1.Inhibitory effect of propylene glycol mannate sulfate on growth of rabbit lens epithelial cells in vitro

AIM:To investigate the inhibitory effect of rabbit lens epithelial cell(RLEC)survival and growth by propylene glycol mannate sulfate(PGMS)on the rabbit capsular bag in vitro.METHODS;Capsular bags were prepared from rabbit eyes after extracapsular cataract extraction(ECCE)and incubated in 0.2,0.4,0.8g/L PGMS in 2,5,10 minutes incubation periods.After treatment,the capsular bags were cultured for 7 days in Dulbecco minimum essential medium(DMEM)supplemented with 50mL/L fetal calf serum(FCS).The specimens were examined with light microscopy and transmission electron microscopy(TEM).Capsular bags without receiving PGMS only served as controls.RESULTS:PGMS inhibited the proliferation of RLEC in the manner of concentration and time dependentment.At the threshold protocol of incubation in PGMS at 0.8g/L for 5 or 10 minutes,proliferative activity of cells were largely arrested and nearly no RLEC was seen on the posterior capsule(P<0.05).Control group had no effect on structure and proliferative activity of RLEC,and the growth proceeded rapidly so that the posterior capsule were totally covered by a confluent monolayer of cell by the end of 7 days.Under TEM,the cells in the control group were tightly arrayed with clearly defined cellular boundary and structure;while cellular deformity and undefined intracellular structure could be seen in the 0.4g/L and 0.8g/L experimental groups.CONCLUSION:PGMS can effectively inhibit the proliferation of RLEC.
3.Inhibitory effect of Meloxicam on the cultured fibroblasts from the excised pterygium

AIM: To investigate the association between cycloxygenase-2 (COX-2) expression and VEGF intervention as well as the inhibitory effect of Meloxicam on the cultured human pterygium fibroblasts (HPF).METHODS: Expression of COX-2 was measured by immunohistochemistry in the cultured HPF from twenty excised pterygium cases. Expression of COX-2 in HPF was measured by Western blot following the treatment of vascular endothelial growth factor (VEGF) at the different concentrations. In addition, the effect of Meloxicam on proliferation of HPF was studied by adding the different concentrations into the cultured HPF plates by Mono-nuclear cell direct cytotoxicity (MTT) reduction assay.RESULTS: COX-2 expression was present in the cultured HPF. The level of the expression increased following VEGF treatment. The proliferation of the cultured HPF decreased following addition of the different concentrations of Meloxicam (from 75μ mol/L to 300μ mol/L) and the magnitude of the inhibition was dose-time dependent.CONCLUSION: COX-2 levels in the cultured HPF werepositively associated with VEGF stimulation and Meloxicam was inhibitory to HPF proliferation.
4.Effectiveness of team-based continuous nursing on improving the life function of children with cerebral palsy

Objective To observe the effect of team-based continuous nursing on improving the life function of children with cerebral palsy. Methods From March 2014 to March 2016, 35 children aged 18-36 months with cerebral palsy were selected as the experimental group;35 matched patients with the same age, same gender, same Gross Motor Function Classification System (GMFCS) and functionally similar were selected as the control group by matching design. Among them, 32 cases were followed up in the experimental group while 30 cases in the control group. In the control group, routine discharge education was adopted, but the experimental group was treated by team-based continuous nursing. The ability of daily living was evaluated by the Pediatric Evaluation of Disability Inventory (PEDI). Neuropsychological development was evaluated by the Gesell Development Scale and Body balance was evaluated by the Berg Balance Function Scale after discharge and also after 6 months follow-up. Results After the intervention for 6 months, the functional skill and assisting assistance of PEDI was (77.56 ± 23.58), (38.78 ± 13.96) points in the experimental group, and (65.12 ± 24.23), (30.35 ± 12.78) points in the control groups, there were significant differences between two groups(t=2.0485, 2.4750, P<0.05). The scores of Gesell Development Scale was (76.12 ± 18.35) points in the experimental group, and (71.85 ± 18.46) points in the control group, there were no significant differences between two groups(P>0.05) , but the experimental group was higher than the control group. The scores of Berg Balance Function Scale was(33.20 ± 4.12) points in the experimental group, and (29.67 ± 4.98) points in the control group, there were significant differences between two groups (t = 3.0488, P<0.05). Conclusions To improve the life function of children with cerebral palsy, the implementation of the team-based continuous nursing had a significant effect.
5.Sample preparation for iodine measurement by arsenic cerium catalytic spectrophotometry

Objective To screen the suitable sample digestion method for measuring iodine in serum by arsenic cerium catalytic spectrophotometric method , and to carry out the methodology test for the newly developed arsenic cerium catalytic spectrophotometry for determination of iodine in serum .Methods Methodology evaluation.Experimentswere on the sample on the sample digestion method of serum iodine with several common digestive agents . After digestion treatment , the concentration of iodine in serum was determined by arsenic cerium catalytic spectrophotometry .The linearity and linear range of the standard curve, detection limit, precision, recovery rate and anti -interference ability of the newlydeveloped arsenic cerium catalytic spectrophotometry were tested .Results Sample pretreatment method included in the newly developed arsenic cerium catalytic spectrophotometry was selected as follows: perchloric acid -sodium chlorate solution was used as the digestive agent to digest 120 min at 130℃.The calibration relation of C =a+blgA( C: iodine concentration ,A: measuring absorbance ) of the newly developed method existed when arsenic ceriumcatalytic reaction was kept at a certain stable temperature range from 13℃ to 30℃ and in certain stable reacting time .The linear range of the calibration curvewas 0 -300 μg/Land the linear correlative coefficient was -0.9996 --0.9999.The intra assay coefficients of variation ( CVs ) were 0.70%,0.70%and 0.74%(n=6), and the inter assay CVs were 0.57%, 0.51% and 0.57% (n=6) for 3 serum samples.The average recovery was 97.9% with a range from 92.3% to 105.8% for 3 serum samples.Conclusions The newly developed arsenic cerium catalytic spectrophotometry has good precision and accuracy .The sample digestion agent is easy to be prepared and reserved , and the instrument is simple and easy to be operated .The method can be widely used as a reliable technique for measuring iodine in serum.
6.Method for measuring different range of iodine concentration in drinking water by arsenic cerium catalytic spectrophotometry

Objective To establish a catalytic spectrophotometric method for determination of iodine in water using the same arsenious acid solution and ceric ammonium sulfate solution as those used in the 2016 version standard method for determination of urinary iodine,and to meet the needs of wide concentration range of water iodine detection.Methods After pretreatment of the water sample with the effective chlorine of sodium dichloroisocyanurate solution for eliminating the interference of reducing substances at room temperature,the concentration of iodine in water was determined by arsenic cerium catalytic spectrophotometry using the same 0.025 mol/L arsenious acid solution and 0.025 mol/L ceric ammonium sulfate solution as those used in the 2016 version standard method for determination of urinary iodine.The linear relationship of the standard curve and the linear rang of different iodine concentration range (0-100,0-400,0-800μg/L),the detection limit,the precision and the accuracy of the sample were tested.Results The calibration relation of C =a + blgA (C:iodine concentration,A:measuring absorbance) in the new method existed when arsenic cerium catalytic reaction was kept at a certain stable temperature ranging from 15 ℃ to 30 ℃ in certain stable reacting time.The linear correlative coefficients absolute value of different iodine concentration range (0-100,0-400,0-800 μg/L) were all greater than 0.999 0,corresponding to the water iodine detection limits were 0.3 μ,g/L (sample volume of 0.80 ml),1.2 μg/L (sample volume of 0.20 ml),and 2.2.μg/L (sample volume of 0.10 ml),respectively.The coefficients of variation (CV) of the three different iodine concentration range were all below 1.0% (n =6).The iodine recovery rate range of a total of 10 different water samples in these three different concentration range was 95.8%-98.7%,98.3%-103.7% and 98.5%-104.5%,and the average recovery rate was 97.6%,100.4% and 102.4%,respectively.In the range of these three different standard curves,water iodine standard materials GBW09114c,GBW09114a and GBW09113c were measured.The relative errors between the results and the given values were < 3.0%,which were in the range of uncertainty of the given value.Conclusion This method verified by methodology experiments has wide linear range,high precision,accuracy,and anti-interference ability,good reproducibility,and is easy to operate,with reduced amount of arsenic waste,reduced environmental pollution,and is suitable for application in different areas to determine water iodine.
7.Effect of two 17?gauge aspirating needles on outcome of transvaginal ultra?sound guided oocyte retrieval for IVF/ICSI

Objective To compare the embryo development and pregnancy outcomes of patients by using two different 17?gauge aspiration needles in transvaginal ultra?sound guided oocyte retrieval. Methods A retrospective study was performed to analysis the embryo development and pregnancy outcomes of patients by using different oocytes aspiration needle:import needle(group A)and domestic needle(group B),including puncture follicles,oocytes rate,cleavage,available embryos,high?quality embryo,the total fertility rate,bleeding, moderately severe OHSS incidence,pregnancy rate,embryo implantation rate and abortion rate,etc. Results There was no significant statistical difference in the two different ovum aspiration needles. Conclusion The embryo development and pregnancy outcomes were similar with both needles. Each needle has its advantages and flaws in transvaginal ultra?sound guided oocyte retrieval ,and we should make a comprehensive choice weighing the practical situation of patients,cost,effectiveness and the doctors' preferences.

OBJECTIVETo observe the effect of total ginsenosides (TG) on monocrotaline (MCT) induced right ventricular hypertrophy rats, and to explore its correlation with calcineurin (CaN) pathway.
METHODSFifty male Sprague Dawley rats were randomly divided into the normal control group, the MCT model group, and the low, middle, high dose TG treatment groups, 10 in each group. All medication was performed by peritoneal injection for 18 days. Right ventricular peak systolic pressure (RVSP), right ventricular hypertrophy index (RVHI), and right ventricular weight/body weight (RVW/BW) were measured. Intracellular free calcium concentrations were measured by Ca2+ fluorescence indicator Fura2/AM. The atrial natriuretic factor (ANF) and CaN mRNA expression of the myocardial tissue were quantitatively analyzed by Real-time PCR. The protein expression of CaN was detected by Western blot.
RESULTSCompared with the MCT model group, preventive treatment of TG at the 3 doses could significantly reduce RVSP, RVHI, RVW/BW, and ANF mRNA expression, and decrease Ca2+ concentration in myocardial cells, CaN mRNA and protein expression in the myocardial tissue.
CONCLUSIONTG could obviously improve MCT-induced right ventricular hypertrophy, which was possibly achieved through suppressing MCT-activated CaN signal transduction.
