1.Effects of EGF on Isolation and Cloning of Mouse Embryonic Stem-like Cells from PGCs

Objective To investigate the effects of the EGF on isolation and cloning of mouse embryonic stem-like cells from PGCs. Methods EGF with different density were added to the culture medium of cells .After a culturing period ,the cloning number of mouse embryonic stem-like cells and the topmost passage would be observed and registered. Results There was not obvious effects on the primary culture of mouse ESC and was distinct on the later passage if EGF were used. Conclusions In the primary passage of mouse ESC may survive without EGF,and the impact of the existence of EGF was better than the one of the inexistence to the cloning of mouse ESC.

Based on amino acid score of food, this article presents an "Amino Acid Score Difference (AASD) " for protein quality evaluation, the minus value of AASD representing the limiting amino acid and its number re- presenting the degree. The minus value in one food can be compensated by the positive value in another when we directly select the rational couple for amino acid complementation in diet. In addition, a calculation formula is introduced for amino acid complementation in two kinds of food or in mixed diet including many foods. It gives an effective means in evaluating the nutritional value of protein in foods and planning an ideal diet by selecting foods on the basis of amino acid complementation

Objective To study the effects of MT egg-milk powder on the contents of lead,zinc,iron and copper in brain and femur of lead poisoning rats. Methods 30 Wistar rats were divided into three groups(n=10): normal control group, lead poisoning group and lead poisoning rats treated with MT egg-milk powder group. After treated with 0.2% lead acetate daily for three weeks, the rats were fed with 500 g?L~ -1 MT egg-milk powder for three weeks. Objective To investigate the expression of heat shock protein 5 (HSPA5) in breast carcinoma and to study its relationship with clinical and pathological factors.Methods HSPA5 from 97 cases of breast carcinoma and 40 cases of adjacent tissues of breast carcinoma were examined by immunohistochemical method.Results The overall positive rate of HSPA5 in 97 cases of breast cancer was 81.4 % (79/97).The positive rates of HSPA5 in cancer tissues and paired normal tissues were 32.5 % (13/40) and 7.5 % (3/40),respectively.The expression rate of HSPA5 in tumor tissues was significantly higher than that in normal tissues (x2 =7.813,P =0.038).The expression of HSPA5 in breast carcinoma was correlated with histologic grade and lymph node metastasis (x2 =22.233,P =0.007; x2 =5.342,P =0.024).Conclusion The occurrence of breast carcinoma and the formation of invasion may be related with overexperssion of HSPA5.
6.Clinical significance of HPV subtype infection and atypical glandular cells on LBP

Objective To research the clinical outcome of atypical glandular cell (AGC)according to various subtypes of HPV infection and histological pathology results.Methods The data of the liquid-bases cytology (LBP),HPV infection and histology in 102 cases of AGC at the gynecology outpatient department of our hospital from January 1 ,2009 to February 28,2014 were collected and performed the analysis on their clinical outcomes.Results Among 67 218 cases of LBP detection,102 cases were AGC with the total incidence rate of 0.15%.In the cases of AGC-NOS,67 cases were normal or benign lesions,11 cases were precancerous lesion and malignant lesions;in the cases of AGC treading to tumor,the benign,precancerous and malignant lesions were in 7,14 and 3 ca-ses respectively.At the same time in the cases of AGC-NOS,HPV infection was in 64 cases,in which 57 cases were high risk infec-tion(type 16,52,45)and 7 cases were low risk infection(type 6,11 ).The single infection,double infection and multiple infection were in 54,6 cases respectively;in the cases of AGC trends to neoplasm,HPV infection was in 19 cases,in which 18 cases were high risk infection(type 52,16,18)and 1 case was low risk infection(type 6),single infection and double infection were in 15 cases and 4 cases respectively.Conclusion AGC may play an important role for the forecast of cervical malignant lesions.The results of differ-ent HPV subtypes infection in AGC related tumors also play a certain role in the prediction of cervical neoplasia.Their combined a-nalysis is the important signal for judging the occurrence of gynecological cervical precancerous lesion and malignant tumor,i.e., AGC combining with the corresponding HPV subtype infection not only can make a judgement for the cervical lesions,but also pro-vides the basis for predicting the high risk existence of gynecological malignant tumor and provides constructive suggestions for Chi-na regional cervical carcinoma vaccine manufacturing and promotion.

Objective: To investigate the antitumor effects of arsenic trioxide (As 2 O 3 ) on solid tumor in vivo , the transplanted hepatic carcinoma of mice were treated with various dosages of As 2 O 3 . Methods: After 50 mice were injected intraperitoneally with 0,2, 4, 6, 8 ?mol/g As 2 O 3 respectively for observation of acute toxicity, 1, 2, 3 ?mol/g were selected for this experiment. Eighty mice were transplanted with hepatic carcinoma cells (1.5?10 6 cells/each side) into both subaxillae. As 2 O 3 in 0, 1, 2 and 3 ?mol/g were injected into right subaxilla respectively once every day for 10 days. At the 30th day after transplanted hepatic carcinoma cell, mice were killed and the size of solid tumors was measured. 8.Cell Division Cycle 2 Protects Neonatal Rats Against Hyperoxia-Induced Bronchopulmonary Dysplasia

Purpose:
Hyperoxia-induced bronchopulmonary dysplasia (BPD) is a lung disease in preterm infants. We aimed to explore the role of cell division cycle 2 (CDC2) on histopathologic changes of lung tissues, as well as the viability, apoptosis, and inflammation of lung cells in rats with hyperoxia-induced BPD.
Materials and Methods:
Hyperoxia-induced BPD in neonatal rats and hyperoxia-induced A549 cells were constructed. The mRNA expression of CDC2 was detected by qRT-PCR. The fibrosis score of lung tissues was evaluated by hematoxylin-eosin staining. The viability and apoptosis of A549 cells were detected by cell counting kit-8 assay and flow cytometry. The protein expressions of bcl-2, bax, and caspase-3 were measured by western blot. The levels of tumor necrosis factor-α (TNF-α), interleukin (IL)-6, and IL-1β in A549 cells were detected by enzyme-linked immunosorbent assay. The pcDNA3.1-CDC2 was injected into rats to determine the role of CDC2 in hyperoxia-induced BPD in vivo.
Results:
The expression of CDC2 was decreased in lung tissues of neonatal rats with hyperoxia-induced BPD and hyperoxia-induced A549 cells. The fibrosis score was increased in the lung tissues of neonatal rats with hyperoxia-induced BPD. Overexpression of CDC2 increased the viability and protein expression of bcl-2; and inhibited the apoptosis, inflammation, and protein expression of bax and caspase-3 in hyperoxia-induced A549 cells. Up-regulation of CDC2 alleviated the histopathologic changes in lung tissues of neonatal rats with hyperoxia-induced BPD.
Conclusion
Overexpression of CDC2 promoted the viability and inhibited the apoptosis and inflammation of hyperoxia-induced cells, and alleviated the histopathologic changes of lung tissues in neonatal rats with hyperoxia-induced BPD.
9.Prediction of preeclampsia in twin-pregnant women

Objective:
To construct a prediction model for preeclampsia (PE) risk in twin-pregnant women, so as to provide the basis for early screening and prevention of PE.
Methods:
A total of 467 twin-pregnant women who underwent prenatal examination and delivered at Huzhou Maternal and Child Health Hospital were selected. Sixty cases with preeclampsia (PE) were included in the case group, and 60 women without PE were included in the control group. General information, blood biochemical indicators and uterine artery resistance index (UtA-RI) were collected. A logistic regression model was used to screen predictive factors and establish a nomogram. The Bootstrap method was performed for the internal validation; the receiver operating characteristic (ROC) curve, calibration curve and decision curve analysis were employed to evaluate the discrimination, calibration and clinical utility of the nomogram, respectively.
Results:
In the case group, there were 47 individuals (78.33%) aged younger than 35 years, 21 individuals (35.00%) with pre-pregnancy body mass index (BMI) of 25 kg/m2 and above, and 33 individuals (55.00%) with in vitro fertilization. In the control group, there were 57 individuals (95.00%) aged younger than 35 years, 8 individuals (13.33%) with pre-pregnancy BMI of 25 kg/m2 and above, and 39 individuals (65.00%) with natural pregnancy. Multivariable logistic regression analysis identified age, pre-pregnancy BMI, method of conception, placental growth factor (PLGF) and UtA-RI as risk prediction factors for PE in twin-pregnant women. The established nomogram had an area under the ROC curve of 0.827 (95%CI: 0.755-0.899), a sensitivity of 0.767, a specificity of 0.733, a good discrimination and calibration, and a relatively high clinical net benefit.
Conclusion
The nomogram established by age, pre-pregnancy BMI, method of conception, PLGF and UtA-RI has a good predictive value for the risk of PE in twin-pregnant women.
10.Observation of clinical effect of apical barrier and perforation repair with mineral trioxide aggregate

Objective:To observe the clinical effect of apical barrier and perforation repair with mineral trioxide aggregate.Methods:Selected 23 samples from out-patients of the department of conservative dentistry of oral disease treatment center of 306 Hospital of PLA.Among them,7 samples were with unshaped root apical,6 samples were with lateral perforation from root canal internal absorption or root fracture,10 samples were iatrogenic lateral perforation.The course of the disease was 0.5-24 months.The images of dental films showed that there was shadow around the root or apical area in all the samples.All the samples received regular root canal treatment.Under root canal microscope,the open sites were sealed with MTA.After the barrier formed,filled the root canal with warm gutta-percha vertical compaction technique.Patients were ordered to re-check on 6 months and 12 months respectively.Results:1 sample dropped out.On 6 months visit,1 sample showed enlarged shadow at the apical area,20 samples showed shrinked shadow,1 sample showed no significant change.On 12 months visit,the shadow vanished in 9 samples;the shadow decreased in 5 samples,and there were 2 samples showed no significant changes.Conclusion:The treatment with MTA on apical barrier and perforation shows acceptable effect in short term observation.The use of microscope helps to enhance the accuracy and leak tightness of MTA filling.
