1.Effects of fluvastatin and Shexiang baoxin pill on expression of 1 lectinlike oxidized low density lipoprotein receptor-1 in a rabbit model of atherosclerosis

Objective:To explore the expression of LOX-1(lectin like oxidizeol low density lipoprotein receptor-1,LOX-1) in aorta of atherosclerotic rabbits and modulating the effect of fluvastatin and Shexiang baoxin pill on the gene and protein expression of LOX-1. Methods:Tirty-two New Zealand white rabbits were randomly divided into four groups: hypercholesterol diet group (fed mth high cholesterol diet),hypercholesterol diet plus Fhevastatin group(fed with high cholesterol diet plus Fhe vastain 25mgl kgld).hypercholesterol diet plus shexiang Bao xin Pill group (fed with high cholesterol diet plus shegxiang Baoxinpill 25mgl kgld).normal diet gwup(fed mith regnlar chow),the protein expression of LOX-1 in aorta of rabbits were examined by immunohistochemsthy,the gene expresswin of LOX-1 in aorte of rabbits were examined by R7-PCK. Results:hypercholesterol diet group(fed with high cholesterol diet),hypercholesterol diet plus fluvastatin group(fed with high cholesterol diet plus Fluvastatin 25mg/kg?d), hypercholesterol diet plus Shexiang baoxin pill group(fed with high cholesterol diet plus Shexiang Baoxin Pill 25mg/kg?d),normal diet group(fed with regular chow),the protein expression of LOX-1 in aorta of rabbits were examined by immunohistochemistry,the gene expression of LOX-1 in aorta of rabbits were examined by RT-PCR. 2.Preparation of IL-2 conjugated immunoliposomes and study on its stability

Objective:To search for a simple and rapid method of preparating the IL-2 conjugated immunoliposomes with high conjugate rate.Methods:The IL-2 conjugated immunoliposomes were prepared by Glutaraldhyde,Undecanal or Derivatization.All methods were analyzed and compared.The conjugate rate of IL-2 was determined by Bradford Assay.Centrifugal acceleration experiment served to prove the liposome stability.Results:IL-2 conjugated liposome by shiff’s base through Undecanal didn’t cause IL-2-self or liposme-self inter-cross link,and conjugate rate was high and stability was good.Conclution:Undecanal is suggested to prepare the IL-2 conjugated immunoliposomes for the anaphase cell experiment.
3.Design and application of self-made water pillow

Objective To explore the effect of self-made water pillow on absolute bed rest patients.Methods Eight-eight patients were randomly allocated into observation group(n=46)and control group(n=42).The patients in the control group received daily care with conventional cotton pillows which were placed at the compression part, while those in the observation group were treated with newly designed water pillows.Both groups received constant turn-over to avoid pressure sores.The two groups were compared in terms of times of daily turn-over,degree of comfort and rate of skin pressuring red.Results The time of daily turn-over in the observation group was less than that of the control group and the degree of comfort was higher and the rate of skin pressuring red smaller all with statistically significant differences(P<0.05).Conclusion The application of the self-made water pillow can effectively prevent pressure sore,enhance comfort degree of absolute bed rest patients and reduce the workload of nursery.It is simple, economical,convenient and safe.
4.Experimental study of Renshen-heche Tablets on tonifying and warming Shen-Yang

Objective: To study the effect of Resen heche tablets (RSHC) on tonyfing and warming Shen Yang. Methods: The mice model of Shen Yang deficiency was established by injecting hydrocortisone. The body weight, autonomic activity and swimming time in ice water in model mice were observed. The level of superoxide dismutase (SOD) in red cell and lipid peroxide (LPO) in plasma of aged rats were analyzed. The effect on anti stress in mice was examined. Results: RSHC had the remarkable effect to improve body weight and autonomic activity and to prolong swimming time in ice water in model mice, and to increase SOD activity and reduce LPO in blood of aged rats, and to elevate the ability of anti fatigue and anoxic tolerance in mice. Conclusion: RSHC drug has the effect on tonifying and warming Shen Yang; can promote growth, enhance organism activity, elevate the adaptation to circumstances and protect injury from free radicals.
5.EFFECTS OF CPT-cAMP ON THE REGENERATION OF THE RETINAL GANGLION CELLS IN ADULT HAMSTERS

Objective To investigate the effects of transplantation of sciatic nerve(SN) and injection of 8-(4-Chlorophenylthio)-cAMP(CPT-cAMP) into the viteous on axotomized retinal ganglion cells(RGCs) in adult hamsters. Methods Twenty adult golden hamsters were ramdomly divided into 4 groups.Optic nerve(ON) was transected and a segment of autologous sciatic nerve(attached graft,AG) was removed and sutured to the proximal stump of ON in regenerating control group(AG group).The animals in experimental groups were further treated with CPT-cAMP injection and/or implantation of a short of segment sciatic nerve(SN) intravitrously.Fluorescent retrograde labeling method and quantitative anatomical techniques were used to measure the number of RGCs. Results 1.In control (AG) retinas,the mean number of regenerating RGCs was 1*!428?284/retina.2.In AG+SN group,the mean number of regenerating RGCs was 2*!220?415/retina.3.In AG+cAMP group,the mean number of regenerating RGCs was 2*!175?364/retina.4.The mean number of regenerating RGCs in AG+cAMP+SN group were 4*!255?820/retina.AG+SN,AG+cAMP groups were significantly different from AG group(P
6.THE MECHANISMS OF IBMX AND CPT-cAMP ON THE REGENERATION OF RGCs

Objective Use three inhibitors to block three different signaling pathway to explore the mechanisms of 3-isobutyl-1-methylxanthine(IBMX) and 8 (4 Chlorophenylthio) cAMP(CPT-cAMP) on the regeneration of retinal ganglion cells. Method Fluorescent retrogratde tracing method and quantitative anatomical techniques were used to measure the numbers of RGCs in control one,control two group and experiment groups. Results 1^ In control group 1(AG),the mean number of regenerating RGC was 1428??284/retina in 4 weeks after surgery; 2^ In control group 2(AG+IBMX+CPT-cAMP) the mean number of regenerating RGC was 4?917?1 489/retina in 4 weeks after surger; 3^ The mean number of regenerating RGC of experiment groups in 4 weeks after surgery were (1) H89 group(AG+IBMX+cAMP+H89):1?426?320/retina; (2)Wortmannin group(AG+IBMX+cAMP+Wortmannin):4143?1?057/retina; (3)PD98059 group(AG+IBMX+cAMP+PD98059):4?154?965/retina. Conclusion The promoting effects of IBMX and CPT-cAMP on regeneration of RGCs can be interrupted by H89 and can't be blocked by wortmannin and PD98059.
7.Determination of ginsenoside in Yixin Capsule by HPLC

AIM: To establish an analytical method of determining ginsenoside in Yixin Capsule by HPLC. METHODS: Ginsenoside Rg_1 and ginsenoside Re in the capsule were separated by HPLC on C_(18) column(250 mm?4.6 mm) with a CH_3CN-0.05 mol/LH_3PO_4(22∶78) as a mobile phase and detected at 203 nm.The flow rate was 1.0 mL?min~(-1). RESULTS: There was a good linear relationship in a range of 1.066-6.396 ?g for ginsenoside Rg_1 and 0.904-5.424 ?g for ginsenoside Re.The average recoveries were 97.1% and 97.3% respectively. CONCLUSION: The method is accurate,rapid and simple for quantitative analysis of Panax Ginseng in Yixin Capsule.
8.CHANGES OF GABA AND GLUTAMATE IMMUNOREACTION IN BRAIN OF RATS WITH EPILEPSY INDUCED BY IL-? OR IL-6

Objective The changes of Glutamic acid(Glu) and GABA immunoreaction in the cerebral cortex and hippocampus of rats with seizure induced by IL-1? and IL-6 were studied.To explore the mechanism of IL-1? and IL-6 in epilepsy. Methods Experimental rats were randomly divided into 3 groups:control group,IL-1? group,and IL-6 group.After intracerebroventricular injection of relevant reagents for 120?min,behaviour changes were observed,Glu and GABA were examined by means of immunohistochemistry in the cerebral cortex and hippocampus of rats. Results The behaviour observation indicated that no seizure happened in control group,seizure in middle degree was observed in IL-1? group and IL-6 group.Compared with in control group,the immunoreaction of Glu showed that the expression was significantly increased in IL-1? group and IL-6 bgroup,while GABA was obviously decreased after intracerebroventricular injection IL-1? or IL-6 at 120?min.Conclusion The machanism of that IL-1? or IL-6 particpated in promotion and abduction epilepsy may be through increasing Glu and decreasing GABA in the cerebral cortex and hippocampus.The nerve excitation is enhanced and then epilepsy occurred.
9.Relationship between the age of menarche,menopause and other factors and postmenopause osteoporosis

Objective To evaluate the relationship between osteoporosis(OP) and the age of menarche and menopause,parity and lactation and bone mineral density(BMD).Methods BMD was measured in 1472 postmenopausal women by Norland XR-36 dual energy Xray absorptiometry(DXA) from May 1999 to April 2003 in our hospital.All data of BMD were compared and statistically analyzed between women with different age of menarche and menopause,parity and lactation period.The diagnostic criteria of osteoporosis was defined by a loss of BMD greater than 2.5 standard deviations below average peak BMD of young adult women.Results In all 1472 subjects,the incidence of osteoporsis is 58.5%(861/1472).There was significant increase in osteoporosis of lumbar spine in women with the age of menarche≥17(119 out of 336 cases were with osteoporosis in lumbar)compared with women with age of menarche≤13(75 out of 276 cases were with osteoporosis in lumbar).In women with menarche age of 11-13,14-16,17-19,lumbar vertebral(L_(2-4)) BMD values were(0.83?0.16),(0.82?0.16),(0.80?0.14) g/cm~2,respectively.There was significant increase in BMD of lumbar in the menarche age group of 11-13 compared with the age group of 17-19 in women within menopause 1-10 years(P
10.Determination of the Content and Dissolution of Hymecromone in Compound Dantong Capsules by HPLC

Objective To establish a method for the determination of hymecromone content and dissolution in Compound Dantong Capsules by HPLC.Methods The content and dissolution of hymecromone was determined by HPLC.The chromatographic conditions were: CAPCELL PAK C18 Column(5 ? m,4.6 mm? 150 mm),methanol-0.01 mol/L phosphate buffer solution(465 ∶ 535) as mobile phase,detection wavelength at 365 nm and the injected volume being 10? L.Dissolution test was performed with 150 mL of water as dissolving medium and with a rotation speed of 100 r? min-1.Results A good linearity of hymecromone was in the range of 0.25 ? g/mL~ 1.75 ? g/mL(r=0.9989),and the average recovery was 99.4 %,RSD=1.60 %(n=5).The dissolutions of 3 batches of samples in 45 minutes were higher than 70 % of the labeled amount,which accorded with the standard.Conclusion This method was simple and accurate,has no interference of other components,and can be used for the quality control and evaluation of Compound Dantong Capsules.
