Journal of Chinese Physician 2015;17(8):1200-1203
Objective To investigate the expression of microRNA in plasma of gallbladder cancer and its clinical significance. Methods miRNA microarray was performed in seven gallbladder cancer tissues and seven adjacent normal gallbladder tissues to identify differentially expressed miRNA.The results were validated in plasmas of 40 gallbladder cancers and 20 health people with real-time polymerase chain reaction (RT-PCR).Results Eleven gallbladder cancer-related miRNAs were identified with miRNA microarray analysis.Among them,miRNA-21,miRNA-370,miRNA-187,miRNA-122,and miRNA-202 were up-regulated,and let-7a,miRNA-200b,miRNA-143,miRNA-31,miRNA-335,and miRNA-551 were down-regulated in gallbladder cancers.Moreover,let-7a,miRNA-21,miRNA-187,miRNA-143,miRNA-202,and miRNA-335 were validated with RT-PCR,which were consistent with miRNA microarray results with significant difference between gallbladder cancers and health people (P < 0.05).Furthermore,miRNA-187,miRNA-143,and miRNA-202 were related to clinical and pathological features(P <0.05).Conclusions These data indicate that The differentially expressed plasma miRNAs are related to gallbladder carcinogenesis and might be the novel biomarkers of gallbladder cancers.
4.The Role of FUS/TLS in the Regulation of Gene Expression and the Correlation with Neurodegenerative Diseases
Journal of Kunming Medical University 2016;37(8):1-4
Neurodegenerative diseases are chronic lesions caused by neuronal degeneration and secondary demyelination.Recent studies have shown that a DNA/RNA binding protein named fused in sarcoma/translocated in liposarcoma protein (FUS/TLS) is associated with neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS),frontotemporal dementia (FTD) and polyglutamine diseases.Mutations or dysfunction in FUS/TLS are responsible for the abnormal processes in the related gene transcription,RNA processing, transportation and translation and are highly correlated with the development of neurodegenerative disease.Discovery and the research of FUS/TLS not only contribute to the understanding of neurodegenerative disease at RNA level, but also provide a new way for the prevention and treatment of neurodegenerative diseases.

Journal of Huazhong University of Science and Technology (Medical Sciences) 2007;27(4):457-60
To investigate the relationship between the expression of cyclooxygenase-2 (COX-2) and menstrual cycle, the regulatory effects of 17-beta-estradiol (E(2)) and medroxyprogesterone acetate (MPA) on the expression of COX-2 in cervical cancer Hela cells were examined. Cervical cancer specimens were obtained from 47 pre-menopausal patients. The phase of menstrual cycle was determined by case history and HE staining of uterine endometrium. COX-2 was immunohistochemically stained by SABC staining and the staining intensity was determined with computerized image analysis system. Hela cells were incubated with alcohol, E(2), E(2)+MPA, MPA for 12, 24 and 48 h respectively. The expression of COX-2 in Hela cells was detected by Western blotting and reverse transcriptase-polymerase chain reaction (RT-PCR). Our results showed that the expression of COX-2 was significantly higher during proliferative phase than secretory phase (P<0.05), but there was no difference in the positive rate between proliferative phase and secretory phase (P>0.05). Incubation with E(2) could significantly enhance the expression of COX-2 continually. On the contrary, E(2)+MPA and MPA alone could decrease the expression of COX-2 as compared with the control and E(2) group (P<0.05 and P<0.01 respectively). It is concluded that the expression of COX-2 in cervical cancer of pre-menopausal patients and Hela cells was regulated by estrogen/progestogen.

Chinese Journal of Tissue Engineering Research 2014;(2):277-282
BACKGROUND:Application of mechanical valve and bioprosthetic valve replacement is an effective means for the treatment of end-stage valve disease. However, their clinical application is constrained by multiple factors. Tissue-engineered heart valves with biological activity hold the potential to overcome the defect of the mechanical valves and bioprosthetic valves. Choosing appropriate cells for cellseeding is a significant aspect for the research of tissue-engineered heart valves. A growing number of somatic cells and stem cells have been used for construction of tissue-engineered heart valves, however, not yet achieve the desired result.
OBJECTIVE:To review the studies on seed cells for tissue-engineered heart valves and the cellseeding methods in vitro on the basis of the cellular components of heart valves.
METHODS:First author searched PubMed and Wanfang database with computer for related articles published from January 2000 to December 2012. The keywords were“tissue engineering, heart valves, cel”. Preferred articles concerned seed cells of tissue-engineered heart valves with the relevance and authority. A total of 39 articles published in the prestigious journals were reviewed.
RESULTS AND CONCLUSION:Endothelial cells and interstitial cells were mainly valvular cellular components. In the early stage, endothelial cells and fibroblasts were used to construct tissue-engineered heart valves. With deep stem cellresearch, the application of pulsatile bioreactor cultivation mesenchymal stem cells holds a great potential to produce tissue-engineered heart valves.
