1.Laparoscopic surgery in gynecological malignant tumors

Laparoscopic surgery is widely used in the treatment of gynecological malignancies,including the radical surgery or staging surgery of early cervical cancer,endometrial cancer and ovarian cancer.Its feasibility and safety of clinical practice in gynecologic malignancies has been approved,and the postoperative follow-up results indicate that the effectiveness of laparoscopic surgery is similar to open surgery.But the laparoscopic surgery treatment of gynecological malignancies is still in dispute.The research will focus on the affects of pneumoperitoneum to the malignant behavior of tumor cells and the mechanism about implantation metastasis of puncture site in future.
2. Association of TNF-863 and CGRP979 gene polymorphims with susceptibility to severe chronic periodonititis i Chinese Han nationality

Objective: To investigate the relationship of TNF-A-863 and CGRP979 gene polymorphisms with the susceptibility to severe chronic periodontitis in Chinese. Methods: Buccal swabs were collected from 100 adult patients with severe chronic periodontitis and 118 healthy adult controls. DNA was extracted from each subjects of the two groups. PCR-LDR technique was used to identify the genotypes of TNF-A-863 and CGRP979. The difference in the genotypes between the two groups was analyzed by statistics software. Results: The genotype of TNF-A--863 was mainly TNF-A-863 A/C in patients with severe chronic periodontitis and TNF-A-863 C/C in healthy controls. There were significant differences in TNF-A-863 distribution between the two groups( P<0.05). We also found that there were significant differences in genotype distribution of CGRP979 between the two groups (P<0.05) ,with A/C predominating in patients with severe chronic periodontitis. Condusion: TNF-A-863 polymorphism is associated with severe chronic periodontitis; A/C of the CGRP979 loci might be a factor for severe chronic periodontitis.
3.Influence of surgical resection extent on prognosis of high-risk gastrointestinal stromal tumors

Objective To investigate the clinical characteristics,surgical resection extent and prognosis influencing factors of pri-mary high-risk gastrointestinal stromal tumors(GIST).Methods The clinicopathological and follow-up data of 48 patients with pri-mary resectable high-risk GIST in this hospital from January 2005 to January 2013 were retrospectively analyzed.The influence of three kinds of different surgical resection methods on the prognosis of GIST was analyzed.Results 34 cases were treated by R0 re-section(70.83%),6 cases by R1 resection(11.50%)and 8 cases by R2 resection(16.67%).All cases were given the postoperative adjuvant therapy of imatinib mesylate.The average medication time was(26.5±13.6)months.The occurrence rate of adverse drug reactions was 100%,and the main symptoms were swelling and leukopenia.The 5-year survival rates in 3 kinds of resection modes were 97.06%,66.67% and 25.00% respectively.The univariate and multivariate prognostic analysis showed that the surgical re-section extent and the tumor rupture were the independent prognostic factors in the high-risk GIST patients.Conclusion The intact operative resection and preventing the tumor rupture are the keys to increase the postoperative survival rate of GIST patients.
4.Effects of natural moxibustion on cytokines and specific transcription factors in asthma rats.

OBJECTIVETo explore the mechanism of natural moxibustion on regulating immune imbalance in asthma rats.
METHODSSeventy SD male rats were divided into a normal group, a placebo group, a dexamethasone group, a big-cake for long-course moxibustion group, a big-cake for short-course moxibustion group, a small-cake for long-course moxibustion group and a small-cake for short-course moxibustion group, ten rats in each one. The rat model of asthma was established by egg albumen sensitization and stimulation in all the groups except the normal group. The natural moxibustion was used in all moxibustion groups, in which big cake of 1 cmX 1 cm size was used in the big-cake groups and small cake of 0.5 cmX 0. 5 cm size was used in the small cake groups. According to relevant acupoints, the natural moxibustion was performed, 5 h per time, once a day. Four times of treatment was considered one course, and three courses were required in the long-course groups and one course was required in the short-course groups. Intraperitoneal injection of dexamethasone was applied in the dexamethasone group, which had the same course as long-course moxibustion group. After the treatment, changes of EOS in peripheral blood of asthma rats were observed; enzyme linked immunosorbent assay (ELISA) was applied to test the contents of IgE, IL-4 and IFN-gamma in the lung tissue; real-time Q-PCR method was adopted to measure the expression level of transcription factor T-bet and GATA binding protein 3 (GATA-3) in the lung tissue.
RESULTSCompared with the normal group, the EOS in whole blood as well as IL-4 and IgE in plasma were all increased in the placebo group (all P< 0. 01), IFN-gamma in plasma was obviously decreased (P<0. 01); while the levels of EOS, IgE and IL-4 were significantly reduced (all P<0. 01), the content of IFN-gamma was increased (P<0. 01) in all moxibustion groups and dexametnasone group. Compared with the normal group, the expression of T-bet mRNA in the placebo group was significantly reduced (P<0. 01). Each treatment group could significantly increase the expression of T-bet mRNA and reduce that of GATA-3 mRNA (P<0. 01). Compared with the short-course moxibustion groups, the expression of T-bet mRNA was obviously increased in the long-course moxibustion group and dexamethasone group (both P<0.01), and that of GATA-3 mRNA was reduced (P<0.01). There was no significant difference between long-course moxibustion group and dexamethasone group (P> 0. 05), and also no significant difference could be seen between big-cake moxibustion group and small-cake moxibustion group (P>0. 05).
CONCLUSIONThe natural moxibustion could obviously reduce airway inflammation in asthma rats. With time passing, the efficacy is enhanced, indicating evident timeliness, which has no apparent relationship with the size of moxibustion cake.

5.Relationship between tumor necrosis factor-αgene-308 promoter polymorphism and susceptibility to development of bronchial asthma in Uighur and Kazakh population in Xinjiang

Objective To investigate the relationship between the polymorphism of the tumor necrosis factor-αgene-308 promoter and bronchial asthma (BA)in Uighur and Kazakh population in Xinjiang.Methods PCR product sequencing method was used to detect the polymorphism distribution of TNF-αgene in 60 BA patients and 60 controls among Xinjiang Uighur and Kazakh population.Then we analyzed the association between different TNF-αgenotypes and BA in Uighur and Kazakh population.Results The distribution of GG,GA and AA genotype was significantly different between the two groups.Statistical analysis showed that BA group had a significantly higher TNF-αA allele frequency than that of the control one (P <0.05).After adjustment for sex and age,we found the A allele was a risk factor for BA pathogenesis (P <0.05).Conclusion The polymorphism of TNF-α gene-308 may be associated with the susceptibility to BA in Xinjiang Uighur and Kazakh population,so TNF-α gene-308 may be considered as a genetic marker for early identification of individuals at high risk for BA,which may play an important role in preventing the development of BA in clinical practice.
6.ANTHOCYANINS ATTENUATE LIPID ACCUMULATION THROUGH ADENOSINE MONOPHOSPHATE ACTIVATED PROTEIN KINASE IN HepG2 CELLS

Objective To study the effects of anthocyanins on hepatic lipid accumulation and to explore the possible mechanisms. Method HepG2 cells were treated with 1,10 and 100 ?mol/L cyanidin-3-0-?-glucoside (Cy-3-g) for 1h,carnitine palmitoyl transferase-I (CPT-I) protein level and acetyl CoA carboxylase (ACC) phosphorylation,triglyceride,fatty acid oxidation,the activity of AMPK,AMPK and p-AMPK protein levels were evaluated. Results Compared with the control,the treatment of HepG2 with 1,10,100 ?mol/L Cy-3-g resulted in significant decrease of intracellular triglyceride,and significant increase of CPT-I protein,and AMPK activity and phosphorylation. However,there were no differences in AMPK protein level among all groups. The treatment of 100?mol/L Cy-3-g promoted the fatty acid oxidation in HepG2. The treatment of 10,100 ?mol/L Cy-3-g significantly increased ACC phosphorylation. Conclusion Anthocyanin Cy-3-g could attenuate hepatic lipid accumulation in HepG2. The mechanism may result from the regulation of the AMPK-ACC-CPT-I signal pathway and promotion of fatty acid oxidation.
7.Effect of glucagon-like peptide-1 agonist on insulin signaling pathway in skeletal muscle of mice with insulin resistance

Objective To study the effect of glucagon-like peptide-1 agonist(exendin-4)on insulin signaling pathway in skeletal muscle of mice with insulin resistance(IR). Methods Thirty male KM mice were divided into normal control group(NC group, n=10)and IR group(n=20). The mice in NC group and IR group were fed with routine diet and high-fat diet for 16 weeks, respectively. After IR models were successfully established, 10 mice in IR group were treated with exendin-4(Ex group, 3μg·kg-1·d-1)for 4 weeks. NC group and IR group were given the same dose of normal saline. Body weight, fasting blood glucose(FBG), and fasting insulin(FINS)were measured before and after the intervention, and homeostasis model assessment for insulin resistance ( HOMA-IR ) was calculated . The muscle glycogen content were measured by Periodic Acid-Schiff Stain ( PAS ) . The expressions of insulinreceptor(IRC), insulin receptor substrate-1(IRS-1), and phosphatidylinositol 3-kinase(PI3K)in the skeletal muscle were measured by Western blot and immunohistochemistry. Results Before intervention, the body weight, FBG, and HOMA-IR of IR group and Ex group were higher compared with NC group(all P<0. 05), but FINS did not rise significantly(P>0. 05). Compared with before intervention, the body weight of three groups after intervention were all increased, but increased more significantly in the IR group. After intervention, the FBG and HOMA-IR in Ex group decreased, compared with IR group and Ex group of non-intervention (all P<0. 05), but FINS did not decrease significantly(P>0. 05). The skeletal muscle cells in NC control were colored extensively and more colored granules, while IR group were less colored, and Ex group were between NC group and IR group. Compared with NC group, the expressions of IRC, IRS-1, and PI3K in the skeletal muscle of IR group were significantly decreased(P<0. 05), not in Ex group(P>0. 05). Compared with IR group, the expressions of IRC, IRS-1 and PI3K in Ex group were higher(P<0. 05). Conclusions Exendin-4 improves insulin resistance by up-regulating the expressions of IRC, IRS-1, and PI3K in the skeletal muscle and promoting the synthesis of muscle glycogen.
8.Atorvastatin attenuates activation and injury of human microvascular endothelial cells induced by oxidized low density lipoprotein

Aim To investigate the effects of atorvas-tatin ( ATV) on activation and injury of microvascular endothelial cells induced by oxidized low density lipo-protein ( ox-LDL) . Methods Cultured human micro-vascular endothelial cells were pre-incubated with ATV for 24 h prior to exposure of endothelial cells to ox-LDL. After exposure of endothelial cells to ox-LDL, the cell viability was measured by MTT method, LDH in supernatants was determined by enzyme activity as-say kit, ICAM-1 in supernatants was assayed by using ELISA method, phosphorylation of NF-κB p65 was de-tected by western blot analysis, transcriptional activity of NF-κB signal pathway was measured by employing dual-luciferase reporter assay system. Results Hu-man microvascular endothelial cells were activated and injured by ox-LDL. Inhibition of the cell viability, re-lease of LDH, expression of ICAM-1, phosphorylation of NF-κB p65 , and up-regulated transcriptional activity of NF-κB induced by ox-LDL were attenuated by ATV. Conclusion ATV can significantly inhibit the activa-tion and injury of human microvascular endothelial cells induced by ox-LDL, and that may be related to inhibition of phosphorylation and transcriptional activity of NF-κB.

10.Isolation and Identification of Coumarin in Dendrobium denneanum Cultivated in Sichuan and Its Quality Standard Study

OBJECTIVE:To establish the quality creteria of Dendrobium denneanum cultivated in Sichuan.METHODS: The reference substance had been isolated and identified.HPLC and TLC were applied for determination and identification respectively.RESULTS: The coumarin control had been successfully isolated and the TLC spots were clear.The linear range of coumarin was 0.096~0.480 ?g with an average recovery of 99.1%(RSD=1.44%,n=6).CONCLUSION: The results can overall reflect the internal quality of D.denneanum cultivated in Sichuan and provide basis for quality evaluation and development of D.denneanum from Sichuan.
