Objective To explore the physical and chemical properties of electric charge modified liposomes and the immune enhancement effect as influenza vaccine adjuvants.Methods Four kinds of electric charge-modified liposomes were prepared with soybean lecithin,cholesterol,N-1-(2,3-dioleyloxy) propyl-N,N,N-trimethyllam-moniumchloride(DOTAP)and 1,2-dioleoyl-sn-glycero-3-phosphocholine(DOPC)(DOTAP and DOPC mass ratio 1:4,2:3,3:2 and 4:1).The tetravalent influenza vaccine was mixed with the four liposomes according to the volume fraction of 1.4:1.0,which was prepared into cationic modified influenza vaccine liposome freeze-dried powder by freeze-thaw freeze-drying method.The liposome freeze-dried powder was redissolved with PBS,the particle size and Zeta potential were measured by laserparticle size analyzer,and the encapsulation efficiency of liposome was measured by Lowry method.231 female KM mice were randomly divided into negative control group(PBS),positive control group(tetravalent vaccine bulk),cationic liposome group(four electric charge modified influenza vaccine liposomes) and neutral liposome group(non-electric charge modified influenza vaccine liposomes),which were injected intraperitoneally at 7,14 and 28 d,0.2 mL per mouse.The cellular immune effect was evaluated by MTT assay and T lymphatic surface labeling,and the humoral immune effect was evaluated by hemagglutinin inhibition(HI) at 3,7,14,28,42 and 56 d of immunization.Results When the mass ratio of DOTAP to DOPC was 4:1,the particle size and Zeta potential of cationic modified liposomes reached the maximum,which were 529.65 nm and 12.05 mV respectively,and the encapsulation efficiency was also high,which was 81.82%.Stimulus index(SI) and CD4~+/CD8~+value of spleen cells of mice in the four cationic liposome groups were significantly higher than those in the negative control group(F=4.651～25.866,each P<0.05),among which,the two indexes in 4:1 cationic liposome group were higher than those in the other three groups;mice in this group produced early humoral immune effect 3 d after immunization,and the antibody titer reached the highest 42 d after immunization and maintained a high level during the whole immunization cycle.Conclusion Cationic liposomes with different electric charge modifications improved the immune effect of influenza vaccine,the immune enhancement effect was the best and the immune effect lasted the longest when the mass ratio of DOTAP to DOPC was 4:1.

Objective To observe the effects of wogonin on lipopolysaccharide(LPS-induced) proinflammatory cytokine production in activated microglia.Methods By using LPS-induced microglia activation model,nitric oxide(NO),tumor necrosis factor-alpha(TNF-?) and interleukin-6(IL-6) secreted by extracellular fluid were measured by ELISA.Results Microglia markedly produced NO,TNF-? and IL-6 in response to LPS.Wogonin,administered at 20-30 ?mol/L,significantly reduced the production of NO,TNF-? and IL-6 in microglia.Conclusion Wogonin has an inhibitory effect on the production of proinflammatory cytokines.

Objective To explore the correlated factors of bilirubin neurotoxicity. Methods From July 2007 to October 2007, 53 hospitalized patients with jaundice in neonatal ward of Peking University Third Hospital were enrolled in the study. The relationship between serum free bilirubin and parameters of brainstem auditory evoked potential(BAEP) was analyzed to screen the sensitive index of bilirubin neurotoxicity. Multivariate analysis was conducted between the sensitive parameter and various factors. Results Partial correlation analysis showed that the wave Ⅴ latency was positively correlated to serum free bilirubin(r=0. 228, P= 0. 020) after the corrected gestational age was controlled. There were no correlation between other latencies or interwave intervals and serum free bilirubin. Linear regression showed the equation was wave Ⅴ latency= 9.721-0. 082 × corrected gestational age+0. 064 × bilirubin/albumin+ 0. 504 × acidemia(F= 7. 534, P＜0.01). Conclusions Wave Ⅴ latency is the sensitive indicator of bilirubin neurotoxicity, which is correlated with the value of bilirubin/albumin and acidemia.

The hypotensive effect of PAMC and its mechanism were studied.In conscious renal hypertensive rats, intraperitonal injection of PAMC once a day for 7d at the dosage of 11 mg/kg ( 1 time/day ) and oral administration of the drug 3 times a day for 14 d at the dosage of 12 mg/kg/d, the arterial pressure decreased 29.0?2.8% and 20.3 ? 1.4%, respectively.In normal anesthetized dogs and rats, intravenous injection of PAMC at the dosage of 0.4 mg/kg, the mean arterial pressure deceased 43.7 ? 7.9 and 39.7 ? 2.3%. In anesthetized rabbits and rats, intraduodenal injection of PAMC at the dosage of 4 mg/kg, the mean arterial pressure decreased 24.5 ? 3.8% and 38.2 ? 11.4%, respectively.The experiments suggested that inhibition of the (?) -adrenal receptors may be the principal mechanism of the hypotensive action of PAMC. Objective:To analyze the distribution of HFE gene (hemochromatosis gene) mutation in normal and iron overload men and discuss the influence of HFE gene mutation on iron overload status of Chinese adult men..Method:The data of 226 normal and 331 iron overload men were drawn from National Nutrition and Health Survey in 2002 as control and case group respectively to analyze and compare the distribution of HFE mutation including C282Y mutation, H63D mutation and S65C mutation in normal and iron overload men.Results:In case group, normal gene was 90.03%, homozygosity for H63D mutation 0.6%, heterozygosity for H63D mutation 9.07%, heterozygosity for S65C 0.3%. In control group normal gene was 94.25%, heterozygosity for H63D 5.75%, and no S65C and C282Y mutation was detected. Conclusion: HFE gene mutation was not the major reason resulting in iron overload in Chinese adult men.

ObjectiveTo assess the reliability and validity of Chinese version of tools in measuring yangsheng.MethodsChinese version of tools in measuring yangsheng was administered to 706 elderly citizens and evaluated for the reliability and validity.ResultsThe fittingness of the both exiting model were unidentified.A model of six factors and twenty-six items was determined using exploratory factor analysis.The fitness of such model was perfect ( RMSEA =0.050,AGFI =0.871,GFI =0.902,NFI =0.903,CFI =0.953 ) and all the construct reliability were more than 0.6 while the average variance extracted were higher than 0.5 except the diet yangsheng factor.The Cronbach's α of Chinese version of tools in measuring yangsheng was 0.918 and Cronbach's α of factors were all above 0.7.ConclusionThe Chinese version of tools in measuring yangsheng appears to have good validity and reliability to estimate the Yangsheng of elderly citizens in China.
