1.Evaluation of image quality of dynamic three-dimensional contrast-enhanced ultrasonography versus CECT/MRI in liver tumors

Objective To evaluate the image quality of dynamic three-dimensional contrast enhanced ultrasonography(3D-CEUS) in liver tumors.Methods Twenty-five patients were examed with 2D-CEUS and dynamic 3D-CEUS.Contrast-enhanced computed tomography or magnetic resonance imaging(CECT/MRI) were performed early or late.Two readers reviewed the images of both modalities and graded the images according to diagnostic criteria as follows:tumor blood fusion in arterial phase(unclear-score 0,moderate-score 1,clear-score 2),tumor boundry and relationship with periphery tissues (unclear score 0,moderate-score 1,clear-score 2).The concordance between the two modalities for showing liver tumors was evaluated by Kappa test.Results Twenty-three of 25 danymic 3D-CEUS were displayed successfully versus total objects in CECT/ MRI.Among them,for showing blood fusion,16 lesions scored 2,7 lesions scored 1,2 lesions scored 0 in danymic 3D-CEUS,meanwhile 18 lesions scored 2,7 lesions scored 1,0 lesion scored 0 in CECT/MRI,the Kappa value was 0.65.For showing tumor boundry and relationship with peripherytissues,16 lesions scored 2,7 lesions scored 1,2 lesions scored 0 in danymic 3D-CEUS,meanwhile 21 lesions scored 2,4 lesions scored 1,0 lesion scored 0 in CECT/MRI,the Kappa value was 0.52.Conclusions Danymic 3D-CEUS was shown to be useful for displaying tumor blood fusion,boundry and relationship with periphery tissues in single liver tuomor,the concordance between dynamic CEUS and CECT/MRI for single liver tuomor was fair.For huge liver tumor and multi-tumors,3D-CEUS can not give overall information.

4.Role of endoplasmic reticulum stress in high fattyacid induced injury in cardiomyocytes

Aim To explore the role of endoplasmic reticulum stress (EPR) in high fatty acid induced injury in cardiomyocytes.Methods Cardiomyocytes were exposed to different concentrations of palmitic acid (0, 0.1, 0.2, 0.4 mmol·L-1) for 24 h and different time points of high palmitic acid (0.2 mmol·L-1) (12, 24, 48 h).Cell viability was measured by CCK8, and the protein expressions of EPR such as GRP78, CHOP, PERKphos, IRE1phos, ATF6 and apoptosis related proteins such as Bcl-2 and Bax were determined by Western blot.Results When the cells were stimulated with palmitic acid (0.1~0.4 mmol·L-1) for 24 h, the cell proliferation rates of 0.2 and 0.4 mmol·L-1 groups were significantly decreased.Cardiomyocytes exposured to high palmitic acid (0.2 mmol·L-1) for 24 h showed an increase in theexpression of EPR related proteins (GRP78, CHOP, PERKphos, IRE1phos and ATF6) and Bax(P<0.05), while Bcl-2 expression was significantly reduced.Pretreatmented with EPR inhibitor pravastatin(10 mol·L-1) significantly increased high palmitic acid induced Bcl-2 expression (P<0.05) and significantly decreased high palmitic acid induced Bax expression (P<0.05).Conclusion Activated EPR might play an important role in treatment of high fatty acid induced myocardial injury.

6.Effect of transforming growth factor β1 on thyroid peroxidase activity and mRNA expression in primary porcine thyrocytes

Objective To observe the effect of transforming growth factor β1 (TGF-β1) on thyroid peroxidase activity and the expression of mRNA in primary porcine thyrocytes, and approach its possible mechanism. Methods In vitro cultured porcine thyrocytes were divided into 0 μg/L(control group), 2, 5, and 10 μg TGF-β1/L groups.After cultured for 72 h, the cell survival rate was measured with methyl thiazolyl tetrazolium (MTT) assay and the thyroid peroxidase (TPO) activity was measured by guaiacol method. The expression of TPO mRNA was determined cell survival rates of 2, 5, 10 μg/L groups(85.26%, 75.14% and 63.21%) were significantly lower(all P < 0.05 ).significantly decreased as compared with the control group [(2.143 ± 0.102), all P < 0.05)], and the TPO of TGF-β1, the lower the expression of TPO mRNA [(0.875 ± 0.078), (0.466 ± 0.044), (0.273 ± 0.007)] compared with the control group[(1.419 ± 0.148), all P < 0.05)]. Conclusions The inhibiting effect of TGF-β1 on primary cultured porcine thyrocytes may be related to its mechanism of inhibiting TPO activity and reduced mRNA expression.
7.Strategies for Generating Regulatory Dendritic Cells and the Clinical Application of Immune Tolerance

Dendritic cells (DCs) are potent antigen presenting cells with dual functions in immune response or im-mune tolerance. Regulatory dendritic cells (DCreg) have negative immunomodulation effects. They can inhibit T cell stimula-tory activity and induce immune tolerance. DCreg can be generated by the induction of drugs, cytokines and cell microenvi-ronment with different mechanisms. DCreg induced tolerance is of great clinical significance in organ transplantation and au-toimmune disease. Recently, several clinical trials have been conducted in type 1 diabetes and rheumatoid arthritis, with re-sults that emphasize the tolerance and safety of DCreg therapy. In this review, we will focus on the strategies for generating DCreg and the clinical application of immune tolerance.
8.Investigation and Analysis on the Label Information of Package Inserts of Pediatric Applicable Drugs of Our Hospital

OBJECTIVE:To provide reference for regulating pediatric clinical drugs. METHODS:Package inserts of pediatric drugs used by the outpatient,inpatient and emergency pharmacies of our hospital during January and March in 2015 were collected. The information about pediatric drugs in the package inserts,including drug name,indications or functions,specification,adminis-tration and dosage,was calculated and analyzed. 815 kinds of drugs applicable to adults and children in our hospital were studied to summarize and analyze the dosage form,specification and dose of pediatric applicable drugs. RESULTS:438 package inserts were collected,including 327 related to chemicals and biologicals among which those imported or produced by sino-foreign joint venture were labeled with more complete information about drug use for children compared to those made in China,and 111 rele-vant to Chinese patent medicines and natural medicines for which there were significantly insufficient information about drug use for children. The applicable drugs with the dosage form for children and those with the specification therefor respectively accounted for 51.17% and 31.65% of the above-mentioned 815 kinds of drugs,and were mainly available as injections,tablets or capsules, granules and oral liquid. The drugs labeled with the dose based on the age group of children such as infant,preschooler,school child and adolescent(1-18 years old)accounted for 61.24% of those with the specification for children,the drugs labeled with the dose for babies (28 d-1 year old) accounted for 26.74% thereof,and the drugs labeled with the dose for neonatus (younger than 28 d)accounted for 12.02% thereof. CONCLUSIONS:The label information and variety of pediatric applicable drugs are marked-ly insufficient. It is suggested that our country should formulate relevant policies,strengthen the development of pediatric drugs and improve the data for clinical use thereof to ensure the safety of clinical use for children.
9.The therapeutic effect of Arsenic Trioxide to the lupus nephritis and the effect of the abnormal expression of transforming growth factor-β1 in the nephridial tissue of MRL/lpr mice

Objective To investigate the roles of Arsenic Trioxide to the lupus nephritis in MRL/ lpr mice and the effect on the expression of TGF-pl in the renal tubule. Methods 45 MRL/lpr mice were chosen for such experiment, then separated in 3 different groups, including arsenic trioxide ( ATO) group, cyclophosphamide (CYC) group and sodium chloride (NS) group. Urine of these mice was kept after the treatment to detect the urine protein levels. The pathological changes and the expression of IgG were observed , and the complement 3 of the nephridial tissue as well as the TGF-pl were detected by immunohisto-chemistry. Result The levels of urine protein in ATO group and CTX group were lower than NS group after the treatment ( P <0.05). The expression of IgG along the glomerular mesangium and capillary loop in ATO and CTX groups was less than NS group ( P <0.05). The expression of C3 had no significant difference within three groups ( P > 0.05). The glomcrulus spherulous cells and the integral of activity in ATO group and CTX group were less than that in NS group ( P <0.05). TGF-β1 wildly presented in the epithelial cells endochylema of the nephric tubule, but it was rarely seen in glomcrulus in NS group. The expression in the nephric tubule of ATO group was less than NS group (20.28 ± 1. 90 vs 68. 23 ± 2. 87, P < 0.01) , and the expression in the nephric tubule in CTX group was less than that in NS group (23. 26 ± 1.71 vs 68. 23 ±2. 87, P <0. 01). Conclusion TGF-pl may take part in the immunopathogenesis of lupus nephritis in MRI/lpr mice. ATO can relieve the inflammation of nephric tubule by down regulation the expression of TGF-β1. ATO can also relieve the tissue damage of kidney in lupus nephritis through reducing the inflammation of glomcrulus and interstitial substance,as well as the expression of the immunocomplex.
10.Related Factors of Stress of Parents of Children with Intellectual Disabilities and Autism: A Comparative Study

Objective To study the related factors of stress of parents of children with intellectual disabilities and autism. Methods 45 parents of children with intellectual disabilities and 45 with autism were surveyed. Results and Conclusion The stress level was very high in the parents of both 2 kinds of children, especially in the parents with autism children (P<0.01). The stress level of parents of both 2 kinds of children correlated positively with the level of disability, and correlated negatively with the family income.
