1.Studies on Content and HPCE Fingerprints of Polysaccharides from the Seed of T.Jackii

OBJECTIVE:To determine the content and establish HPCE fingerprints of polysaccharides of the T.jackii seed.METHODS:The content of polysaccharides of the T.jackii seed was determined by phenol-vitriolic colorimetry and the fingerprints of polysaccharides were established by HPCE.RESULTS:The content percentage of polysaccharides in the seed of T.jackii was6.56%,with average recovery rate at98.3%and RSD at2.31(n=5).There were two kinds of polysaccharides in the seed of T.jackii.CONCLUSION:There is no significant difference with regard to the contents and components of polysacc_ harides in the T.jackii seeds from different origins.

OBJECTIVE:To study the active fraction of the seeds of Hovenia Dulcis Thunb.to treat acute alcoholism.METHODES:The seeds of Hovenia Dulcis Thunb.were divided into five parts:superitical fluid extraction(SFE)part,chloroform extraction part,ethyl acetate extraction part,n-butanol extraction part and water extraction part.The effects of the active fraction of the seeds of Hovenia Dulcis Thunb.on ebriety latency,ebriety rate,mortality,and ethanol concentration-time curve of the alcoholism mice were investigated.RESULTS:The ethyl acetate extract could prolong the ebriety latency of the mice,decrease ebriety rate and mortality of the alcoholism mice,and significantly decrease serum ethanol concentrations at 0.5,1,and 1.5h after alcohol administration(P
3.Comparison of the clinical effect of breast-conserving surgery and modified radical mastectomy in the treatment of early breast cancer

Objective To compare the clinical efficacy of breast-conserving surgery and modified radical mastectomy for early breast cancer and their influence on quality of life.Methods 78 patients with early stage breast cancer were divided into two groups by random number table,and 39 cases in each group.The control group was given modified radical mastectomy,while the observation group was given breast-conserving surgery.The clinical efficacy,surgical conditions and quality of life scores were compared between the two groups.Results The operative time,blood loss,incision length and the average hospital stay of the observation group were (60.23 ± 5.21)min,(44.73 ± 4.22) mL,(4.72 ± 1.02) cm and (9.52 ± 0.86) d,which were significantly lower than those of the control group(t =7.88,8.32,8.89,6.42,all P < 0.05).The postoperative physical factors,mental factors,psychological factors and social factors scores of the observation group were (67.44 ± 3.87),(48.64 ± 3.73),(117.34 ± 8.33)and (61.56 ± 5.32),which were significantly higher than those of the control group (t =5.43,5.87,6.29,7.11,all P < 0.05).The postoperative 3-year local recurrence,distant metastasis and survival rate showed no significant differences between the two groups (x2 =1.22,1.09,1.29,all P > 0.05).Conclusion Breast-conserving surgery in the treatment of early breast cancer has significant effect,and it has less trauma,less bleeding and cosmetic results,and it can be used as the preferred method for the treatment of early breast cancer,which is worth to be further applied in clinical.
4.Influence of Anxiety and Depression in Digestive system Cancer Patients' Prognosis

Objective:To study the effect of anxiety and depression in 46 digestive system cancer patients' prognosis.Methods:The differences of SDS mark?SAS mark and ≥ 50 mark percentage were compared between patients and common mould in nationwide adopting SDS and SAS,moreover the prognosis was compared between varying degrees of anxiety and depression in patients.Results:The SDS mark?SAS mark and ≥ 50 mark percentage in patients were obviously higher than that in common mould in nationwide;the patients with anxiety and depression had bad prognosis.Conclusion:The results suggest that anxiety and deprssion is common feeling in digestive system cancer patients and seriously influence the patients' prognosis,psychotherapy must be conducted and it may contribute to improve the patient's prognosis.
5.On the correlation between different stages of non-small cell lung cancer with Treg cells and cytokines

Objective To investigate the expression of CD4+CDhi25CDlow127 regulatory T cells (Treg cell), IL-2, IL-10 and IFN-γin peripheral blood of patients with different TNM stages of non-small cell lung cancer( NSCLC) , and to evaluate their correlation. Methods The proportion of CD4+CDhi25 CDlow127 Treg cells to CD4+cell in peripheral blood of 55 NSCLC patients were determined by flow cytometry, and levels of IL-2, IL-10 and IFN-γ were measured by ELISA. The proportion of CD4+CDhi25 CDlow127 Treg cells, IL-2, IL-10 and IFN-γ of patients with different TNM stages of NSCLC were compared, and correlation analysis was carried on. Results The proportion of CD4+CDhi25 CDlow127 Treg cells increased obviously together with the increasing of TNM stages of NSCLC(P<0. 05),so did the level of IL-10 (P<0. 05). On the other hand, the level of IL-2 and IFN-γdecreased apparently with the increasing of TNM sta-ges of NSCLC(P<0. 05). CD4+CDhi25CDlow127Treg cells and IL-10 were positively correlated(P <0. 05). IL-2 and IFN-γ were positively correlated too(P<0. 05). CD4+CDhi25CDlow127Treg cells and IL-10 were all negatively correlated with IL-2 and IFN-γ(P<0. 05). Conclusion The increasing of CD4+ CDhi25CDlow127Treg cells and IL-10 and the de-creasing of IL-2 and IFN-γ are possibly one of the reasons of antitumor immunity suppression in terminal NSCLC patients. The surveillance for them can be reference indicators to evaluate antitumor immunity status of terminal lung cancer patients.
6.The influence of early mechanical ventilation and perfluorocarbons gasification inhalation on inflammatory factor levels in patients with severe pancreatitis

Objective To investigate the clinical effect by early mechanical ventilation and perfluorocarbons gasification inhala-tion on patients with severe acute pancreatitis (SAP) complicated with acute respiratory distress syndrome (ARDS) .Methods 34 cases of SAP and ARDS patients accepted early mechanical ventilation and vaporization of perfluorocarbon inhalation therapy (ob-servation group) ,and 34 patients received mechanical ventilation for early treatment alone (control group) ,The efficacy ,oxygenation index(PaO2/FiO2 ) and inflammatory cytokines IL-1 ,6 ,8 ,and TNF-a level of two groups were compared .Results The total effi-ciency of the observation group was 94 .12% ,which was significantly higher than the control group that was 76 .47% (P<0 .05) . After treatment ,the arterial oxygen saturation (SaO2 ) ,arterial oxygen pressure (PaO2 ) ,PaO2/FiO2 of the observation group in-creased significantly ,and higher than the control group(P<0 .05) .After treatment ,IL-1 ,6 ,8 ,TNF-αof the observation group were significantly lower than the control group(P<0 .05) .Conclusion The treatment of early mechanical ventilation with perfluorocar-bon inhalation gasification could control the inflammation in patients with SAP complicated with AROS ,which could improve the cure rate .
7.lnhibition effects of Bevacizumab on migration of human Tenon capsule fibroblasts

Abstract? AlM: To investigate the inhibition effect of bevacizumab on human Tenon capsule fibroblasts ( HTFs ) and discuss the countermeasures of bleb scarringscarring in glaucoma surgery countermeasures.? METHODS: Adopted cell recovery method and followed the aseptic principles, we performed the culture of HTFs which came from the Central Laboratory of Shaanxi People's Hospital cell library. Wound Healing assay:We scraped a cell-free zone on the cell surface when the cells reached confluence at 80%. The control group was added to serum-free DMEM medium. The HTFs of bevacizumab group were stimulated with 1mg/mL concentrations without DEME for 0, 24, 48, and 72h. The scratch width was observed and measured.? RESULTS: HTFs were long fusiform shape under microscope, the nucleus is in the center of the cell with larger nucleus, abundant cytoplasm, were arranged in a whorled growth out of shape, strong ability to proliferate, conform to general forms of fibroblast. The morphological and biological characteristics of cells after cryopreservation and resuscitation remain unchanged. Wound healing assay: 0h, equal to the initial width of the two groups, 24h when the migration distance of the two groups of cells are basically the same, 48h when the control group cell migration distance is greater than that in the bevacizumab processing group, 72h when the control group scratches basichealing, bevacizumab treated cells migrate closer than 48h no significant change, and a lot of cells died.?CONCLUSlON:Cell recovery method can successfully cultured HTFs, which was stability on morphology and biological properties, laying the cellular basis for experimental research. Fibroblast itself has a strong ability to migrate, outside - derived bevacizumab can inhibit HTFs migration evidently and it will cause excessive cell death when. Bevacizumab has certain extent inhibitory effect on HTFs migration, and it is likely to become one of the important drugs for creating bleb scarring after glaucoma surgery in the future.
8.Promoting effect of connective tissue growth factor on expression of E-cadherin in human Tenon capsule fibroblasts

Background Scarring of filtration channel following glaucoma filtering surgery is a main cause of the failure of the surgery.The proliferation,epithelial-mesenchymal transition and extracellular matrix remodeling of fibroblasts are thought to be the primary pathological mechanism of scarring.Connective tissue growth factor (CTGF) plays a promoting role in the formation of scar.Whether CTGF participates in mesenchymal-epithelial transition of human Tenon capsule fibroblasts (HTFs) is not clear yet.Objective This study attempted to investigate the effect of CTGF on the mesenchymal-epithelial transition of HTFs in vitro.Methods HTFs were cultured and passaged in high glucose DMEM medium with 10% fetal bovine serum,and the cells of generation 3-6 were used in this experiment.The cells were divided into the blank control group and CTGF-treated group and were routinely cultured in the blank control group.CTGF was added in the medium in the CTGF-treated group,with the final concentration 50 ng/ml CTGF.Immunofluorescence staining was used to identify and locate the expression of E-cadherin protein in the cells,and Western blot assay was employed to quantitatively analyze the expression level of E-cadherin protein in 48 hours after culture.Results The HTFs grew well with the spindle-like shape and vortex-like arrangement.The red fluorescence (E-cadherin protein) in the cytoplasm and blue fluorescence in the cellular nucleus were seen in the CTGF-treated group,but only nucleus with blue fluorescence were obtained in the blank control group.Western blot assay showed that the E-cadherin protein expression was absent in the blank control group,however,the relative expression level of E-cadherin protein in the cells was 0.63± 0.08.Conclusions E-cadherin protein is not expressed in fibroblast derived from mesenchymal tissue.However,CTGF can induce the expression of E-cadherin in HTFs.This study suggests that CTGF promotes the mesenchymal-epithelial transition of HTFs in vitro.

ⅡB. Our investigation also showed that the activity of PAS, LDH and PFK was stronger in type Ⅱ fibers than in type Ⅰ. It was concluded that each type fibers in the rat plantaris exhibit significant differences in metabolic and functional activity. The type ⅡA fibers present both high activity of glycolytie enzyme and oxidase, and reflect that these enzymes may play very important role in energy metabolism. Further study remains to bedertaken.
10.STUDIES ON THE RELATION BETWEEN INHIBITORY EFFECT OF MYELIN MEMBRANE PROTEINS ON NEURITE OUTGROWTH AND cAMP LEVELS IN NEURONS

Objective To study the relationship between the inhibitory effects that myelin membrane proteins exerted on nerve regeneration and the changes of intracellular cAMP levels in neurons. Methods To observe the effects of myelin membrane proteins extracted from central nervous system(CNS) by density centrifugation on the outgrowth of neurite of cerebella granule cells in culture and detect the changes of intracellular cAMP levels of the neurons with radioimmunoassay. Results 1\^Myelin membrane proteins of CNS inhibited neurite outgrowth of cultured cerebella granule cells. 2\^The cAMP level in neurons decreased in 5 minutes after contacting myelin membrane proteins and reached to the lowest level in 12 hours of contacting. Conclusion The inhibitory effects of myelin membrane proteins on the outgrowth of neurites may be related to the inhibitory factors which cause the decrease of cAMP level in neuron through the passway of signal transduction pathway.
