1.Preparation and Quality Control of Diclofenac Sodium Hollow Suppository

OBJECTIVE: To prepare diclofenac sodium hollow suppository and establish its quality control. METHODS: The hollow type suppository was prepared with mixed fatty glyceride as the base; the content of diclofenac sodium was deter-mined by ultraviolet spectrophotometry. RESULTS: The linear range of diclofenac sodium was 5~25 ?g?mL-1(r=0.999 7),with an average recovery rate of 102.04%(RSD=1.08%). CONCLUSION: The preparation technology of diclofenac sodium hollow suppository is simple and feasible,and the quality of the suppository is stable and controllable.
2.Expression and significance of CD147 and collagen Ⅳ in gastric carcinoma

Objective To study the expression of CD147 and collagen Ⅳ protein in gastric carcinoma.Methods Immunohistochemical assay was used to detect the expression of CD147 and collagen Ⅳ protein in 119 cases of gastric carcinoma, 20 cases of normal gastric tissues, and the clinical data was analyzed.Results In 119 cases of gastric carcinoma, the positive expression of CD147 protein was 83.2 %(99/119), the positive expression of collagen Ⅳ protein was 31.9 %(38/119). The expression of CD147 and collagen Ⅳprotein had relationship with metastasis of lymph nodes,depth of invasion and clinical phases (P <0.05). There was negative significant correlation between the expression of CD147 and collagen Ⅳ protein in gastric carcinoma (P<0.01). Conclusion The expressions of CD147 and collagen Ⅳ protein may be used as one of the marks to study gastric carcinoma.

4.Chemical comparison of different Farfarae Flos by NMR-based metabolomic approaches.

1H NMR-based metabolomic approach combined with multivariate statistical analysis was used to evaluate the quality of 21 Farfarae Flos (FF) samples from different growth regions. Principal component analysis showed that wild and cultivated FF could be separated clearly, suggesting a big chemical difference existed between them. Supervised PLS-DA analysis indicated that the wild samples showed higher levels of secondary metabolites, such as bauer-7-ene-3β, 16α-diol, chlorogenic acid, rutin, 7-(3'-ethylcrotonoyloxy)-1α-(2'-methyl-butyryloxy)-3, 14-dehydro-Z-notonipetranone (EMDNT), tussilagone, β-sitosterol and sitosterone. This is consistent with traditional experience that the quality of wild samples are better than that of cultivated ones. The content of pyrrolizidine alkaloids senkirkine also differed greatly among samples from different habitats. The Pearson correlation analysis showed that senkirkine is positively correlated with 4, 5-O-dicaffeoylquinic acid, 3,5-O-dicaffeoylquinic acid, 3,4-O-dicaffeoylquinic acid, rutin, kampferol analogues, to a statistically significant extent. The correlation between the toxic compounds and the bioactive components in FF should be further studied.
5.Application of arthroscopy in treatment of sports traumatic ankle arthritis

To introduce the methods and experience of arthroscopy treating different types of sports traumatic ankle arthritis.Methods The study involved 25 patents with spots traumatic ankle arthritis treated under ankle arthroscopy from January 2008 to October 2010.American Orthopaedic Foot and Ankle Society (AOFAS) ankle-hindfoot scoring system was applied to make functional evaluation before and after operation.Also,anterior drawer stress radiographs were used to assess the stability of joint.All patients had arthroscopic examination and received corresponding treatments,including synovial scar tissue resection,cartilage surface trimming,and microfracture management.Five patients underwent stage Ⅰ extramalleolus ligament repair.Results Arthroscopy showed simple synovitis in four patients,impingement syndrome developed from synovitis with fibrous scar tissue formation in 10,tibial or talus cartilage lesions in 11 and congestion of fresh injury of fibular end in four.A total of 23 cases were followed up for average 13.5 months (range,6-24 months).The ankle-hindfoot score was increased from preoperative (54.3 ± 6.2) points to (81.5 ± 7.9) points at 1 month,(82.9 ± 2.5) points at 3 months,( 83.1 + 2.1 ) points at 6 months and ( 83.5 ± 3.9 ) points at 12 months,with statistical differences ( t =13.01,20.58,21.10,19.11 respectively,P<0.05).The anterior drawer stress radiographs showed that the anterior translation of talus improved from preoperative ( 15.2 ± 2.5 ) mm to postoperative (3.5 ± 0.2 ) mm,with statistical difference ( t =9.33,P < 0.05 ).The anterior drawer test and talus tilt test were both negative for all patients.Concluslons Ankle arthroscopy is characterized by micro trauma and fast recovery.Symptomatic treatment under arthroscopy can obviously relieve the uncomfortable symptoms following sports traumatic ankle arthritis.
6.The abnormalities of noninvasive urodynamics in early diabetes cystopathy

Objective To study the abnormalities of noninvasive urodynamios in early diabetes eystopathy (DCP) and provide diagnosis evidences in its early stage. Methods According to the disease course (less or more than 1 year), 85 patients with type 2 diabetes mellitus (T2DM) were divided into new lydiagnosed diabetic group and non-newly diagnosed diabetic group. Thirty healthy cases were involved in normal control group. All of them were checked with the technology of noninvasive urodynamics to measure maximal flow rate, average flow rate, the volume leading to first bladder sensation and residual urine volume. Results As to the 32 newly diagnosed diabetic group, maximal flow rate was (18.4±6.9) ml/s, and average flow rate was (10.6 ± 5.3) ml/s, 18 cases were detected to have bladder residual urine, with the average residual urine volume of (13.2 ± 17.3) ml, and the DCP detection rate was 56.2%(18/32). As to the 53 non-newly diagnosed diabetic group, maximal flow rate was (14.7 ± 6.6) ml/s, and average flow rate was (9.5±4.7) ml/s,38 cases were detected to have bladder residual urine, with the average residual urine volume of (19.3 ± 18.4) ml, and the DCP detection rate was 71.7%(38/53). There was no residual urine detected in normal control group. Their maximal flow rate was (25.7 ± 5.9) ml/s, and average flow rate was (18.0 ± 4.9)ml/s. Compared with that in normal control group, maximal flow rate, average flow rote and residual urine volume decreased in both newly diagnosed diabetic group and non-newly diagnosed diabetic group(P< 0.01). As compared with that in newly diagnosed diabetic group, maximal flow rate in non-newly diagnosed diabetic group decreased obviously (P< 0.05). Conclusions The abnormalities of urodynamics may happen in the early stage of diabetes, and maximal flow rate may be as the most sensitive index. With the new technology of noninvasive urodynamies, we DCP can be diagnosed in early stage and evaluated the function of bladder dynamically.
7.Change in corneal biomechanical properties and influence factors in population with myopia

Background Corneal refractive surgery has significant effects on corneal biomechanical properties.However,there are few study on large scale population with corneal biomechanical properties in the myopic population before corneal refractive surgery.Objective This study was to evaluate the change of corneal biomechanical properties and influence factors in myopic eyes using ocular response analyzer (ORA).Methods A prospective cohort study was carried out in Beijing Tongren Eye Center from April 2010 to January 2011.Corneal hysteresis (CH) and corneal resistance factor (CRF) were detected using ORA in 1 792 eyes of 896 myopic subjects who were going to receive corneal refractive surgery under the informed consent.According to different spherical equivalent (SE) the eyes were grouped into-0.25-<-3.0 D group,-3.0-<-6.0 D group,-6.0-<-9.0 D group,-9.0-<-12.0 D group,-12.0-<-15.0 D group and ≥-15.0 D group and according to different central corneal thickness (CCT),the eyes were grouped into <500 μm group,500-<550 μm group,550-<600 μm group and≥600 μm group.The CH value and CRF value in different SE groups or different CCT groups were compared,and the factors influencing CH and CRF were analyzed by Pearson correlation analysis.Results The mean CH and CRF value were (9.84±1.52)mmHg (1 mmHg =0.133 kPa) and (10.46±1.71) mmHg,respectively in the myopic eyes.The CH values were (10.35±1.53),(10.07±1.55),(9.81±1.46),(9.71±1.59),(9.35±1.55) and (9.23±1.28) mmHg in the-0.25-<-3.0 D group,-3.0-<-6.0 D group,-6.0-<-9.0 D group,-9.0-<-12.0 D group,-12.0-<-15.0 D group and ≥-15.0 D group,respectively,showing a significant difference among the groups (F=20.69,P<0.01),and the CH values in the-6.0-<-9.0 D group,-9.0-<-12.0 D group,-12.0-<-15.0 D group and ≥-15.0 D group were significantly lower than those in the-6.0-<-9.0 D group,-9.0-<-12.0 D group (all at P<0.01).There was no significant difference in the CRF values among different SE groups (F =0.65,P =0.49).CH and CRF values were increasing with the raise of CCT,with evidently differences among the CCT<500 μm group,500 μm ≤ CCT<550 μm group,550 μm≤ CCT<600 μm group and CCT≥ 600 μm group (CH:F=110.99,P<0.01;CRF:F =84.35,P<0.01),and the CH and CRF values were significantly higher in the 550 μm ≤ CCT<600 μm group and CCT ≥600 μm group than those in the CCT<500 μm group (all at P<0.01).CH showed a positive correlation with SE (r =0.24,P<0.01),and no significant correlation between CRF and SE (r =0.03,P =0.20).The CH and CRF showed the positive correlations with the CCT (r=0.42,0.57,both at P<0.01).In addition,CH and CRF values increased as the increases of SE,corneal curvature (CC) and CCT values,with the regression equation of CH =-7.87+0.08SE+0.16CC+0.02CCT (R2=0.26,P<0.01) and CRF=-11.42+0.14CC+0.03CCT (R2=0.34,P<0.01),respectively.The CH and CRF did not change with the age and gender in the subjects.Conclusions Corneal biomechanical properties including CH and CRF were positively correlated with CCT,CC and SE.
8.The different effect of negative emotion induced by background music on verbal and spatial working memory: evidence from ERP study

Objective To investigate the different effect of negative emotion induced by background music on verbal and spatial working memory. Methods Fifteen participants were exposed in relaxing or horrible background music and did the working memory task. After each emotional condition, the 9-point Self Assessment Manikin scale was registered. n-back paradigm was employed as WM task and the WM task contained two different types, the verbal WM and spatial WM. In the same time,The ERPs during each emotional condition was recorded,and the anterior LPC and posterior P300 were analyzed. Results The valence,arousal and dominance were 5.40±0.91,3.93 ±0.79 and 5.33 ± 1.29 in relaxing condition,while the same dimensionalities in negative condition were 2.00 ± 0.75,6.47 ± 0.99 and 2.33 ± 0.89. The self reported data showed that horrible background music induced the negative emotional state, which were observed from the decrease of valence (t(14)= 15. 902, P <0.01) and dominance (t(14) =6.708, P<0.01) while the increase of arousal (t (14)=9.906, P<0.01). The ERP data showed,there was a main effect in emotion with anterior LPC (F (1.14)= 7.37, P < 0.01) and in both verbal and spatial WM,the amplitude of LPC were reduced in negative emotional condition. However, there was an interaction of emotion and type on amplitude of P300 (F (1.14)= 11.25, P < 0.01), selective influence of negative emotion in spatial WM were seen (F (1.14)= 12.43, P<0.01). Conclusion The WM was influenced by negative emotion induced by negative background music ,and the spatial WM was more vulnerable to negative emotion.
9.Outcome of Community-based Rehabilitation Care: A 3-year Investigation

Objective To evaluate the effects of community rehabilitation care. Methods A 3-year community-based rehabilitation care services in Desheng Community Health Service Center were investigated with questionnaire. Results Those of the knowledge of rehabilitation awareness, keeping in a psychological well being, communicating with others, participating in rehabilitation and other community activities, independent in household activities, the activities of daily living, all improved significantly during the project. Conclusion Community-based rehabilitation care plays a important role in community-based comprehensive rehabilitation services, which may improve the quality of life of people with disabilities and chronic dysfunction.
10.Effect of basic fibroblast growth factor coacervates on experimental neuropathy in diabetic rats

OBJECTIVE To explore the therapeutic effect of basic fibroblast growth factor (bFGF) coacervates on diabetic peripheral neuropathy (DPN) in diabetic rats.METHODS Poly (ethylene argininylaspartate diglyceride) (PEAD), heparin and bFGF were dissolved in saline at the mass ratio of 50:10:1 to obtain bFGF coacervates. The loading efficiency of bFGF in the coacervates was analyzed by Western blotting. The release profile of bFGF from the coacevates was detected by ELISA. Male SD rats were ip injected with streptozotocin 65 mg · kg- 1 to establish a diabetic model,and DPN occurred 8 weeks later. The DPN rats were randomly divided into free-coacervate group, bFGF group and bFGFcoacervate group. For bFGF group, bFGF 200 μg·kg-1 was im injected once daily for 3 d. In bFGF-coacervate group, bFGF coacervate solution (244 μL) equal to bFGF 200 μg · k - 1, was im given only once. DPN rats in free- coacervate group were im given the same volume of vehicle(PEAD + heparin) only once. Ten normal age peer rats were taken as normal control group.Footprint analysis was conducted each week to evaluate motor function. On the 30th day after treatment,the rats were sacrificed, and sciatic nerves of both sides were harvested for pathological observation through HE staining. Apoptosis in nerve tissue was detected by DAPI staining, and Ki67 and proliferating cell nuclear antigen (PCNA) protein levels were detected by Western blotting. RESULTS Western blotting and ELISA analysis indicated that bFGF-coacervates were well prepared at a mass ratio of 50:10:1,and controlled bFGF release for at least 35 d. The result of rat behavior evaluation and pathological index test indicated that, compared with normal control group, the sciatic function index (SFI) in free-coacervate group decreased significantly(P<0.01), the internal nerve fibers were accompanied by irregularity and serious demyelination, and there was a large number of apoptotic nuclei and low expressions of Ki67 and PCNA proteins (P<0.01).After injection with bFGF or bFGF-coacervates, the SFI increased progressively (P<0.05, P<0.01), and the proportion of fibers with myelin abnormalities and apoptotic cells was significantly reversed. Moreover, the levels of Ki67 and PCNA was evidently enhanced on the 30th day post- operation (P<0.05, P<0.01). Compared with bFGF group, the results of those detection indicators in bFGF-coacervate group were better (P<0.05, P<0.01). CONCLUSION PEAD and heparin complex can load bFGF with high efficiency, and control its release in a steady manner. For DPN rats,treatment with bFGF-coacervates is more effective than bFGF alone.
