Visfatin is a recently discovered adipokine secreted by visceral adipose tissue,corresponds to pre-B cell colony-enhancing factor(PBEF), which is composed of 473 amino acid coding a 52kDa polypeptide and expressed in bone marrow,liver,muscle and so on.Visfatin binds and activates the insulin receptor,exerting insulin-mimetic effects;promotes adipose tissue to differentiate,synthesis and accumulate,which is correlated with obesity;promote vascular smooth muscle cell to mature,participating in the processes of inflammatory response and cell differentiation and apoptosis.The multi-function of visfatin increases new contents of correlated disease pathogenesis and new targets of disease management.
2.The effect of combination of gliclazide and sitagliptin on the treatment of poorly obese obese patients with type 2 diabetes mellitus

Objective To investigate the effect of psychological intervention combined with gliclazide and sitagliptin on the treatment of poorly obese obese patients with type 2 diabetes mellitus. Methods 96 obese patients with type 2 diabetes mellitus who had poor control of blood glucose after insulin treatment were randomly divided into control group and experimental group,48 cases in each groups. Gliclazide and sitagliptin (FBG), glycosylated hemoglobin (HbA1c), 2h postprandial blood glucose (2hPG), fasting insulin (FIns) were measured before and after 15 weeks of treatment in the control group. situation. Results FBG, HbA1c, 2hPG and FIns were significantly lower in the experimental group than in the control group (P<0.05). Conclusion The combination of gliclazide and sitagliptin on the treatment of poorly obese type 2 diabetes mellitus with insulin treatment can significantly reduce the blood glucose level of patients and promote it.
3.Experimental study of TGF-β2 antisense oligodeo- xynucleotide as an anti-scarring agent in glaucoma surgery

AIM: Currently available anti-scarring regimens for glaucoma filtration surgery have potentially blinding complications and safer alternatives would be beneficial. This experiment is to investigate the effect of TGF-β2 antisense oligodeoxynucleotide on differentiation, proliferation of subconjunctival fibroblast following glaucoma filtration surgery.METHODS: Glaucoma filtration surgery were performed on both eyes of 28 rabbits. TGF-β2 antisense oligodeoxynudeotide was subconjunctivally injected in the right eyes (A group), and TGF-β2 missense oligodeoxynucleotide (B group)or PBS(C group) was used at the same method in the left eyes as controls. Rabbits were killed at 4,7,14 and 28 days after surgery. Intraocular pressure (IOP), bleb characteristics were recorded at different time point. Subconjunctival fibroblasts were examined by immunohistochemistry and electron microscopy.RESULTS: The IOP of rabbits in group A was significantly lower at 14 days (6.74± 1.18 mmHg) and 21 days (8.15± 1.97mmHg) after operation than the IOP in group B (8.53± 1.04,9.72± 1.09 mmHg)(P <0.01) and group C(8.79± 1.21, 9.43±1.27 mmHg) (P <0.05). The mean bleb survival time was longer (17.2 days) in group A than that of group B (14.5 days) and group C (13.5 days)(P<0.05). The population of the cells expressing α -smooth muscle actin(α -SMA) and proliferating cell nuclear antigen (PCNA) was significantly reduced in group A compared with the group B and C. The ultrastructure of fibroblast was not altered by TGF-β2 anti-sense oligodeoxynucleotide.CONCLUSION:TGF-β2 antisense oligodeoxynucleotide can prevent the scar formation after glaucoma surgery by inhibit the differentiation and proliferation of subconjunctival fibroblast. It could be a potentially useful anti-scarring alternative for the prevention of late surgical failure.
5.Improvement of osteoblast bioactivity and osteoprotegrin gene expression of titanium surface by anodic oxidation

BACKGROUND:Nanostructure formation on titanium surface by anodic oxidation has good biocompatibility with bone tissue. OBJECTIVE:To observe the surface morphology and crystal ine constitution of nanopores microstructure on titanium surface formed by anodic oxidation and to further observe its influence on the MC3T3-E1 osteoblast cells’ biological behavior and the gene expression of osteoprotegerin. METHODS:Nanopores forming on titanium surface by anodic oxidation was prepared as experimental group and polished titanium as control group (12 samples for each group). Mouse MC3T3-E1 osteoblasts were co-cultured with polished pure titanium plate group and anodic oxidation nanopores group. After 7 days of inoculation, cellmorphology was observed using scanning electron microscopy, MTT method was used for the cellproliferation test and the growth curve was made. Gene expression of osteoprotegerin was also detected. RESULTS AND CONCLUSION:After anodic oxidation, a homogeneous and uniform array of nanopores formed;however it had no significant influence on the crystal ine phase of the titanium sample surfaces. Titanium surface with nanopore structure was more favorable than polished titanium surface for cellattachment and spreading. cells on the anodized surface with nanopores had higher celldensity and bigger metal coverage area. cells on the nanopores surface also exhibited a polygonal shape with many filopodia extending in al directions. MTT method showed that the anodized nanopore surface had higher cellamount than the as-polished titanium, and the former was about 1.4 times of the latter group after 7 days of culture. The gene expression level of osteoprotegerin in the MC3T3-E1 cells cultured on anodized titanium surface with nanopores was significantly higher than that on the as-polished titanium (P<0.01). The anodic oxidation treatment is more advantageous for the osteoblasts adhesion and gene expression of osteoprotegerin, thereby promoting the growth of osteoblasts.

Trans-cinnamaldehyde, the main component of volatile oil from cassia twig or Cinnamomum cassia, which is a traditional Chinese herbal medicine. Trans-cinnamaldehyde is a kind olefine aldehyde of organic compounds and has many pharmacological properties, such as anti-inflammatory, anti-tumor, anti-bacterial, antidiabetic, anti-obesity, and neuroprotection etc. The compound has preventive and therapeutic effects on the nervous system, cardiovascular, cancer, diabetes and other diseases. Trans-cinnamaldehyde, as a preventive care of nature medicine, has great clinical and market potential. This paper gives a review about the pharmacological effects and mechanism of trans-cinnamaldehyde researched in the latest five years.
