China Biotechnology 2006;0(12):-
There is immense interest in the long-chain polyunsaturated fatty acids(LC-PUFAs) on account of their health-beneficial properties. There has been considerable interest in the production of LC-PUFAs in transgenic plants to provide a cheap, sustainable and clean source of these fatty acids.Several main LC-PUFAs,their roles and metabolic pathway in plant are introducedbriefly, progresses on biosynthesis of LC-PUFAs in transgenic plant are reviewed, and strategies for improving the yields of LC-PUFAs in plant are also discussed.
2.Advances in Application Studies of FAD2 on Biosynthesis of Industrial Fatty Acids
China Biotechnology 2006;0(05):-
Many unusual fatty acids present in the seed oils of non-food plants are modified at the ?12 position either by the addition of epoxy or hydroxy groups or by the formation of triple (acetylenic) bonds or conjugated double bonds. The introduction of these functionalities into C18 fatty acids is catalyzed by a family of divergent forms of the fatty-acid ?12-desaturase (FAD2) enzyme. Genes encoding these enzymes have now been cloned from a variety of organisms and expressed in transgenic oilseeds. However, the accumulation of the intended fatty acids in transgenic plants has been relatively disappointing, and the mechanisms of these unusual fatty acids biosynthesis and transferring from phosphatidylcholine into storage triacylglycerols still need to be elucidated.

OBJECTIVETo study the effect of Flos Daturae alkaloids (FDA) on TGF-beta1-1uuuu;U epithelial-mesenchymal transition (EMT) of human pulmonary adenocarcinoma A549 cells.
METHODSA549 cells in vitro cultured were randomly divided into 5 groups, i.e., the blank control group, the TGF-beta1 group, the low dose FDA group, the medium dose FDA group, and the high dose FDA group. The morphologies of A549 cells were observed. Expressions of cytokeratin (CK)-19 and alpha-smooth muscle actin (alpha-SMA) were detected by Western blot and real-time PCR at 24, 48, and 72 h, respectively.
RESULTSA549 cells in the TGF-beta1, group turned from cobblestone to spindle shape gradually. Those in low, medium and high dose FDA groups showed similar shapes to those of the TGF-beta1 group. There was no statistical difference in the morphology of A549 cells among the 3 dose FDA groups (P > 0.05). Western blot showed that, when compared with the blank control group, the expression of CK-19 was down-regulated, but the expression of alpha-SMA was up-regulated in the TGF-beta1 group (P < 0.01). Compared with the TGF-beta1, group, the expression of CK-19 was up-regulated, but the expression of alpha-SMA was suppressed in low, medium and high dose FDA groups (P < 0.01). The CK-19 expression obviously increased, but the alpha-SMA expression was suppressed in high dose FDA group at 72 h (P < 0.01). Real-time PCR results showed, as compared with the TGF-beta1 group, the mRNA expression of CK-19 was increased, but the mRNA expression of alpha-SMA was reduced in low, medium and high dose FDA groups (P < 0.01).
CONCLUSIONSFDA had no effect on EMT morphological changes of TGF-beta1 induced A549 cells. FDA could reverse characteristic markers of A549 cells during EMT to some extent, such as expressions of CK-19 and alpha-SMA. The expression of CK-19 (as the epithelium marker) increased and the expression of alpha-SMA (as the mesenchymal marker) was reduced. Besides, they were most obviously seen in the high dose FDA group at 72 h in a dose- and time-dependent manner.

Chinese Journal of Postgraduates of Medicine 2011;34(35):13-15
Objective To explore the long-term effect of open reduction and internal fixation for the treatment of displaced acetabular fracture, and analyze the influence of risk factors. Methods The clinical data of 62 patients with acetabular fracture from August 2005 to February 2009 was analyzed retrospectively. All the cases were treated with open reduction and internal fixation. The long-term effect and related risk factors were analyzed. Results All the cases were followed up for 24-52(38.8 ± 2.6) months.According to the Matta standard of replacement of fracture, there were 43 cases with anatomic reduction,14 eases with satisfactory reduction,5 cases with unsatisfactory reduction.Fracture union was obtained in all the patients.Based on the modified Merled' Aubigne -Postel clinical grading system, the result was excellent and good in 50 patients, fair and poor in 12 patients, with excellent rate of 80.65% (50/62).Postoperative complications including traumatic arthritis were seen in 7 patients, heterotopic ossification in 3 patients and femoral head avascular necrosis in 1 patient. The related risk factors of clinical results of displaced acetabular fracture were age,Letournel-Judet fracture type, operation time, cartilage surface damage of the femoral head and quality of reduction (P < 0.05 ).However, gender,AO fracture type, surgical approach and hip dislocation were not affecting factors of the acetabular fracture(P > 0.05 ).Conclusions Open reduction and internal fixation can result in a satisfactory clinical outcome.Age,Letournel-Judet fracture type, operation time, cartilage surface damage of the femoral head and quality of reduction are independent risk factors affecting postoperative long-term functional outcomes.

Object To observe the effects of rehmaionoside A on mice leukopenia models induced by cyclophosphamide. Methods Mice leukopenia models were prepared by ip cyclophosphamide and then treated with rehmaionoside A. The levels of body weight, peripheral hemogram, reticulocyte count, spleen index, bone marrow karyocyte count and contents of bone marrow DNA were determined. Results Rehmaionoside A could obviously raise the counts of white blood cell, red blood cell, platelet, reticulocyte and bone marrow karyocyte, contents of bone marrow DNA, and body weight of the leukopenia model. Conclusion Rehmaionoside A has apparent leukogenic and antianemic actions.

AIM: To investigate the clinicopathological features of orbit primary liposarcoma. METHODS: By means of histological, ultrastructural and immunohistochemical studies, the clinical and pathological features of two cases of orbit primary liposarcoma were analyzed and compared with 6 cases of primary retroperitoneum liposarcoma. RESULTS: Two cases of orbit primary liposarcoma were young persons. The majority of cases of primary retroperitoneum liposarcoma were aged over 40 years. The tumors were located at the retrobulbar region, and involved the wall of eyeball and optic nerve. The tumor cells of case 1 were round, with abundant eosinophilic granules in the cytoplasm, and there were lipoblasts in the tumor cells of two cases. Local recurrence following excision occurred in case 2.Immunohistochemical expression of S100 protein was positive in all cases, and CK (AE1/AE3), desmin, MyoD1, melanoma were negative. Ultrastructure observation showed lipid drops in the tumor cells in case 2 of orbit primary liposarcoma. CONCLUSION: Primary liposarcoma of the orbit may recur. The diagnosis was based on histological,ultrastruc-tural observation and immunohistochemical staining.
