1.STUDY ON THE LEVELS OF HOMOCYSTEININ AND NEUTROPHIL GELATINASE ASSOCI-ATED LIPOCALIN IN PATIENTS WITH CHRONIC HEART FAILURE COMBINED INSUFFI-CIENT RENAL FUNCTION

Objective To investigate the levels of homocysteinin and neutrophil gelatinase associated lipocalin and its rela-tionship in patients with chronic heart failure combined insufficient renal function .Methods 58 patients with chronic heart failure were enrolled in from June 2013 to June 2014 in Beijiao hospital affiliated Southern medical university .Thereinto, according to pa-tient’s condition, 28 patients were chronic heart failure concurrence insufficient renal function, 30 patients were chronic heart failure. Left ventricular ejection fraction were determined by cardiac ultrasound , the levels of serum BNP, Hcy, NGAL and creatine were de-termined.Results Compared with chronic heart failure patients,the levels of Hcy and NGAL were increased in patients with chronic heart failure concurrence insufficient renal function (p<0.05).Correlation analysis showed that the levels of Hcy is positively corre-lated with the levels of NGAL (p<0.05);the levels of Hcy and NGAL were positively correlated with BNP and creatine (p<0.05), but negatively correlated with LVEF (p<0.05), respectively.Conclusion Circulating levels of Hcy and NGAL can reflect the de-velopment of chronic heart failure concurrence insufficient renal function .
2.CORRELATIONAL STUDY BETWEEN HOMOCYSTEININ AND CORONARY ATHEROSCLE-ROTIC DISEASE ON THE CORONARY ARTERY DISEASE

Objective To investigate correlational study on the levels of homocysteinin with the severity of coronary artery in patients with coronary atherosclerotic disease.Methods 60 patients with coronary atherosclerotic disease were enrolled in from Janu-ary 2013 to December 2014 in Beijiao hospital affiliated Southern medical university .All patients received coronary artery angiograph and the levels of circulating homocysteinin were determined.According to the levels of circulating homocysteinin, 60 patients were de-vided into coronary atherosclerotic disease group and coronary atherosclerotic disease group with hyperhomocysteinemia ( HHcy) .The levels of homocysteinin and the severity of coronary artery in patients with coronary atherosclerotic disease and correlation between the two group were analyzed.Results Compared with coronary atherosclerotic disease patients,the levels of homocysteinemia and Genisi score were increased in patients with coronary atherosclerotic disease with HHcy (p<0.05).Correlation analysis showed that the lev-els of homocystein is positively correlated with Genisi score (p<0.05).Conclusion HHcy is a important risk for coronary athero-sclerotic disease and positively related with the severity of coronary artery in patients with coronary atherosclerotic disease .
3.Research progress of molecular mechanism on miR-29 family for regulating the formation, development and metastasis of breast cancer

MicroRNAs (miRNAs), a non-encoding small molecular RNA, is entitled to regulate gene expression after transcription, which plays a key role in the progress of tumorigenesis, growth and metastasis. MiR-29 family consists in miR-29a, miR-29b and miR-29c, all featured by the regulation of breast cancer. This review will summarize the regulation and molecular mechanism of miR-29 family in all critical stages of breast cancer, and evaluate the value of miR-29 as a new diagnostic and prognostic biomarker or therapeutic target.
4.Analysis on clinical application of full-automatic erythrocyte sedimentation rate analyzer in ditecting EDTA-anticoagulated blood

Objective To compare the efficacies of automatic erythrocyte sedimentation rate (ESR) analyzer using EDTA as blood anticoagulant ,traditional Westergren method and ESR analyzer using sodium citrate as blood anticoagulant in detecting ESR , in order to chose the appropriate one for clinical application .Methods A total of 40 samples were collected and divided into 3 groups .The ESR was detected by using XC‐A30 automatic ESR analyzer using EDTA as blood anticoagulant (XC ESR analyzer) , Westergren method and VISION‐C automatic ESR analyzer using sodium citrate as blood anticoagulant (VISION ESR analyzer) ,re‐spectively .Statistical analysis was used to comparing the detection results of ESR among the three methods .The repeatability test was carried out for evaluating repeatability of VISION ESR analyser .Results There was statistically significant difference in ESR detected by using Westergren ,XC ESR analyzer and VISION ESR analyzer(F=86 .497 ,P=0 .000) .There was statistically signifi‐cant difference in ESR between that detected by using XC ESR analyzer and that detected by using Westergren (t= -4 .08 ,P=0 .004) .The ESR detected by using VISION ESR analyzer was positively correlated with that detected by using Westergren (r=0 .975 ,P=0 .000) .There was no statistically significant difference in ESR between that detected by using VISION ESR analyzer and that detected by using Westergren(t=0 .65 ,P=0 .638) .The repetitive tests of analyzer shown that the within‐run coefficient of variation(CV) and between‐run CV both were less than 10% .Conclusion The ESR detected by using VISION ESR analyzer which is simple operation and with credible result has good correlation with that detected by using Westergren .The VISION ESR analyzer might be suitable for clinical application .
5.Advances in molecular-targeted therapies for gastric cancer and gastroesophageal junction adenocarcinoma

The prognosis of advanced gastric cancer and gastroesophageal junction (GEJ) adenocarcinoma is poor. Although che-motherapy prolongs patient survival and improves quality of life to a greater extent best supportive care compared to, the median over-survival of patients with advanced gastric cancer is limited to approximately 7-10 months. With remarkable progress in the understand-ing of molecular mechanisms, molecular-targeted agents have been developed and evaluated in international randomized phaseⅢclini-cal trials. These agents may change the treatment mode of this disease. A ToGA study initially demonstrated that the trastuzumab, the monoclonal antibody of HER-2, as a molecular-targeted agent, in combination with chemotherapy, can prolong the overall survival of patients to 13.8 months. Several agents targeting angiogenesis, c-Met, PARP, and immunotherapy are currently subjected to clinical tri-als. This review summarizes the current status of molecular-targeted therapies for gastric cancer and GEJ adenocarcinoma.
6.A case report of transverse colon desmoplastic small round cell tumor

Desmoplastic small round cell tumor( DSRCT) is a kind of highly uncommon malignant tumor which is reported in very recent years.Since it has been firstly reported by Gerald in 1991,DSRCT has only a-round one hundred reported cases globally, and very limited cases domestically.This report illustrates the first case of DSRCT in our hospital so as to share and to discuss with clinicians.
7.Construction of human Alzheimer's disease specific single chain antibodies library

Objective To construct the human Alzheimer's disease (AD) specific single chain anti‐bodies (scFv) library for sceening human AD scFv to Aβ1‐42 oligomers .Methods RNA was isola‐ted from 40 ml peripheral blood taken from 18 AD patients .Variable heavy (V H ) and variable light (VL ) genes were amplified by RT‐PCR and linked to the scFv fragments that were then cloned into the phage vector pCANTAB5E .The scFv library was constructed by electroporating E .coli TG1 cells into the pCANTAB5E and rescuing the assistant phage M13K07 .Results The total RNA was extracted .The VH and VL genes were amplified .Electropharesis showed that the length of VH and VL genes was 360bp and 300bp respectively and the length of linked scFv was 750bp .The 2 .4 × 109 scFv library was thus constructed and identified by BstN I digestion .Elec‐tropharesis showed that the length of BstN I‐digested scFv fragments varied ,indicating that the library is of a good diversity .Conclusion The human AD phage scFv library we constructed lays a foundation for screening the antibodies to Aβ1‐42 oligomers and the treatment of AD .
8.Identification of a novel human leucocyte antigen allele B*07:110

BACKGROUND:In recent years, with the development of China Marrow Donor Program and the improvement of human leukocyte antigen (HLA) typing technique, novel aleles of human leukocyte antigen have been discovered constantly in China. OBJECTIVE:To identify and confirm a novel HLA alele in a Chinese individual. METHODS: A new HLA alele was found during routine human leukocyte antigen genotyping by PCR-sequence specific oligonucleotide probes and sequencing-based typing. RESULTS AND CONCLUSION:The HLA-B locus hybridization probe reaction patterns of this sample did not match with any known HLA-B aleles or alelic combinations. Exons 2, 3 were sequenced in both directions using HLA-B sequence primer and group-specific sequencing primer. The obtained sequence had 2nts change from B*07:02:01 at nt 226 and nt 228 where A->G (codon 76 ATA->GTG) and resulting in a coding change, 76 isoleucine (I) was changed to valine (V). This nucleotide sequence has been submitted to the GenBank nucleotide sequence database and it is available under the accession number HM989017. A novel HLA alele, HLA-B*07:110, was identified, and was named officialy by the WHO Nomenclature Committee.
9.Counselor verbal response modes of psychic hot-line counseling

Objective:To explore the counselor verbal response modes of psychic hot-line counseling and its characteristics.Methods:Through a qualitative study with category analysis method,8 pieces of audio recordings were collected,in each of which the entire duration was between 30 minutes to 50 minutes.And all of these record-ings were transcribed word by word.Meaningful 1202 pieces were deeply analyzed.Results:There were 5 core counselor verbal response types in hot-line counseling,greeting response,set-up response,listening response,follow-up response,and affecting response,all of which had 24 concept categories.All of the online counselors used the 5 types of verbal response.The listening response and follow-up response were the highest frequent types.Greeting response and set-up response were used mostly in the initial stage of counseling.Listening response,and follow-up response were used mostly in the stage of describing problem.Affecting response was sharply increased in the stage of analyzing problem.Greeting response and set-up response were used mostly in last stage of counseling.Conclu-sion:It suggests that there are 5 core counselor verbal response types and 24 concept categories in hot-line counse-ling.The listening response and follow-up response are the highest frequent types.Different types of verbal re-sponse modes used by counselor in different stages due to different intention.
10.A Molecular-Imprinted Sensor for Trace Detection of Gibberellin Based on Ferrocenecarboxylic Acid Multiply Marked Dendrimer

A novel strategy employing dendrimer to multi-label the template molecule to improve the sensitivity of molecularly imprinted electrochemical sensor was proposed. The determination relies on a competition reaction between poly ( diethylene-triaminepenta-acetic acid )-glycol ester ( PDTPA )-ferrocene-carboxylic acid ( FcA ) labeled gibberellins acid 3 ( GA3 ) and GA3 in the sample. Since one cavity corresponds with multiple FcA, instead of only one FcA, the intensity of the detecting signal was greatly enhanced, so was the sensitivity of the sensor. Experimental results showed that the molecularly imprinted electrochemical sensor was sensitive to GA3 detection at a concentration from 2. 0í10-9 to 1. 0í10-7 mol/L, with a detection limit of 9. 3í10-10 mol/L. In addition, the sensor had good reproducibility and its feasibility was verified in the analysis of series of real beer samples.
