1.Detection of Helicobacter Pylori Functional OipA Gene and Its Significance

Objective To evaluate the relationship between functional oipA gene of Helicobacterpylori and digestive disease. Methods Biopsy gastric mucosa were obtained from 360 patients who would get gastroscopy.Hp were isolated from urease positive samples and partly urease negative samples,and improved by microscope,urease experiment and 16SrRNA PCR. OipA and cagA of the isolates were obtained by PCR and the statas of oipA signal region were analysised after sequencing. Analyze the relevance between functional oipA gene of Helicobacter pylori and digestive disease.Results 106 isolated Hp were obtained,in which 72 strains with cagA gene positive,87 with oipA signal region gene positive (80 the statas of signal region was on and 1 was off;6 could not be decided).The frequency of functional oipA were both significantly higher than cagA in ulcer and atrophic gastritis.Conclusion The relationship between functional oipA gene and digestive disease was more closer than that of cagA.
2.Establishment of An Indirect ELISA Method with Helicobacter Pylori Recombinated Antigene OipA6 as Coating Antigen

Objective To establish an indirect ELISA method with Helicobacter pylori (HP)recombinated antigene OipA6 as coating antigen after prokaryotic expression and purification.Methods The ELISA plate was coated with Hp OipA6 recom-binant protein as antigen.Serum specimens and enzyme labeled anti human IgG were added.The reaction conditions was op-timized.The results of indirect ELISA were compared with the detection of oipA gene.Results When Hp OipA6 recombi-nant protein was 1.0 mg/ml,the condition of coating was 2~8℃ one night,the dilution of serum specimens was 1∶100 and enzyme labeled anti human IgG was 1∶4 000,the incubation time of enzyme labeled anti human IgG was 40 min,the chro-mogenic time was 15 min,the result of ELISA was the best.The sensitivity and specificity of the indirect ELISA were both higher than the detection of oipA gene.Conclusion It suggested that the indirect ELISA,in which Hp OipA6 recombinant protein were coated,may be an appropriate assisted method in detection of high virulent Hp infection.
3.Glycyrrhizic acid ointment in the treatment of chronic eczema in guinea pigs

Objective To evaluate the therapeutic effect of glycyrrhizic acid ointment on chronic eczema in guinea pigs.Methods Totally,60 Kunming male mice,60 Kunming mice and 60 albino guinea pigs were respectively classified into 6 groups:model group,ointment base group,dexamethasone group,high-,medium-and low-dose glycyrrhizic acid group.To observe the effect of glycyrrhizic acid ointment on the swelling of mouse auricles,dexamethasone and different doses of glycyrrhizic acid ointment were topically applied to both auricles of male mice in corresponding groups followed by the establishement of mouse auricle swelling model induced by xylene.One and a half hours later,the mice were sacrificed,and the weight of swelling auricles was weighed.Scratching behavior was induced by dextran 40 in Kunming mice following the application of dexamethasone or glycyrrhizic acid ointment.The latent period and frequency of scraching were recorded for every Kunming mice.To establish a model of eczema,guinea pigs were sensitized with dinitrochlorobenzene (DNCB,5%) in the neck and back and challenged by DNCB (0.1%) on the inner surface of right ears once a week for 4 weeks;subsequently,both auricles were treated with dexamethasone or glycyrrhizic acid ointment once a day for 10 days.Then,the thickness of auricles of guinea pigs was measured,and skin tissue was obtained from the right ear of guinea pigs and subjected to a pathological analysis.Results The high- and medium-dose of glycyrrhizic acid ointment could suppress the xylene-induced swelling of mouse auricles (P < 0.05 or P < 0.01 ).The latent period of scratching could be lengthened by high dose of glycyrrhizic acid (P < 0.05).The three doses of glycyrrhizic acid all reduced scratching frequency in Kunming mice (P < 0.05 or P < 0.01 ).Attenuated swelling of auricles (P < 0.05 or P < 0.01 ) and relieved pathological changes were observed in guinea pigs after treatment with glycyrrhizic acid ointment.Conclusion Glycyrrhizic acid ointment shows a therapeutic effect on chronic eczema in guinea pigs.

Objective To investigate the analgesic efficacy and safety of continuous epidural infusion of 0.2% levobupivacaine in patients after lower abdominal surgery.Methods Sixty ASA Ⅰ - Ⅱ patients undergoing lower abdominal gynecologic operation under epidural anesthesia were randomly divided into two groups : group L levobupivacaine (n = 30) and group R ropivacaine ( n = 30) . An epidural catheter was placed at L2-3 . After operation the catheter was connected through a 3-way Stopcock to a Graseby 9 500 infusion pump for continuous infusion of 0.2% levobupivacaine or 0.2% ropivacaine at a rate of 4 ml?h-1 and a Graseby 3300 infusion pump for PCEA with 0.01% morphine (bolus dose = 2ml, lockout interval = 10 min, total dose limit = 16 ml?4 h-1) (1) VAS score (0 = no pain, 100 = severe pain). (2) the ratio of the number of attempts calculated (D1) to the number of successfully delivered doses (D2) (D1/D2), (3) modified Bromage Motor scale score, (4) Bruggmann comfort scale (BCS), (5) Ramsay sedation score and (6) the severity of side effects were recorded 0, 2,4, 8, 12, 20 and 24h after operation.Results The demographic data (age, body weight, height) and duration of operation were comparable between the two groups. 6.Quality analysis of urinary fluoride detection ability from 2010 to 2012 in Guizhou endemic fluorosis monitoring network laboratories

Objective To evaluate the urinary fluorine detection quality of endemic fluorosis monitoring network laboratories in Guizhou Province and promote the improvement of quality control.Methods From 2010 to 2012,according to the method for determination of fluoride in urine by ion selective electrode,in 11 endemic fluorosis monitoring network laboratories in Guizhou,an quality control assessment was done.In the internal quality control assessment,standard substance detection inspection was used for personnel parallel samples and equipment comparison,t test was used to compare the inter-group differences.In the external quality control inspection,blind urine fluorine sample was tested,and a Z score value evaluation method was used to compare the relative error of the test results.Results In the assessment of internal quality control,the parallel comparison test results of sample,personnel and instrument showed that the differences were not statistically significant (t =-1.81-1.80,P > 0.05).In 2010,in the external quality control inspection,there was 1 laboratory |Z|≥3,and 2 test results |Z| >2;in 2011,there was 4 test results |Z| > 2;in 2012,there was 5 test results |Z| > 2;in 2010,there was one laboratory in a blind test,relative error of the results > 20%;in 2011 and 2012,there was a laboratory in a blind test,relative error of the results > 10%.Conclusion The endemic fluorosis monitoring network laboratory testing ability in Guizhou is relatively good,but there is an obvious individual laboratory system error.

Objective To explore the effects and the mechanism of manumycin on KAT-18 cells Methods Human ATC (anaplastic thyroid cancer) KAT-18 cells were used. The cytotoxicity was analyzed by SRB assay. Apoptosis and cellular nitric oxide were detected by flow cytometry using annexin v and NO sensor dye. Superoxide anion was measured with a fluorescent plate reader by DHE.GSH was assayed by fluorescent Monochlorobimane. The SOD activities were assayed by colorimetric methods. The protein expression of Mn-SOD was determined by western blot. Results Manumycin decreased the viability of KAT-18 cells in a dose-dependent manner. Manumycin induced apoptosis significantly and NO generation simultaneously. Manumycin also induced superoxide anions generation. Manumycin reduced intracellular GSH in a time-course manner. However , manumycin did not decrease the SOD activity after 6 h treatment and Mn-SOD expression. A delayed induction of SOD activity was observed after 24 h manumycin treatment. N-acetyl-L-cysteine blocked NO and superoxide anions generation and apoptosis induction by Manumycin. Furthermore , NAC protected KAT-18 cells from the cytotoxicity of manumycin. Conclusion Manumycin induces apoptosis and has cytotoxic effects on KAT-18 cells. Cellular NO and superoxide anions generation are required for Manumycin-induced apoptosis in KAT-18 cells.
10.Helicobacter pylori oipA gene switch status and its association with gastic diseases

Objective To understand the status of oipA gene of Helicobacter pylori (H.pylori) and to evaluate the relationship between oipA gene and digestive disease, Methods Biopsy gastric mucosa were obtained from 360 patients with digestive disease under gastroscopy. H.pylori was isolated from urease positive and some negative samples, and then identified by microscope, urease test and 16 S rRNA PCR. oipA and cagA genes of the isolates were amplified by PCR and the status of oipA signal region was analyzed after sequencing. The relationship between functional oipA gene of H. pylori and digestive disease was investigated. Results One hundred and six isolats were obtained. Seventy-two strains were positive for cagA gene and 87 were positive for oipA gene in signal region. Eighty strains were on open status in signal region called functioal oipA gene and 1 was on off status. The status of other 6 strains were uncertain. The frequency of functional oipA in ulcer and atrophic gastritis were higher than that of cagA(100% and 78.26% vs 58.33% and 39.13%).Conclusion Functional oipA gene is closely related with ulcer and atrophic gastritis.
