3.Therapeutic strategy for retinoblastoma in China

Retinoblastoma (RB) is the most common intraocular malignant tumor in children. With advanced clinical technologies there are more and more methods available to treat retinoblastoma, and make it is possible to delivery individualized protocol combined traditional treatments with modern regimen to patients now. In order to improve the survival rate and the life quality of RB patients in China, it is very important to make a suitable system of standardized therapy based on results from developed countries and health policies of our own country.
4.Research progress of ultrasonography and computed tomography in the preoperative assessment of cervical lymph nodes metastases of esophageal cancer

Esophageal cancer is one of the most common malignant tumors in China.At present, the best treatment of esophageal cancer is surgery.For the patients with cervical lymph nodes metastases, three-field lymph nodes dissection is the main solution.Ultrasonography and computed tomography (CT) of the neck play a decisive role in the preoperative examinations for patients with esophageal cancer, and should be used as routine examinations before surgery at the qualified hospital in order to improve the accuracy of preoperative assessment of cervical lymph nodes metastases of esophageal cancer and screen potential patients with esophageal cancer undergoing three-field lymph nodes dissection.
5.Ⅰ-stage resection of colon cancer and synchronous liver metastasis after conversion therapy

Objective To investigate the safety and clinical effect of Ⅰ-stage resection of colon cancer and synchronous liver metastasis after conversion therapy.Methods The retrospective descriptive study was adopted.The clinical data of 1 patient with sigmoid cancer with synchronous multiple liver metastases who was admitted to the Zhongshan Hospital of Fudan University in April 2013 were collected.The patient was diagnosed as sigmoid cancer with multiple lymph nodes surrounding colon and 5 metastatic lesions found in liver by preoperative imaging examination,and the maximum diameters of 2 metastatic lesions were 4.5 cm and 3.6 cm.The pathological results of colonoscopy indicated adenocarcinoma.After discussion of the multidisciplinary team,liver metastatic lesions were significantly reduced through mFOLFOX6 chemotherapy combined with bevacizumab treatment,and then patient underwent Ⅰ-stage resection of colon cancer with synchronous liver metastasis by the surgical procedures of partial hepatectomy + 3D laparoscopy-assisted radical resection of sigmoid cancer.The patient received convention treatment of antibiotic,nutrition support therapy and mFOLFOX6 adjuvant chemotherapy.The changes of lesions in liver and tumor markers were observed by tumor marker test,abdominal/pelvic CT and MRI in the upper abdomen after conversion therapy.The operation time,volume of intraoptrative blood loss,number of lymph nodes dissected and vital sign were observed in the operation.The liver function,time for out-off-bed activity,time to anal exsufflation,time of drainage tube removal,duration of hospital stay,complications,results of pathological examination and recurrence and mnetastasis of tumor were observed after operation.After discharge from hospital,the patient underwent monthly tumor marker test,B ultrasound and chest X-ray examination till January 2014 and abdominaL/pelvic CT,magnetic resonance imaging (MRI) in the upper abdomen and colonoscopy every 6 months.The follow-up of outpatient examination and telephone interview was performed to detect the recurrence and metastasis of tumor up to September 2015.Results After preoperative 4-cycle chemotherapy,the diameters of 2 metastatic lesions in liver reduced to 3.2 cm and 2.0 cm,the levels of carcinoembryonic antigen(CEA) and CA19-9 (tumor marker) were reduced to 95.9 μg/L and 252.4 kU/L.The patient underwent successful Ⅰ-stage resection of colon cancer and synohronous liver metastasis.The operation time,volume of intraoperative blood loss,numbers of lymph nodes dissected and cancerous nodes were 208 minutes,250mL,14 and 1,respectively.The patient had no blood transfusion and metastasis,with the stable vital signs and good postoperative recovery.The levels of alanine transaminase (ALT) and aspartate transaminase (AST) were 1 147 U/L and 2 631 U/L at postoperative day 1,101 U/L and 37 U/L at postoperative day 7,respectively.The out-off-bed activity,anal exsufflation,drainage tube removal and discharge from hospital were occurred at postoperative day 2,3,7 and 9,respectively.No anastomotic fistula,bleeding and infection were occurred aftcr operation.The patient was diagnosed as with ulcerated sigmoid adenocarcinoma in T3N1cM1 stage (combining with liver metastasis).The patient received adjuvant therapy of mFOLFOX6 and oral capecitabine at postoperative day 40,and was followed up for 30 months with a good quality of life and without metastasis and recurrence.Conclusion Ⅰ-stage resection of colon cancer with synohronous liver metastasis can be used for initial unresectable colon cancer with liver metastasis after conversion therapy,and it is safe and feasible,with a good therapeutic effect.
6.Na/Ca exchange plays a role in calcium homeostasis and calcium-induced calcium release of cardiac myocytes

ABSTRACT Calcium homeostasis is of crucial importance for the function of cardial myocytes in normal electrical and mechanical processes. The sarcoplasmic reticular Ca-ATPase and the sar-colemmic Na/Ca exchanger contribute mainly to the steady state of calcium in the myoplasm. The Na/Ca exchanger serves as the principal calcium extrusion mechanism and regulates calcium content of the sarcoplasmic reticulum by regulating the resting [Ca2+]i level, through which the Na/Ca exchanger regulates the force of contraction sequentially. Very small calcium entry can trigger significant sarcoplasmic reticular calcium release (CICR). During the upstroke and plateau phasesof the action potential, There is evidence that indicates Ca2+ influx via the Na/Ca exchanger, and the depolarisation-induced calcium entry on Na/Ca exchange may contribute mainly to triggering intra-cellular calcium release. Na/Ca exchange should be reevaluated as a route through which Ca2+, as a triggering signal, enters cardiac myocyte during excitation-contraction coupling.
7.Isolation of CD44~+/CD24~(-/low) and side population cells from MDA-MB-453 cells and the analysis of their activation of Wnt and Notch pathway

Objective:To explore stem cells phenotype of human epidermal growth factor receptor 2(HER2)-overexpressing breast cancer cells,MDA-MB-453 by isolation of CD44+CD24-/low and side population(SP).Analyze the relationship between SP cells and the gene amplification levels of the key proteins,?-catenin and Notch-1 in Wnt and Notch signal pathways.And study the correlation of SP cells and the gene amplification of HER2 and human epidermal growth factor receptor 3(HER3).Methods:CD44+CD24-/low and SP phenotype were studied in MDA-MB-453(HER2-overexpressing cell line)and MCF7(HER2 negative cell line)respectively by flow cytometry.The gene amplification of ?-catenin and Notch-1 in Wnt and Notch pathways were also studied by reverse transcriptase polymerase chain reaction(RT-PCR).The gene amplification levels of the proteins mentioned,HER2 and HER3 were roughly compared in MDA-MB-453 cells and their SP by RT-PCR.Results:There was no CD44+CD24-/low subpopulation in MDA-MB-453 cells,However,SP could be observed in MDA-MB-453.The gene amplification levels of ?-catenin and Notch-1 in Wnt and Notch pathways were both observed in MDA-MB-453 and MCF7.Their gene amplification levels were more evident in SP of MDA-MB-453 than in MDA-MB-453 cells.However,the similar gene amplification levels of HER2 and HER3 were observed.Conclusion:SP can be enriched in breast cancer stem cells of MDA-MB-453.Wnt and Notch pathways are activated in MDA-MB-453,which may be related to maintenance and activity of breast cancer stem cells of MDA-MB-453.
8.Effects of different ways of administration on the P2X_1 purinoceptor-mediated vasoconstriction in the rat mesenteric artery

Aim To investigate the effects of ?,?-methylene ATP ( ?,?-MeATP) on the P2X1 purino-ceptor-mediated vasoconstriction by different administration ways in the rat mesenteric artery. Methods Isometric vasoconstrictive responses to ?,?-MeATP,administered in non-accumulative manner or single concentration manner,were recorded in the rat isolated mesenteric arterial rings. Results ?,?-MeATP ( 10 -7 ~ 10 -4 mol ? L -1 ) administered in both of the two manners produced concentration-dependent vasocon-strictive responses in the rat isolated mesenteric artery. The vasoconstrictive responses to ?,?-MeATP in single-concentration administration group were greater than those in non-accumulative administration group when the vasoconstriction was standardized either by tissue wet weight or by maximal response to 120 mmol ?L-1 KCl ( P
9.THE CONNECTIONS OF THE VISUAL PORTION OF THE THALAMIC RETICULAR NUCLEUS IN GOLDEN HAMSTERS

The thalamic reticular nucleus (RT) is a sheet-like nucleus which surrounds the anterolateral aspect of the dorsal thalamus. In this study, we present data on the connections of the dorsocaudal portion of the RT in hamsters using anterograde, retrograde and transneuroual fiber tracing techniques. After injection of one eye with a mixture of ~3H-proline and ~3H-fucose (2 animals; 10 and 15 days survival time), transneuronal label was located in the dorsocaudal portion of the RT (dc RT) on both sides of the thalamus.After separate injection of horseradish peroxidase (HRP) into the thalamic lateral posterior nucleus (LP), dorsal and ventral nuclei of the lateral geniculate body (LGd, LGv) (8 animals, one day survival time), HRP labelled neurons wereobserved in the dorsal, middle and ventral area of the ipsilateral dc RT respectively.A small iontophoretic injection of a mixture of ~3H-proline and ~3H-leucine was made into different sites of area 17 in each of 3 hamsters (one day survival time),in each case an elongated patch of label was located in nearly the entire rostrocaudal extent of the ipsilateral dc RT. A caudal Injection in area 17 resulted in labelling the dorsal area while a progressively more rostral injection resulted in labelling a more ventral area of the dc RT.Thus, the dorsocaudal portion of the RT can be defined as the visual portion of the nucleus because it has connections with many of the known visual centres, namely, the visual cortex, LP, LGd, LGv Furthermore, because of the topographic connection with area 17 and the visual thalamic nuclei, the notion that the dc RT as a nucleus which is capable of processing specific and localised visual information. should be considered.
10.Effects of Lead Exposure on Study, Memory and Autonomic Activities of Mice

Objective To study the neurotoxicity of lead to mice and the mechanism. Methods Two months old(grown) and two weeks old(infancy) mice were employed, 1 ?l of 1 mol/L lead acetate was given by brain ventricle injection, 3 d, 5 d, 1 week, 2 weeks and 3 weeks after treatment with lead, the change of autonomic activities and the ability of study and memory of the mice were examined. Results The latency period of Lashley Ⅲ water maze prolonged significantly in grown group and the infancy group compared with the control groups at every time point of test. Conclusion Lead exposure can damage the function of study and memory of mice, and increase the frequency of autonomic activities of theirs.
