1.Removal of Cu,As,and Pb from chrysoidine by supercritical CO_2 extraction

Objective To make an attempt at some conditions on extracting heavy metals in chry-soidine by supercritical CO_2 extration.Methods Taking sodium diethyldithiocarbamate trihydrate(NaDDC?3H_2O) as chelating agent and ethanol as entrainer,the orthogonal test was designed and ICP-MS used to mensurate the contents of Cu,As,and Pb in chrysoidine under different conditions.According to the results,the factors including extracting pressure,temperature,chelating agent dosage,and time were studied.Results When the test sample was 10 g,the optimum condition was that extracting pressure: 25 MPa,the temperature : 60 ℃,chelating agent dosage: 2 g,extracting time: 3 h,and the ethanol dosage: 10 mL.After extracting reaction,the contents of heavy metal in chrysoidine reached United States FDA standard.Conclusion The contents of heavy metal in Chinese herb medicine are notably decreased,and this way provides a new thought and research technique to decrease the contents of heavy metal.
2.CD133, a stem cell marker, in neuroblastoma

BACKGROUND:Neuroblastoma is the most common solid tumor in infants and children. Targeting cancer stem cel therapy can be expected to cure cancer radicaly, but because of smal number, anti-apoptotic and anti-chemotherapy capacity, cancer stem cels are not sensitive to the radiotherapy and chemotherapy.
OBJECTIVE:To explore the expression of stem cel marker CD133 in neuroblastoma and its clinical significance.
METHODS:Fifty-eight children with neuroblastoma admitted at Department of Pediatric Surgery, the First Affiliated Hospital of Xinjiang Medical University, China from June 2004 to February 2014 were enroled, including 46 cases of neuroblastoma and 12 cases of ganglion neuroblastoma. Then, the expression level of CD133 was analyzed by immunohistochemistry method, and the relationship between pathological types, International Neuroblastoma Staging System stage, survival time after surgery and the expression level of CD133 was explored.
RESULTS AND CONCLUSION:There were 22 cases (48%) of neuroblastoma positive for CD133, and 4 cases (33%) of ganglion neuroblastoma positive for CD133. CD133 mainly expressed in the tumor cel cytoplasm. The expression rates of CD133 in different clinical stages were significantly different (P=0.011), which were 21% for stages 1 and 2, 64% for stages 3 and 4, 36% for stage 4S. And the positive rates of CD133 between patients with good prognosis and patients with poor prognosis were significantly different (P=0.031), which were 29% and 57%, respectively. The life cycle of CD133-negative patients was significantly longer than that of CD133-positive infants (P < 0.05). There were tightly connections between CD133 and the occurrence, development, and prognosis of neuroblastoma, and thus, CD133 is of great significance to assess the survival time after surgery and improve of the diagnosis and treatment of neuroblastoma.
3.Reducing bacterial aerosols by dental procedure with Xipayi mouth rinse or H_2O_2 pre-rinse

Aerosol and splatter formed during dental procedure were collected pre and post rinsing with Xipayi mouth rinse or 1% H2O2,cultured 48 hours with blood agar plates,counted the bacterial colonies.Both groups were effective in reducing bacterial aerosols(P 0.05).Bacterial aerosols by dental procedure can be reduced by using mouthwash pre-operationally,both Xipayi mouth rinse and 1% H2O2 are effective mouthwashes.
4.Debates on Plasticity of Stem Ccells (review)

Traditionally, stem cells in adult tissues were believed to be restricted in their differentiation potential and limited to generate the types of cell present in the tissue in which the stem cell resides. Recently, however, many studies indicate that stem cells may display a broader differentiation potential, termed plasticity or transdifferentiation, than what was previously thought. But the validity of this understanding has been challenged by some investigators. In this review, the authors present some standpoints on stem cell plasticity and discuss some phenomena of stem cell plasticity observed on animal models those have been generally accepted currently.
5.Development of Computer Real Time Monitoring System for Rational Clinical Use of Drugs

OBJECTIVE:To promote rational clinical use of drugs by the aid of computer.METHODS:The databases for basic data of drugs,irrational use of commonly-used drugs,compatibility of agents for injection and consultation for drug use in periods of prequancy and lactation were established.Modules for inquiry,addition,revision,deletion and maintaince were designed.The computer real time monitoring system for rational clinical use of drugs was thus developed.RESULTS&CONCLUSION:This system is simple,flexible and quick,and can check the prescription and provide consultation for compatibility between drugs.It is suitable for extensive use in clinical practice.
6.Effects of extract of astragalus on hippocampal delayed neuronal death in rats

Aim To study the effects of extract of astragalus on hippocampal delayed neuronal death of totalcerebral ischemia and 7 days reperfusion in rats.Methods Global ischemia was made by four-vessel occlusion. Electron microscope was used to observe the ultramicrostructure of dorsal hippocampal neurons.Light microscope was used to survey the structure of hippocampal neurons and to count the number of normal neurons in CA1 sector. In the face of escalating problems with pathogen control, the development of proper formulations of existing antibiotics is as important as the development of novel antibiotics. Daptomycin is a lipopeptide antibiotic with potent activity against Gram-positive bacteria. Currently, only injectable solution of daptomycin has been approved for clinical use. In the present study, the formulation of PEGylated liposomal daptomycin (PLD) was prepared and optimized, and its efficacy against methicillin-resistant Staphylococcus aureus (MRSA252) strains was investigated. The obtained PLD had a mean vesicle diameter of (111.5 +/- 15.4) nm and a mean percent drug loading of (5.81 +/- 0.19) % with high storage stability. Potent activity of PLD against MRSA was demonstrated in vitro with a more sustained effect than that of conventional liposomal daptomycin and daptomycin solution. In addition, intravenous administration of a single dose (equal to human use) of PLD significantly increased the survival of mice in a MRSA252 systemic infection model compared with other formulations. Drug distribution in the lung was significantly enhanced following administration of PLD, and no measurable tissue lesions or pathological changes were detected during PLD treatment. Taken together, PEGylated liposomes loaded with daptomycin may represent a promising approach to reduce MRSA252 infections, especially those involving bloodstream dissemination, such as hematogenous pulmonary infection.

8.Effect of Picroside II on ERK1/2 Signal Pathway in Cerebral lschemic Injury Rats.

OBJECTIVETo explore the neuroprotective effect and mechanism of picroside II on extracellular regulated protein kinases1/2 (ERK1/2) signal transduction pathway in cerebral ischemia injuryrats. METHODS The middle cerebral artery occlusion (MCAO) model was established by inserting a monofilament into middle cerebral artery. Totally 96 successfully modeled Wistar rats were divided into the modelgroup, the treatment (picroside II) group, the Lipopolysachcaride (LPS) group, and the U0126 group according to random digit table. Each group was further divided into 3 subgroups, i.e. 6, 12, and 24 h sub-groups. Picroside II (20 mg/kg) was peritoneally injected to rats in the treatment group 2 h after ischemia.LPS (20 mg/kg) and Picroside II (20 mg/kg) were peritoneally injected to rats in the LPS group 2 h after ischemia. U0126-EtOH (20 mg/kg)and Picroside II (20 mg/kg) were peritoneally injected to rats in the U0126group 2 h after ischemia. Equal volume of normal saline was peritoneally injected to rats in the control groupand the model group. The neurobehavioral function was evaluated by modified neurological severity score(mNSS) test. The structure of neurons was observed using hematoxylin-eosinstaining (HE) staining. Theapoptotic cells were detected using terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The expression of phosphorylated extracellular signal-regulated protein kinase1,2 (pERK1,2) in cortex was detected using immunohistochemistry (IHC) and Western blot.
RESULTSAfter cerebral ischemia injury neurological impairment score increased, the damage of neuron in the cortical area was aggravated, apoptotic cells increased in the model group as time went by. The expression of pERK1/2 increased more significantly in the model group than in the control group (P <0.05). The damage of neuron in the cortical area was milder, while apoptotic cells decreased, the expression of pERK1f2 obviously decreased more in the treatment group and the U0126 group (P < 0.05). The early damage of neuron in the cortical area was more severe, apoptotic cells and the expression of pERK12 were comparatively higher in early stage of the LPS group, but the expression of pERK1/2 was somewhat decreased in late stage.
CONCLUSIONSActivating ERK12 pathway could mediate apoptosis and inflammatory reactions of neurons after cerebral ischemia injury. Picroside II could protect the nerve system possibly through reducing activation of ERKI2 pathway, inhibiting apoptosis of neurons and inflammation reaction.

9.Co-delivery of paclitaxel and cyclosporine by a novel liposome-silica hybrid nano-carrier for anti-tumor therapy via oral route.

In this study, we developed a novel liposome-silica hybrid nano-carrier for tumor combination therapy via oral route, using paclitaxel and cyclosporine as a model drug pair. Optimization of the preparation of the drug-loading formulation and characterization of its physicochemical parameters and drug release profile were performed in vitro. Then in vivo pharmacodynamics and pharmacokinetics studies were performed. The results showed that the obtained formulation has a small particle size (mean diameter of 100.2 +/- 15.2 nm), a homogeneous distribution [the polydispersity index was (0.251 +/- 0.018)] and high encapsulation efficiency (90.15 +/- 2.47) % and (80.64 +/- 3.52) % for paclitaxel and cyclosporine respectively with a mild and easy preparation process. A sequential drug release trend of cyclosporine prior to palictaxel was observed. The liposome-silica hybrid nano-carrier showed good biocompatibility in vivo and co-delivery of cyclosporine and paclitaxel significantly enhanced the oral absorption of paclitaxel with improved anti-tumor efficacy, suggesting a promising approach for multi-drug therapy against tumor and other serious diseases via oral route.

10.Baculovirus per os Infectivity Factors Are Involved in HearNPV ODVs Infection of HzAM1 Cells in vitro

Baculoviruses produce two viral phenotypes, the budded virus (BV) and the occlusion-derived virus (ODV). ODVs are released from occlusion bodies in the midgut where they initiate a primary infection. Due to the lack of an in vitro system, the molecular mechanism of ODV infection is still unclear. Here we present data demonstrating that Helicoverpa armigera nucleopolyhedrovirus (HearNPV) ODV infected cultured Hz-AM1 cells in a pH dependent manner. The optimal pH for ODV infection was 8.5, which is same to that in the microvilli of midgut epithelial cells, the ODV native infection sites. Antibodies neutralization analysis indicated that four HearNPV oral infection essential genes p74, pif-1, pif-2 and pif-3 are also essential for HearNPV ODV infection in vitro. Thus, HearNPV-HzAM1 system can be used to analyze the mechanism of ODV entry.
