Twenty-two patients survived for 18 months with g ood quality of life.ConclusionAccurate selection for donor and recipient, suitable surgical procedure, excelle nt myocardial preservation, effective protection of recipient's renal function, early prevention and management of right graft-heart dysfunction can significa ntly decline incidence of right heart failure at postoperative early stage.

3.Expression of connexin 43 in ovarian cancer and its relationship with chemoresistance

Objective To examine the expression of protein kinase C (PKC), connexin43(Cx43) and non-phosphorylated Cx43 in ovarian cancer, and discuss the role of phosphorylated of Cx43 in chemoresistance in ovarian cancer. Methods We examined the expression of Cx43, non-phosphorylated Cx43 and PKC in ovarian cancer tissue by immunohistochemistry, and compared their expression in chemosensitivity group and ehemoresistance group. Cisplatin resistant ovarian cancer cell line SKOV3/DDP cells were treated by staurosporine (a kind of PKC inhibitors). Then expression of Cx43, non-phosphorylated Cx43 and PKC were tested. Meanwhile, we tested chemosensitivity of SKOV3/DDP cells by ATP bioluminescence tumor chemosensifivity assay (ATP-TCA). Results (1) Immunohistochemically,the rates of positive expression of Cx43 and non-phospharylated Cx43 were 54%, 14% respectively in the chemoresistance group, which were 83%, 59% in the chemosensitivity group respectively (P<0.05). The rate of positive expression of PKC in 28 chemoresistance ovarian cancer cases (64%) was higher than that in 29 chemosensitivity cases (31% ,P<0.05). Both of them were significantly lower in ehemoresistanee group than in chemosensitivity group (P<0.05). In addition, the expression of PKC was negatively correlated with the expression of Cx43 and non-phosphorylated Cx43. The correlation coefficients were -0. 626 and -0. 714, respectively (P<0.05). (2) Immunohistochemically, PKC was down regulated, and Cx43 and non-phosphorylated Cx43 were up regulated in SKOV3/DDP cells after staurosporine treatment. The longer the staurosporine worked, the more expression of Cx43 was. (3) By ATP-TCA, SKOV3/DDP cells were resistant to paclitaxel and cisplatin. The tumor growth inhibition was higher in the group of paclitaxel or cisplatin combined staurosporine than in the group of paclitaxel or cisplatin alone. The sensitivity was intermediate in the group combined with low concentration staurosporine (1×10-8 moL/L), and the sensitivity was high in the group combined with high concentration staurosporine (1×10-7 mol/L). Conclusions Phosphorylation of Cx43 caused by PKC leads to decrease in the expression of Cx43. This effect makes ovarian cancer cells less chemosensitive. Phosphorylation of Cx43 caused by PKC can be inhibited by staurosporine.
4.Lower respiratory tract in patients with malignant tumor of fermentation bacteria infection of bacterial drug resistance analysis

Objective To understand the drug resistance in cancer patients with secondary non-fermenting bacterial lower respir-atory tract infection in order to provide a basis for clinical rational use of antibacterial drugs.Methods The lower respiratory tract specimens were collected from the patients with malignant tumor and identified by the fully automated microbial identification sys-tem,the drug susceptibility test was performed by using K-B method and the drug susceptibility test results were judged according to CLSI 2012 standard.The data were analyzed by the WHONET 5.6 software.Results 172 strains of non-fermenting bacteria were isolated from the lower respiratory tract specimens in the patients with malignant tumors,in which Pseudomonas aeruginosa was maximum,accounted for 45.9%,followed by Acinetobacter baumannii and Stenotrophomonas maltophilia ,accounted for 36. 0% and 10.5 % respectively.The drug susceptibility test showed that five kinds of non-fermenter demonstrated the high resistance or multi-resistance to multiple antibacterial drugs.Conclusion Non-fermenting bacterial multi-drug resistant phenomenon is seri-ous,clinic should pay attention to non-fermenting bacterial infection and drug resistance monitoring,antibacterial drugs should be rationally used according to the drug susceptibility test results in order to reduce the generation of drug-resistant strains.
5.The experiment study of several resin-removal methods on enamel surface

Objective To observe the effect of several residual adhesive methods on the enamel surface ,and conduct lab evalua-tion .Methods Sixty premolars extracted because of orthodontic treatment .And all the teeth were randomly divided into 3 groups . Group 1:tungsten carbide burs + silicon particles ;Group 2 :ultrasonic scaling + silicon particles ;Group 3:silicon particles ,each with 20 premolars .After underwent several methods ,the surface roughness differences ,operation time were determined and ob-served with the scanning electron microscope .And the result was statistically analyzed .Results There were significant differences in the surface roughness and operation time among the three groups (P<0 .05) ,The scanning electron microscope after polishing showed that the teeth surface had different degrees of injury ,the silica particles group had less superficial scratch .Conclusion The tungsten carbide burs and ultrasonic instrument for debonding before the silica particles had less superficial scratch .
6.Clinical characteristics and drug resistance analysis in ICU patients with hospital-acquired infections of Burkholderia cepacia

Objective To analyze the clinical characteristics and resistance to commonly used antibacterial drugs in the intensive care unit(ICU)patients with hospital-acquired Burkholderia cepacia infection in order to provide the basis for clinical rational treatment.Methods The drug resistance situation in 32 strains of Burkholderia cepacia isolated from the bacterial culture speci-mens submitted by ICU of our hospital from January 2011 to December 2012 was performed the retrospective analysis.Results In the detection sites,the infection was mainly distributed in the lower respiratory tract (62.5%),followed by deep venous catheter (12.5%);the drug susceptibility test revealed that 32 strains of Burkholderia cepacia showed the natural resistance to multiple antibacterial drugs,but which were still sensitive to minocycline,chloromycetin,meropenem,ceftazidime,cefoperazone/sulbactam, these drugs could be used as the first choice of drugs for treating Burkholderia cepacia infection.Conclusion The drug resistance phenomenon of Burkholderia cepacia is very serious in the ICU patients.Clinic should pay great concern to the infections caused by multi-drug resistant Burkholderia cepacia ,the microbiological testing should be conducted as early as possible,and the antibacterial drugs should be rationally selected according to the drug susceptibility testing results.
7.Impaired islet ?-cell function and insulin resistance in adult male rats born with intrauterine growth retardation

Objective: To investigate the islet ?-cell function and insulin sensitivity in adult male rats born intrauterine growth retardation caused by uterine placenta insufficiency. Methods: Bilateral uterine artery ligation (UAL) was performed on day 17 of gestation in the pregnant Wistar rats; sham-operated pregnant rats served as controls. The birth weights of the offspring in the UAL group were below the mean values of the birth weights of the control group more than 2SD, defined as IUGR. The tests were done in the adult male offspring (n=9 for each group). The glucose tolerance test was processed and Modified Beta-Cell function Index (MBCI) was calculated to evaluating the islet ?-cell endocrine function. Hyperinsulinemic- euglycemic clamp, the gold standard method to test the insulin resistance in the periphereal tissue was performed and insulin induced glucose infusion rate (GIR) was used to evaluate the insulin sensitivity. 9.Establishment of ISSR marker technology and optimization of its system in Prunella vulgaris

Objective To establish and optimize the ISSR-PCR reaction system for Prunella vulgaris and lay foundation for its genetic diversity research.Methods The single-factor and orthogonal design were applied for optimizing seven factors in the ISSR-PCR reaction system including Mg2+,dNTP,primers,Taq DNA polymerase,the template DNA,annealing temperature,and cycles.Results The suitable PCR reaction system contained 2.2 mmol/L Mg2+,175 ?mol/L dNTP,0.75 ?mol/L primer,1.0 U Taq DNA polymerase,and 30 ng template DNA in total 20 ?L reaction solution.On this basis,18 primers were screened with stable amplification and rich polymorphism from 92 ISSR primers.The optimal annealing temperature for ISSR-PCR reaction was proposed by gradient PCR.Conclusion It is a way to establisb the ISSR-PCR system for orthogonal design combining with single-factor test.And it is proved to be stable and credible for the result of 24 P.vulgaris populations.This optimized ISSR reaction system would provide the basis for the genetic analysis of P.vulgaris.
10.Effects of tolterodine on detrusor hyperreflexia for patients with spinal cord injury

ObjectiveTo evaluate the efficacy and tolerance of tolterodine in treating detrusor hyperreflexia in patients with spinal cord injury.Methods50 patients of spinal cord injury patients with detrusor hyperreflexia were involved in this study.The data from voiding diary were evaluated 7 days before and after the patients being treated with tolterodine 2 mg twice daily.ResultsAfter 12 weeks of treatment,the functional bladder volume increased from(114.0±44.3) ml to(207.7±54.9) ml,the mean frequency of micturition decreased from(8.9±2.7) /d to(4.6±2.5) /d,the mean volume of incontinence decreased from(646.7±348.9) ml to(426.6±291.3) ml,the mean frequency of incontinence decreased from(7.8±3.1) /d to(4.8±3.2) /d,which were all statistically different before and after treatment.5 patients felt dry mouth,but they could tolerated it.ConclusionTolterodine is an effective and well tolerable antimuscarinic agent for treating detrusor hyperreflexia after spinal cord injury.
