The molecular modifications of Herpes Simplex Virus Type Ⅰ (HSV-1) proteins represented by acetylation and phosphorylation are essential to its biological functions.The cellular chromatin-remodeling/assembly is involved in HSV-1 associated gene transcriptional regulation in human cells harboring HSV-1 lytic or latent infections.Further investigation on these biological events would provide a better understanding of the mechanisms of HSV- 1 viral gene transcriptional regulation.
2.Role of Chinese Materia Medica Preparations in Hospital Chinese Traditional Medicine Work

OBJECTIVE:To discuss the role of Chinese materia medica preparations in hospital traditional Chinese medicine work for references of the development of Chinese materia medica preparations in hospital. METHODS: The importance and role of the research & development, preparation and use of hospital Chinese materia medica preparation in the development chain of Chinese medicine were analyzed. RESULTS & CONCLUSION: The research and preparation of hospital Chinese materia medica preparation make up for the deficiency of the industrial production of Chinese materia medica preparations and play a part in the inheritance and innovation of traditional Chinese medicine.
3.Rapid determination of monoamine neurotransmitters and their metabolites by high performance liquid chromatography

BACKGROUND: The determination of monoamine neurotransmitters and their metabolites is important in the experimental.research of Parkinson disease (PD) and experimental diagnosis of phaochromocytoma.OBJECTIVE: To develop a high performance liquid chromatography (HPLC) method for simultaneous and rapid determination of monoamine neurotransmitters and their metabolites which include epinephrine(E), norepinephrine(NE), dopamine(DA), 5'-hydroxytryptamine(5'-HT), 5-hydroxy3-indoleacetic acid(5-HIAA), homovanillic acid(HVA) and 3,4-dihydroxyphenylacetic acid(DOPAC).DESIGN: Case control observation and a randomized controlled animal experiment.SETTING: Department of Clinical Laboratory, Affiliated Hospital of Medical College, Qingdao University; Haiyang Branch, Affiliated Hospital of Medical College, Qingdao University.PARTICIPANTS: This experiment was carried out at Staff Room of Physiology, edical College, Qingdao University and Department of Clinical Laboratory, Affiliated Hospital of Medical College of Qingdao University from October to December 2003. ①Two patients with phaeochromocytoma were all confirmed by CT, MRI or pathologic section as well as physical sign. Twelve health volunteers were all university students and were excluded to suffer hypertension, endocrine system diseases and other diseases. ② Total 12 healthy female Wister rats weighed between 150 g to 180 g were chosen and randomly divided into control group and PD group with 6 rats in each.METHODS: The standard chromatograms of the standard solution of mixtures of seven substances (E, NE, DA, 5'-HT, 5-HIAA, HVA and DOPAC)were prepared and the precisions and recovery rates of the same samples were determined respectively. Urine samples of 24 hours obtained from 12 healthy volunteers and 2 patients with phaeochromocytoma were analyzed respectively by HPLC. The unilateral forebrain bundles in PD group were destroyed with 6-hydroxydopamine (6-OHDA) for monitoring release characteristics of monoamine neurotransmitters and their metabolites in healthy and marred caudate putamen nucleus striatum of normal and PD rats striatum.MAIN OUTCOME MEASURES: ①Determination of the precisions and recovery rates of monoamines. ②The contents of catecholamines in 24 hours urine of 12 health volunteers and 2 patients with phaeochromocytoma .③The release characteristics of monoamines and their metabolites in rats striatum. RESULTS: The results and analysis of catecholamines substances in 24hours urine were all obtained from 12 health volunteers and 2 patients with pheochromocytoma. There were no animals died in the experinment, and all of them were involved in the result analysis. ①There was a favorable linear relationship in the range of 2-1 000 μg/L for DA, E, NE and DOPAC, 10-1 000 μg/L for 5-HIAA, HVA and 5-HT for 30-1 000 μg/L with HPLC. The minimal detection limit of DA, E, NE and DOPAC was 2 μg/L, 5-HIAA 10 μg/L, HVA and 5-HT 30 μg/L. The coefficient of variation (CV) in the same batch was 3.0%-8.6%, and the CV in the different batchs was 3.8%-9.7%. The average rates of recovery of DA, NE, E and other 4 kinds of substances were 107.3%, 89.4%, 80.1% and 89.2%-95.7%, respectively.②The contents of CA in 24 hours urine of the 2 patients with phaeochromocytomas were higher than those of the 12 healthy volunteers. One of patient samples showed that E was mainly higher than that of control group. Other samples showed that NE was mainly higher than that of control group. Otherwise , E and NE were ten times higher than that of control group. ③The contents of DA and its metabolites such as DOPAC and HVA in intact sides of hemiparkinsonian rats had no significant changes ,while the contents of DA, DOPAC and HVA in lesion sides were significantly decreased compared with the controls (P < 0.01 ).There were no significant changes in the contents of 5-HT and its metabolites 5-HIAA. ④The turnover rates of DA (DOPAC/DA and HVA/DA) in intact and lesion sides of hemiparkinsonian rats were all higher than those of the controls(P < 0.05).CONCLUSION: The method we developed is suitable for the experimental research of normal and hemiparkinsonian rat models and for the diagnostic testing of phaeochromocytoma.
4.Effect of human growth hormone on cell cycle kinetics in colonic cancer cells

Objectives:The effects of human growth hormone (hGH) on cell cycle kinetics were studied in human colonic cancer cell line, LS-174-T cell. Methods: The cells were cultured in RPMI-1640 medium for 24 hours with hGH at concerntration of 50, 100, 200 ng/ml with or without cisplatin. Cell cycle kinetics was analyzed by flow cytometry. Results: The significant effects of hGH on cell cycle kinetics were found in the cell line. Percentage of S phase increase (P<0.01) and percentage of G2/M phase decreased (P<0.01).DNA Index (DI) didn't increase.The cells had a lot of apoptosis and percentage of S phase decreased (P<0.01) with addition of cisplatin. Conclusions: In vitro hGH may induce the proliferation of colonic cancer cells. Cisplatin may suppress the stimulatory effects of hGH on S phase and increase the apoptosis of colonic cancer cells.
6.An experiment on ectopia osteogenesis of rhBMP-2/nHA/Co compound membrane

Objective:To evaluate the ectopia osteogenesis ability of rhBMP-2/nHA/Co compound membrane.Methods:Two membranes were fabricated:(1) nHA/Co group(2) rhBMP-2/nHA/Co group.Both of them was evaluated after implantation under thigh muscle of nude mice for 10,20,30 days.Results:More bone-like structures in rhBMP-2/nHA/Co group were found,than that in nHA/Co group.Conclusion:Collagen and nano-HA are suitable sustained release systems for rhBMP-2 in animal experiment and the rhBMP-2/nHA/Co compound membrane can effectively induce ectopia osteogenesis.
7.Effects of curcumin on solid tumor and acute myeloid leukemic cells

Objective To investigate the effects of curcumin on the proliferation, cell cycle distribution, and apoptosis of the acute myeloid leukemic cell line HL 60 and the hepatocarcinoma cell line QGY. Methods MTT method was used to detect the biological activities of curcumin at different concentrations and at different time. The cell cycle distribution was analyzed by flow cytometry, and changes of the cellular ultrastructure were observed by electronic microscopy. Results Curcumin could inhabit the growth of HL 60 and QGY in dose and time dependent manners. IC50 values of curcumin at 72 h to HL 60 and QGY were 24.8 ?mol/L and 49.5 ?mol/L, respectively. The cell growth of HL 60 was arrested at S and G 2/M stages (apoptosis peak: 8.65%), and that of QGY was arrested at S stage (apoptosis peak: 10.84%). Curcumin could lead to fat degeneration in HL 60 cells and cell degeneration and necrosis of QGY cells, resulting in apoptosis of QGY cells. Conclusion Curcumin has inhibitory effect on the growth of HL 60 cells through its anti proliferation and on QGY cells through the induction of apoptosis of QGY cells.
8.Efficacy of laryngeal mask airway Supreme in patients undergoing gynecological laparoscopic surgery

Objective To assess the efficacy of laryngeal mask airway Supreme (LMA Supreme) used in patients undergoing gynecological laparoscopic surgery. Methods Eighty ASA Ⅰ or Ⅱ patients, aged 40-64 yr,weighing 50-70 kg, undergoing elective gynecological laparoscopic surgery, were randomized into 2 groups ( n = 40 each): LMA Supreme group (group S) and LMA ProSeal group (group P). Mallampati test was performed before operation in both groups. The patients were classified as Ⅰ or Ⅱ . LMA Supreme and LMA ProSeal were inserted in group S and P respectively after induction of anesthesia with midazolam 0.05 mg/kg, propofol 2 mg/kg, fentanyl 3 μg/kg and vecuronium 0.1 mg/kg. A gastric tube was inserted through the drain tube of the LMA. The number of attempt, success rate, LMA placement time, success rate of gastric tube placement, cuff volume, peak airway pressure, and airway seal pressure, BP, HR, ECG, SpO2, PET CO2 and complications were recorded. The duration of surgery and pneumoperitoneum, and emergence time were also recorded. The fiberoptic bronchoscope scores were assessed after successful LMA placement. Results There were no significant differences in the duration of operation and pneumoperitoneum, emergence time between the two groups (P > 0.05). The BP, HR,SpO2, PET CO2 and peak pressure were within the normal range during operation in both groups. The first attempt success rates of LMA placement and gastric tube placement were significantly higher, the LMA placement time was significantly shorter, and the fiberoptic bronchoscopic scores were significantly higher in group S than in group P ( P < 0.05). Conclusion LMA Supreme is helpful for ventilation during operation with less complications and can be used effectively for gynecological laparoscopic surgery.
9.Research on treatment of 120 cases of pulmonary Interstitial fibrosis with prednisone anti Shuxuening Injection

Objective To study the clinical effect ofprednisone combined with Shuxuening injection on pulmonary interstitial fibrosis. Methods 120 cases were randomly recruited into a treatment group and a control group. The control group was treated with prednisone, and the treatment group was additionally given Shuxuening injection on the basis of the control group. The changes of vital capacity (VC), PO_2, PCO_2 and imaging were observed. Results The total effective rate was 81.67% and 66.67% in the treatment group and the control group Respectively, showing significant difference (P<0.05). VC and PO_2 were raised in both groups after the treatment (P<0.05), while the treatment group showed better results than the control group (P<0.05). PCO2 were also raised in both groups after the treatment (P<0.05), while there was no significant diference between the two groups (P > 0.05). There were 27 cases (45.00%) and 19 cases (31.67%) showed improvement in imaging in the treatment group and the control group respectively, manifesting a significant difference (P<0.05) . Conclusion Treatment of prednisone with Shuxuening injection is better than prednisone on pulmonary interstitial fibrosis. This method is worthy of clinic promotion.
