Objective To investigate the expression of DNA damage and repair gene Apurinic/apyrimidinic endonuclease (APE1) protein in nasal NK/T cell lymphoma, and elucidate its clinical implication. Methods Expression of APE1 proteins was detected immunohistochemically in 10 normal lymph nodes and human nasal NK/T cells from lymphoma of 64 patients and their integral optical density was determined by means of image analytic system. The proliferation index and apoptosis index were determined by means of immunohistochemical staining and terminal dUTP nickend labeling (TUNEL) technique. Results 1. Nuclear, nucleus/cytoplasmic and cytoplasmic types of APE1 positive staining could be noted in nasal NK/T cell lymphoma. Expression of APE1 gene in nucleus was significantly strengthened compared to that in nucleus/cytoplasm and cytoplasm. OBJECTIVE:To establish a method for simultaneous determination of the effective constituents rhein,emodin and chrysophanol in jiangzhiyin by HPLC.METHODS:SHIMADU VP-ODS column was used as analytical column,methanol-0.2%phosphoric acid(80∶20)as mobile phase,the flow rate was set at1ml/min,the detection wavelength at225nm,column temperature at room temperature.RESULTS:The linear ranges of detected rhein,emodin and chrysophanol in jiangzhiyin were0.015～0.15?g(r=0.9996),0.014～0.14?g(r=0.9998)and0.0085～0.085?g(r=0.9993)respectively;The average recoveries were99.26%(RSD=2.00%),98.72%(RSD=1.80%)and99.76%(RSD=2.3%)respectively.CONCLUSION:The determination results are stable and accurate,and the method is feasible and reliable,which can be used for the quality control of jiangzhiyin.

Objective To evaluate the effects of propofol on apoptosis in the hippocampal neurons of fetal rats in vitro.Methods The isolated hippocampal neurons were seeded into 96-well plates or 24-well plates at a density of 5 × 104 cells/ml.The cells were randomly divided into 5 groups (n =18 each) using a random number table:control group (group C),in tralipid group (group Ⅰ) and propofol 1,10,100 μmol/L groups (P1-3 groups).In group Ⅰ,10％ intralipid was added to the culture media until the final concentration reached 100 μmol/L.In groups P1-3,propofol was added to the culture media until the final concentration reached 1,10 and 100 μmol/L,respectively,and the cells were then incubated for 3 h.The cell apoptosis was assessed by flow cytometry.The expression of Bcl-2 mRNA and caspase-3 mRNA was detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR).The expression of Bcl-2 and actived-caspase-3 protein was determined by Western blot analysis.The apoptosis rate was calculated.Results Compared with group C,the apoptosis rate was significantly increased,the expression of Bcl-2 mRNA and protein was down-regulated,and the expression of caspase-3 mRNA and actived-caspase-3 protein was up-regulated in P1-3 groups (P ＜ 0.05).There was no significant difference in the parameters mentioned above between group Ⅰ and group C (P ＞ 0.05).Conclusion Propofol induces apoptosis in isolated hippocampal neurons by inhibiting Bcl-2 expression and enhancing caspase-3 activity in fetal rats.

Objective To improve the sleep quality of patients by evaluating the effect of relaxation training in combination with listening to music on sleep quality of patients with intracranial aneurysm who underwent endovascular interventional therapy. Methods 55 patients who reached the entrance standard were divided into the observation group (28 cases)and the control group(27 cases).The control group received routine nursing measures. The observation group were given relaxation training in combination with listening to music based on routine nursing measures. The sleep quality was appraised by Pittsburgh Sleep Quality Index (PSQI) and patients subjective feeling was investigated by self-designed questionnaires. Results The relaxation training in combination with listening to music ameliorated the sleep quality in the observation group compared with that of the control group(P＜0.01).Conclusion The relaxation training in combination with listening to music could improved the sleep quality of patients with intracranial aneurysm who underwent endovascular interventional therapy.

Objective To evaluate the effect of relaxation training combined with listening to music on controlling the pain of patient with intracranial aneurysm before interventional therapy.Methods To determin the pain degree and subjective reception of the patients on admission and after relaxation training combined with listening to music.Results Pain degree relieved obviously after relaxation training combined with listening to music[(7.31±2.24)vs(3.76±2.12),P＜0.01];self-made questionnaire showed the positive evaluation of relaxation training combined with listening to music was 95%.Conclusion Relaxation training combined with listening to music can relieve the pain and general malaise bacause of absolute bedrest before interventional therapy.

Objective To summarize the clinical experience of laparoseopie chdeeystectomy(LC).Methods Clinical records of 276 cases treated by LC were retrospectively reviewed.Results LC Was successfully accomplished in 265 cases and conversion to open surgery was required in 11 eases;The success rate of operation was 96%;and conversion rate of opening surgery was 4%;In the study,no severe complications occurred.Conclusion selecting fitable inpatients;With perfect technique and highly responsibility,LC could be performed safely.

For investigation on the lactose absorption and tolerance in milk and yogurt, 30 healthy adults were tested by a non-invasive and sensitive hydrogen breath test after drinking half a pound of each daily separately. The results showed that 4 persons absorbed the lactose in the milk completely (13.3%), but 26 showed malabsorption (86.7%), among them, 1 case was slight malabsorption (3.3%), 16 moderate (53.3%) and 9 severe in degree (30%). The intolerance rate and intolerance index of lactose in milk were 53.3% and 0.9; but 6.7% and 0.067 in yogurt respectively.The milk lactose intolerant person expired more hydrogen as 60.34?30.57 ml, while drinking yogurt the hydrogen dropped to 14.97 ?33.64ml (P

Objective To investigate the cooperative antifungal effect of antibacterial peptide MUC7 combined with Bifidobacte‐ria in vitro .Methods The antifungal effect was observed and measured by viable count and the Oxford cup method .Results Two methods exhibited more potent antifungal effect on Candida albicans ,Candida albicans ,Candida krusei and Candida parapsilosis in MUC7 combined with Bifidobacteria group .The colonies′numbers in MUC7 combined with Bifidobacteria group were 2 .00 ± 1 .13 , 2 .00 ± 1 .42 ,5 .00 ± 2 .03 ,2 .00 ± 1 .39 respectively by viable counting ,which was lower than thoes in the saline group and Bifidobacterium group (P<0 .01) ,these two groups were significant lower than those in MUC7 group (P<0 .05);the inhibition zone in MUC7 combined with Bifidobacteria group were (29 .00 ± 2 .17) ,(31 .00 ± 3 .25) ,(29 .00 ± 2 .89) ,(30 .00 ± 3 .36)mm de‐tected by the Oxford cup method ,which showed a significantly difference with the saline group ,Bifidobacterium group and MUC7 group(P< 0 .05) .Conclusion Antibacterial peptide MUC7 combined with Bifidobacterium exhibits good antifungal effect which may provide a foundation for the further research on a new generation of antifungal Bifidobacterium preparation .

The underlying effect of different concentrations of neogenin on proliferation, apoptosis and the related proliferative factors in human trophoblasts was explored in order to understand the function of neogenin during placentation. TEV-1 cell line was cultured and the expression of netrin-1 was detected by using indirect cellular immunofluorescence. Exponentially growing TEV-1 cells were treated by different concentrations of neogenin (0, 1, 5, 10, 50 ng/mL) for 24 h. Cell viability was measured by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay. TEV-1 cell apoptosis was assessed by flow cytometry (FCM). The expression of netrin-1 mRNA and protein in TEV-1 cells was examined by using real-time PCR and Western blot, respectively. It was found that immunoreactivity for netrin-1 was observed in cytoplasm of the trophoblasts. Immediately after treatment with different concentrations of neogenin for 24 h, the netrin-1 expression began to increase. Real-time PCR revealed that the expression level of netrin-1 mRNA was 37.59+/-10.25 times higher than control group when TEV-1 cells were exposed to 50 ng/mL neogenin (P<0.01), and the same tendency was seen by using Western blot. MTT results showed that proliferation of TEV-1 cells was independent of neogenin. Meanwhile, apoptosis was significantly increased to (22.15+/-6.15)% at 50 ng/mL neogenin and (6.55+/-0.25)% without neogenin (P<0.01). It is suggested that neogenin regulates proliferation and apoptosis of TEV-1 cells. And it can enhance the ability of TEV-1 cells to express netrin-1 in a dose-dependent manner. Neogenin may play an important biological role in the normal human pregnancy and contribute to the physiological pregnancy process.
