OBJECTIVE:To study the active fraction of the seeds of Hovenia Dulcis Thunb.to treat acute alcoholism.METHODES:The seeds of Hovenia Dulcis Thunb.were divided into five parts:superitical fluid extraction(SFE)part,chloroform extraction part,ethyl acetate extraction part,n-butanol extraction part and water extraction part.The effects of the active fraction of the seeds of Hovenia Dulcis Thunb.on ebriety latency,ebriety rate,mortality,and ethanol concentration-time curve of the alcoholism mice were investigated.RESULTS:The ethyl acetate extract could prolong the ebriety latency of the mice,decrease ebriety rate and mortality of the alcoholism mice,and significantly decrease serum ethanol concentrations at 0.5,1,and 1.5h after alcohol administration(P

OBJECTIVE:To determine the content and establish HPCE fingerprints of polysaccharides of the T.jackii seed.METHODS:The content of polysaccharides of the T.jackii seed was determined by phenol-vitriolic colorimetry and the fingerprints of polysaccharides were established by HPCE.RESULTS:The content percentage of polysaccharides in the seed of T.jackii was6.56%,with average recovery rate at98.3%and RSD at2.31(n=5).There were two kinds of polysaccharides in the seed of T.jackii.CONCLUSION:There is no significant difference with regard to the contents and components of polysacc_ harides in the T.jackii seeds from different origins.

Objective To compare the clinical efficacy of breast-conserving surgery and modified radical mastectomy for early breast cancer and their influence on quality of life.Methods 78 patients with early stage breast cancer were divided into two groups by random number table,and 39 cases in each group.The control group was given modified radical mastectomy,while the observation group was given breast-conserving surgery.The clinical efficacy,surgical conditions and quality of life scores were compared between the two groups.Results The operative time,blood loss,incision length and the average hospital stay of the observation group were (60.23 ± 5.21)min,(44.73 ± 4.22) mL,(4.72 ± 1.02) cm and (9.52 ± 0.86) d,which were significantly lower than those of the control group(t =7.88,8.32,8.89,6.42,all P ＜ 0.05).The postoperative physical factors,mental factors,psychological factors and social factors scores of the observation group were (67.44 ± 3.87),(48.64 ± 3.73),(117.34 ± 8.33)and (61.56 ± 5.32),which were significantly higher than those of the control group (t =5.43,5.87,6.29,7.11,all P ＜ 0.05).The postoperative 3-year local recurrence,distant metastasis and survival rate showed no significant differences between the two groups (x2 =1.22,1.09,1.29,all P ＞ 0.05).Conclusion Breast-conserving surgery in the treatment of early breast cancer has significant effect,and it has less trauma,less bleeding and cosmetic results,and it can be used as the preferred method for the treatment of early breast cancer,which is worth to be further applied in clinical.

AlM: To investigate the inhibition effect of bevacizumab on human Tenon capsule fibroblasts ( HTFs ) and discuss the countermeasures of bleb scarringscarring in glaucoma surgery countermeasures.? METHODS: Adopted cell recovery method and followed the aseptic principles, we performed the culture of HTFs which came from the Central Laboratory of Shaanxi People's Hospital cell library. Wound Healing assay:We scraped a cell-free zone on the cell surface when the cells reached confluence at 80%. The control group was added to serum-free DMEM medium. The HTFs of bevacizumab group were stimulated with 1mg/mL concentrations without DEME for 0, 24, 48, and 72h. The scratch width was observed and measured.? RESULTS: HTFs were long fusiform shape under microscope, the nucleus is in the center of the cell with larger nucleus, abundant cytoplasm, were arranged in a whorled growth out of shape, strong ability to proliferate, conform to general forms of fibroblast. The morphological and biological characteristics of cells after cryopreservation and resuscitation remain unchanged. Wound healing assay: 0h, equal to the initial width of the two groups, 24h when the migration distance of the two groups of cells are basically the same, 48h when the control group cell migration distance is greater than that in the bevacizumab processing group, 72h when the control group scratches basichealing, bevacizumab treated cells migrate closer than 48h no significant change, and a lot of cells died.?CONCLUSlON:Cell recovery method can successfully cultured HTFs, which was stability on morphology and biological properties, laying the cellular basis for experimental research. Fibroblast itself has a strong ability to migrate, outside - derived bevacizumab can inhibit HTFs migration evidently and it will cause excessive cell death when. Bevacizumab has certain extent inhibitory effect on HTFs migration, and it is likely to become one of the important drugs for creating bleb scarring after glaucoma surgery in the future.

The primary focal area of carcinoma of unknown primary (CUP) can not be determined by clinical examination.It is difficult to find the primary site and its prognosis is poor.The traditional diagnostic approaches mainly include pathological examination,endoscopy,imaging technology and so on.However,positron emission tomography is the preferred approach.In recent years,gene expression profiling has become a new diagnostic approach to find the source of CUP.The detection rate by gene expression profiling is high,and the approach can accurately identify the primary site.The recommended therapies include classic platinum-or paclitaxel-based chemotherapy,radiotherapy or surgery,and site-directed therapy by gene expression profiling for patients with CUP improves their survival rate.

Objective To investigate induction of apoptosis of human ovarian cancer CoC1 cells by 5-allyl-7-gen-difluoromethylenechrysin(ADFMChR)in vitro,and its molecular mechanism. Methods The proliferative and growth inhibition of CoC1 cells treated with ADFMChR was respectively measured using(3,4,5-dimethylthiazol-2-yl)2,5-diphenyl tetrazolium bromide(MTT)colorimetric assay and clone-formation in soft agar assay. The apoptosis of CoC1 cells induced by ADFMChR was determined by DNA agarose gel electrophoresis assay. Effect of ADFMChR on PPAR-γ, Bcl-2. Bax protein expression level of CoC1 cells was detected by western blotting. Results The proliferation of CoC1 cells could be significantly inhibited by ADFMChR in a dose-dependent manner. The IC50 was 7. 76 μmol/L The ladder-shaped band could be shown in DNA agarose gel electrophoresis after treatment with ADFMChR at 30. 0 μmol/L for 48 h and the ladder-shape band disappeared with GW9662. Western blot analysis showed that expression of PPAR-γ and Bax proteins were up-regulated and protein levels of Bcl-2 were depressed after treatment with ADFMChR in a concentration-dependent fashion. However,the effects of ADFMChR on Bcl-2 and Bax proteins were blocked or diminished in the presence of GW9662. Conclusions The effect of ADFMChR on induction of apoptosis in CoC1 cells may be mediated by activation of PPAR-γ, sequentially accompanied by reducing of protein levels of Bcl-2 and increasing of Bax expression.

Objective To study the treatment effect of self-made huoxue tincture for treating skin injury and phlebitis caused by extravasation of chemotherapeutic drugs.Methods Patients(30 cases)with skin injury and phlebitis caused by extravasation of chemotherapeutic drugs were divided into the observation group(20 cases)and the control group(10 eases).The observation group underwent treatment with huoxue tincture,while the control group adopted local hydropathic compress with 50%magnesium sulfate.Results The effective rate of the observation group was higher than that of the control group,P<0.05.Conclusions Hydropathic compress with huoxue tincture was effective in the treatment of skin injury and phlebitis caused by extravasation of chemotherapeutic drugs.It may be widely applied in clinic.

Objective To insert VEGF-R3(FIT-4)gene fragment into cloning vector and identify it.Methods By T-A cloning,the VEGF-R3 gene fragment amplified by RT-PCR was cloned on the PGEM-T Easy plasmid vector and transfected into E.coli JM109.the recombinant clones were screened by"white-blue plaque selection" and tested by the methods of single restrictional enzyme.Results The VEGF-R3 gene fragment amplified by RT-PCR showed a smear after the electrophoresis on agarose Gel and obtained white clones by"white-blue plaque selection".The electrophoresis result of the recombinant clones tested by the single restrictional enzyme was accordant with expectant result.Conclusion The recombinant clones through the methods T-A may be used in the further gene expression and gene therapy studies.
