability of non-sensitive words than that with sensitive words,and the directed forgetting effect of elder people tonon-sensitive words in negative emotion is weaker than that in neutral emotion.

Objective To deduce the interactions between genes from time series microarray data.Methods We used inter-transaction association rules mining technique and GO (Gene Ontology) annotation to analyze the microarray data. Results Using 2-fold-change method, 119 differential expression genes were identified from total 10 080 genes or ESTs, whose expression levels varied significantly on 6 periods of fetus cerebellar development. As a result, about 1 300 inter-transaction association rules were extracted and 10 top rules were kept for their maximum J-measure values. A genes association network graph was deduced based on the 10 top rules. Conclusion Inter-transaction association rules are able to deduce the interactions between genes from time series microarray data and the gene expression status can be predicted based on the association rules.

Objective To compare the outcomes between radiofrequency modification and augmentation reconstruction in the treatment for partial anterior cruciate ligament( ACL) tears. Methods Fifty?four patients with partial ACL tears were divided into radiofrequency modification group( n=16) and augmentation reconstruc?tion group( n=38) according to different treatment methods. Minimum follow?up was 2 years. Objective:To compare the antihypertensive effects of tuina at Taichong (LR 3),Neiguan (PC 6) and Qiaogong (Extra) to screen the best tuina protocol for primary hypertension due to liver-fire flaming-up.Methods:A total of 102 patients with primary hypertension due to liver-fire flaming-up were randomly divided into a Taichong (LR 3) group,a Neiguan (PC 6) group and a Qiaogong (Extra) group according to the random number table,with 34 cases in each group.Patients in the three groups received 4-week tuina treatment with the corresponding acupoint respectively.The blood pressure was recorded by benchtop mercury sphygmomanometer before tuina,immediately after tuina treatment,30 min and 60 min after tuina treatment with the patient in a supine position in a quiet treatment room.The total effective rate was observed.Results:The systolic and diastolic blood pressures of the three groups all decreased after treatment (all P＜0.05).The decrease of systolic and diastolic blood pressure at different time points after treatment in the Qiaogong (Extra) group and the Taichong (LR 3) group were better than those in the Neiguan (PC 6) group (all P＜0.05).Qiaogong (Extra) group had the highest total effective rate though there was no statistical difference in the total effective rate among the three groups (P＞0.05).Conclusion:Treating primary hypertension due to liver-fire flaming-up with tuina at Taichong (LR 3),Neiguan (PC 6) and Qiaogong (Extra) can reduce systolic and diastolic blood pressure respectively,among which Qiaogong (Extra) has the highest total effective rate.

BACKGROUND: Long-term utilization of steroid hormone easily induces avascular necrosis of the femoral head (ANFH). Cornu Cervi Pantotrichum possesses bone growth factor and can promote wound healing and tissue repairing, which may has effect on treatment of ANFH. OBJECTIVE: To investigate therapeutic effect of antler powder on ANFH induced by corticosteroids in rats. METHODS: Totally 42 male wistar rats were randomly divided into the blank control, model and high-, moderate- and low-dose treatment groups. Exception those in the blank control group, all rats were intragluteally injected with dexamethasone at 30 mg/kg twice per week for 6 weeks to induce ANFH. Then the rats were treated with antler powder by oral gavage at 200, 400 and 800 mg/kg dosage, once per day for 60 days. Same volume of physiological saline was gavaged in the blank control and model groups. Serum lipid level and bone mineral density (BMD) of femur were measured, pathological change and expression of vascular endothelial growth factor (VEGF) were investigated. RESULTS AND CONCLUSION: Steroid hormone intervention resulted in obviously ANFH in rats. After treatment with antler powder, the degree of necrosis was significant reduced. Compared with the model group, BMD were increased, total cholesterol, triacylglycerol, and low density lipoprotein levels were decreased, and the VEGF expression increased in the treatment groups. The results suggest that antler powder has a positive curative effect on ANFH by promoting bone formation, fat metabolism and increasing VEGF expression.

Objective To explore the effect of isofluranee on receptor for advanced glycation end products (RAGE) expression of hippocampus and learning and memory function in rats. Methods 24-month Sprague-Dawley male rats (n=45) and 4-month Sprague-Dawley male rats (n=45) were randomly divided into six groups with 15 rats each group. Group C1 (aged control group),group C2(adult control group) breath 30% oxygen and air mixed gas; Group S1(single inhalation of isoflurane aged group),Group S2(single inhalation of isoflurane adult group)were anesthetized with 1.5% isoflurane,breath 30% oxygen and air mixed gas for 2h;Group R1(Repeated inhalation of isoflurane aged group), group R2(Repeated inhalation of isoflurane adult group) were anesthetized with 1.5% isoflurane,breath 30% oxygen and air mixed gas 2h a day for three days. Eight rats randomly selected from each group were killed and their hippocampus were immediately isolated for detection of RAGE expression by immunohistochemistry and RT-PCR after accomplished treatment 24h. The remained rats' learning and memory function were assessed using Morris water-maze test. Results The results of Morris water-maze test showed that the times of acerossing the original platform and the time consumption of staying the original platform quadrant was shorter in group S1 and group R1,but the escape latency was longer than group C1(escape latency C1 (9.42± 2.63)s,S1(13.20±3.85)s,R1(17.20±3.44)s, F=12.773, P<0.05). In the group R2,the escape latency was longer but the times of accrossing the original platform and the time consumption of staying the original plat-form quadrant was shorter than group C2 (times of accrossing the original platform C1(7.30±2.40), S1(3.90± 2.42),R1(3.44±2.40), F=7.448, P<0.01).To contrast with the group C2,there were no significant differ-enees in spatial probe test in the group S2(P>0.05). The levels of mRNA and protein of RAGE in hippocampus was significantly higher in group S1 and group R1 than group C1(RAGE mRNA expression C1(0.11±0.02),S1 (0.56±0.09), R1(0.73±0.14), F=179.447, P<0.01). To contrast with the group C2, there were no differ-ences found in the levels of mRNA and protein of RAGE in group S1(P>0.05), but it was higher in the group R2 (RAGE mRNA express C2(0.22±0.04), R2 (0.41±0.08), F=40. 209, P < 0. 01). Conclusion Isoflurane can reduce learning and memory function in both aged and adult rats, but aged rats are particularly significant im-pacted. This effect may be induced by the increase of RAGE expression in hippocampas.
