Eukaryotic DNA element called Matrix Attachment Regions (MARs) can function on regulating the structure and activity of chromosome. Traditional quantitation in vitro and indirect functional analysis can not always reflect MAR-involved physiological state. In order to study transcription regulation and make a try in methodism,? 1-antitrypsin MAR (?1-AT MAR) is cloned and incorporated into pEGFP-C1 vector. Non-MAR-containing and MAR-containing plasmids were then transfected into HEK-293 cells with LipofectamineTM 2000 respectively. Positive cell clones were assayed after 20 days of selection by G418. Semiquantitative RT-PCR and fluorescence microscope analysis show that this MAR has a positive effect on modulating nearby gene expression. Further, co-localization with newly CMV promoter and RNA polymeraseⅡ(RNAPⅡ) was detected by chromatin immunoprecipitation (ChIP), The PCR result demonstrates that more RNAPⅡwas recruited to the CMV promoter to initiate transcription in presence of MAR. ChIP can be used to confirm the MAR-mediated transcriptional activation and provide more reliable information than RT-PCR in real time. The technology is also providing a platform for our research in gene expression regulation.

AIM:To discuss the application effect of adjustable sutures in glaucoma filtering operation after trabecular resection.
METHODS: Seventy-eight cases ( 101 eyes ) suffered from glaucoma were randomly divided into two groups, observation group and control group. Thirty-nine cases ( 51 eyes ) in the observation group underwent trabeculectomy with adjustable sutures, the control group (39 cases, 50 eyes) only adopted trabeculectomy.RESULTS: Compared Preoperative IOP in two groups, the difference was not statistically significant (P>0. 05). After 6mo, IOP were decreased compared with preoperative in two groups, and that in observation group was lower than control group, the difference was statistically significant ( P < 0. 05 ). Six months after operation, there were 1 eye with shallow anterior chamber Ⅰ and 2 eyes with non- functional bleb in observation group, and the complication rate was 5. 9%. While there were of 6 eyes with shallow anterior chamber, in which 4 eyes at gradeⅠ, each one at gradeII and Ⅲ. Non-functional blebs in 5 eyes and a scleral flap adhesion complications rate in the control group was 24. 0%, significantly higher than that in the observation group, the difference was statistically significant ( P<0.01).
CONCLUSION: The adjustable sutures combined with trabeculectomy for glaucoma can significantly reduce the postoperative complications. The curative effect is exact and clinically applicable.

To investigate the relationship between diabetic macular edema (DME) and serum E-selectin level.
●METHODS: Totally 90 patients with diabetic retinopathy in our hospital from October 2009 to October 2013 were observed. Retrospective analysis of the correlation between macular edema degree and serum soluble E -selectin ( sE - sel ). Ninety patients with diabetic retinopathy were divided into two groups: DME group and non - DME group. The level of sE - sel and microalbuminuria (MA) of two groups were determined and compared.
●RESULTS: There was significant difference in MA, sE-sel levels of groups ( P < 0. 05). The value of diabetic retinopathy patients with sE - sel and MA, FBG were positively correlated ( r = 0. 728, P < 0. 05; r = 0. 651, P <0. 05).
●CONCLUSlON: Elevated levels of sE-sel is a risk factor for diabetic macular edema, and sE-sel was involved in the pathogenesis of diabetic microangiopathy.

Glyoxalase system,a most effective defence system is present in cytosol of cells to scavenge ?-oxoaldehydes,which are reactive intermediates of AGEs formation.Glyoxalase Ⅰ is the key enzyme of this system.?-oxoaldehydes and AGEs are involved in the development of Alzheimer′s disease (AD). Therefore,the expression level and activity of glyoxalase I are essential to pathogenesis of AD.

Objective: To investigate the effect of all trans retinoid acid (ATRA) on bone cell proliferation and differentiation. Methods: Newborn rat calvarial osteoblastic cells were isolated and the metabolism of the osteoblastic cells were determined by MTT, Goldens method and immunocytohistologic method. Results: 10 -5 , 10 -6 , 10 -7 , 10 -8 , 10 -9 mol/L ATRA could increase osteoblastic cells proliferation after 72 h culture; 10 -5 , 10 -6 , 10 -7 mol/L ATRA could increase ALP activity. The expression of cyclin D 1 was decreased. Conclusion: ATRA stimulates the proliferation and differentiation of rat calvarial osteoblastic cells in vitro. Cell cycle relative proteins may play an important role in control of cell proliferation and differentiation induced by retinoic acid and derivatives.

S1P (sphingosine 1-phosphate) is one of the metabolite products of membrane sphingomyelin, and it plays an important role in formation, transformation and progression of many malignant tumors as well as the regulation of cell proliferation, apoptosis and angiogenesis. Furthermore, S1P plays different roles in regulation of biological behaviors of malignant tumors depending on the types of the cells and S1PR (sphingosine 1-phosphate receptor). For ovarian cancer and colon cancer, S1P improves the growth of tumors cells by S1PR1/S1PR3, but for malignant melanoma, it inhibits the cell growth by S1PR2. There is a difference in the downstream pathways mediated by difficult receptors. This review summarizes the advances in research on the roles of S1P in regulating biological behaviors of tumor cells and different signal pathways of S1P mediated by various receptors. AIM: To observe the effect of cannabinoid receptor (CB1R) agonist WIN55-212-2 on the expression of brain-derived neurotrophic factor (BDNF), c-Fos and protein kinase A beta-catalytic subunit (PKAC-β) in cerebrum cortex after intracerebral hemorrhage (ICH) in rats. METHODS: The intracerebral hemorrhage model of rat was made by the injection of collagenase Ⅶ, and WIN55-212-2 was intraperitoneally (ip) injected 30 min later. The rats were killed for sampling the brain tissues as specimens 24 h after ICH. The methods of immunohistochemical analysis and Western blotting were adopted to detect the expression of PKAC-β and BDNF. The mRNA expression of PKAC-β, c-Fos and BDNF was determined by RT-PCR. RESULTS: WIN55-212-2 obviously improved some nervous deficit symptoms and increased the expression of BDNF at mRNA and protein levels with upregulating the mRNA expression of c-Fos and downregulating the expression of PKA at mRNA and protein levels in the ipsilateral cerebral cortex. The proteins of PKAC-β, c-Fos and BDNF were expressed on the membrane or nucleus of the neuron or in the cytoplasm of glial cells, respectively. CONCLUSION: The expression of BDNF is induced not only by upregulation of c-Fos, but also by downregulation of PKA in WIN55-212-2 treated rats.
