1.Development and aging of C57/BL6 mouse hippocampus

Objective To investigate the morphological changes during the development and aging of C57/BL6 mouse hippocampus.Methods Serial sections and stereology were used to quantitatively analyze the development of C57/BL6 mouse hippocampus at different growth stages.Results Hippocampus primordium first appeared at embryonic day 12(E12d).A "C" outline could be seen in the pyramidal layer of Ammon's horn(CA),and the extra-arm's blanket of the dentate gyrus(DG)'s granular layer formed at E18d.After birth,CA developed and maturated gradually.The blanket of DG's granular layer formed at postnatal day 7(P7d).At P21d,the inner arm's thickness of DG's granular layer was equal to that of the extra-arm,and the subgranular layer was present until month 15(15M).The increase of the volume of hippocampus,CA,DG and CA's layers was slow before P7,became fast from P7d to P14d and slowed down again after P14d,but became stable after 3M.Conclusion Mouse hippocampus is formed at E12 and becomes basically mature at month 3.The volume of aging mouse hippocampus has no obvious changes.
3.Protective effect of allitridi on hippocampus of rats with cerebral ischemia-reperfusion and P53 expression

AIM:To observe the protective effect of allitridi on hippocampal neuron of rats with cerebral ischemia-reperfusion (I/R) injury and to investigate its effects on P53 expression in hippocampus.METHODS: The global cerebral ischemia-reperfusion models were established by 4-vessel occlusion. Allitridi at doses of 10, 20 or 30 mg/kg was injected through rat’s tail vein, half dose at 30 min before brain ischemia and another half dose at 10 min after reperfusion were injected, respectively. The hippocampus of rat was removed 24 h after reperfusion. Toluidine blue staining was applied to estimate morphologic changes. Flow cytometry was used to evaluate neuronal apoptosis rate of hippocampus. Immunohistochemistry was used to observe the expression of P53 protein.RESULTS: Compared with sham group, survival neuronal density in I/R group was significantly depressed. The rate of neuronal apoptosis and the expression of P53 protein were significantly increased. Allitridi significantly increased the number of survival neurons in hippocampus compared to I/R group. Meanwhile, allitridi remarkably inhibited the rate of neuronal apoptosis and the expression of P53 protein.CONCLUSION: Allitridi has protective role against brain ischemia reperfusion injury. The mechanism may be involved in blocking P53 protein expression in hippocampus of rats with ischemia-reperfusion.
4.Curative effect observation of premature ovarian failure treated by oral taking traditional Chinese medicine with functions of tonifying kidney, nourishing blood and soothing liver and external application

Objective To evaluate the clinical efficacy of oral taking traditional Chinese medicine with functions of tonifying kidney, nourishing blood and soothing liver and external application treatment of premature ovarian failure(POF). Methods 40 patients of deficiency of liver-yin and kidney-yin type POF in Gynecology Department of Xiyuan Hospital CACMS from July 2009 to July 2013 were collected and administrated with traditional Chinese medicine with functions of tonifying kidney, nourishing blood and soothing liver and external treatment for 3 months. The changes of menstruation situation, symptoms and serum sex hormone were compared before and after the treatment. Results ① Menstruation integral were reduced after the treatment, showing a statistical significance compared with those before the treatment(t=5.694,P<0.01). ② Symptoms integral were reduced after the treatment, showing a statistical significance compared with those before the treatment(t=21.828,P<0.01). ③ Serum FSH, LH were decreased and Serum E2 were increased after the treatment,showing a statistical significance compared with those before the treatment(P<0.05). ④ The total effective rate was 92.5%(37/40). Conclusion Traditional Chinese medicine with functions of tonifying kidney, nourishing blood and soothing liver are effective for treating POF. These methods had proven to improve ovarian function by decreasing high level of FSH and LH, whereas enhancing the estrogen level, and therefore promote menstruation cycle.
5.Pharmacodynamic studies on cardioprotective function of Zhurihen dripping pills in experimental animals

Objective To study the cardioprotective function of Zhurihen dripping pills in experimental animals.Methods By hypoxia tolerance model and isoproterenol hydrochloride induced acute hypoxia model, measuring the time of death in mice.For rat serum LDH, CK, SOD and MDA were measured to observe protective effect of Zhurihen dripping pills on vasopressin-induced acute myocardial ischemia model.The effect of Zhurihen dripping pills on latency and durations were observed in acute arrhythmia induced by inhaled chloroform – epinephrine rabbits model.Results The hypoxia tolerance in mice and myocardial hypoxia tolerance significantly improved by Zhurihen dripping pill, and prolonged the survival time in mice.The serum level of LDH, CK, SOD and MDA significantly improved, and protected the impaired myocardial cell in rats.The latent period of arrhythmia was significantly prolonged and duration of arrhythmia was shortened in rabbits.Conclusion Zhurihen dropping pills an improve myocardial oxygen deficiency, anti-arrhythmia effect and protect myocardial cells.

Neonatal intrahepatic cholestasis caused by citrin deficiency (NICCD) is an autosomal recessive disease caused by SLC25A13 gene mutations, and is characterized by delayed jaundice clearance, liver dysfunction, and elevated aminoacidemia. The confirmed diagnosis depends on gene analysis. Citrin deficiency is one of the important causes of neonatal intrahepatic cholestasis in China. Recently more and more researches about NICCD were reported. The paper summarized the epidemiology, pathogenesis, clinical characteristics, and progresses in diagnosis and treatment of NICCD.
7.Effectiveness of iterative metal artifact reduction for reduction of metal artifact in chest CT scanning

Objective To assess the effectiveness of iterative metal artifact reduction (IMAR) on metal artifacts reduction in thorax scan.Methods Thoracic phantom with two pedicle screws implanted in both sides of the T5 vertebrae was used,with the scan parameters of 130 kV and CARE Dose 4D,the phantom was scanned with and without the screws respectively.Images without screws were reconstructed with FBP.Images with screws were reconstructed with FBP and IMAR respectively.Three ROIs were selected on tissues including aorta,pulmonary and paravertebral soft tissue on image slice adjacent to the screws.The CT value and standard deviations (noise) of ROIs were measured,and the deviation of CT value (△HU) was calculated as the difference between CT values in images with and without screws.Twenty-six cases who received chest CT examination and with pedicle screw implant in scanning range were collected.The scanning parameters and image reconstruction methods were the same as phantom scan.The CT value (HU) of metal artifacts adjacent to vertebrae and dorsal soft tissue was measured,and the image quality of reconstructed image by two skilled radiologists independently was evaluated.Results In the phantom after implanted screws,the noise were significantly reduced by IMAR compared to FBP in all the three ROIs of aorta,pulmonary and paravertebral soft tissue (P<0.05),and the △HU was significantly smaller in IMAR compared to that with FBP (P<0.01).In 26 patients,there were significant differences in CT value of vertebral bone tissue and dorsal soft tissue between FBP and IMAR (P<0.05),and the subjective evaluation scores of the two image reconstruction methods showed a statistically significant difference (P<0.05).Conclusion IMAR can significantly reduce streak artifacts of metal implant and adjuste the CT values of artifact affected tissues to make it more close to the true value without metal implant.
8.Medicinal experience, toxicity and efficacy of five types of Yunnan ethnic medicine

The five volumes of Annals of National Medicine in Yunnan contain more than 1040 kinds of ethnic medicines and national medicines experiene that are commonly used by 25 minority nationalities in Yunnan.Among them,there are 147 species of toxic drugs,including five with fatal toxicity,14 with strong toxicity,57 with toxicity,and 71 with mild toxicity.In order to guarantee the validity and safety of national drug use,Yunnan Institute of Materia Medica applied modern science and technology to study the safety,efficacy and pharmacognosy of 30 kinds of commonly used toxic folk drugs contained in Annals of National Medicine in Yunnan.This paper chooses five typical national medicinal herbs Stephania delavayi,Euphorbia neriifolia,Alocasia cucullata,Achillea wilsoniana and Chamaenerion angustifolium,and summarizes their national medicinal and toxicity experience.The results of acute toxicity test showed that the five national medicinal herbs are toxic.The pharmacodynamic experiments showed that S.delavayi has analgesic action,E.neriifolia has anti-inflammatory action,A.cucullata has analgesic and antitussive effects,Ach.wilsoniana has analgesic effect,and C.angustifolium has analgesic and anti-inflammatory effects.
9.STUDY ON ULTRASTRUCTURE, RAS ONCOGENES AND CHANGES IN MUCUS OF COLON CARCINOMA

The characteristics of ductal adenocarcinoma of colon under eieciror: microscopy were stout and dense microvill, with small roots buried in the top of cytoplasm. The ullrarnorphological changes of carcinomatous infiltration was similar to the picture observed in vitro cultivation of cancer cells.In 104 cases out of 159, there was an over expression of ras oncogenes P21, indicating that carcinoma of colon was related to activation of ras oncogenes. Examination with monoclonal antibodies yielded a positive rate of 83.4%- 95.7%, indicating that most of the carcinomas had colon related antigens. This phenomenon should play an important role in raising the diagnostic accuracy, and it also should serve as an indicator of effective treatment. In 62 cases of mucoid adenocarcinoma and 30 cases of signet ring cell carcinoma, there was a marked reduction of mueoitin sulfate with an increase in sialic acid mucus. The letter was a glycoprotein containing hydroxyl mucus, and it facilitated the separation of cancer cells to infiltrate and metastasize.in 134 cases, AgNOR was quantified, and there were 13.05? 1.48 granules in each mucleas. There was more abundant AgNOR in undifferentiated cancer, while the number of AgNOR fluctuated between 10.56 to 11.83 in papillary adenocarcinoma, ductal adenocarcinoma, and mucoid adenocarcinoma. indicating the qunantity of AgNOR was related to degree of malignancy in colonic carcinoma.
10.Human bone morphogenetic protein 3(hBMP3) inhibits growth of human osteosarcoma cells MG63 and U2OS

Objective The biological effects of BMP3 on osteosarcoma were investigated by treating human osteosarcoma cell lines MG63,and U2OS with human BMP3(hBMP3).Methods Osteosarcoma cells in experimental groups were respectively treated with AdBMP-3 and rhBMP3-CM,control groups with AdGFP and rGFP-CM,the blank group with neither.Their ability of proliferation,apoptosis,transmigration and differentiation were respectively detected by trypan blue exclusion test,terminal deoynucleotidyl transferase mediated dUTP nick end labeling(TUNEL) and acridine orange-ethidium bromide fluorescent stain(AO/EB),transwell-room and alkaline phosphatase(ALP) activity reagent kit.Results(1) All the indexes detected were not significantly different between two control groups.(2) Compared with control groups,the cell survival rate showed a significant decrease in experimental groups.(3) The apoptosis indexes increased.(4)The number of trans-membrane cell decreased.(5)The activity of alkaline phosphatase increased after treatment with AdBMP3 and rhBMP-3 for 3 days in MG63,5 days in U2OS.Conclusion hBMP3 inhibit osteosarcoma cells growth and promote bone formation.
