Objective:To observe the tansdifferentiation of bone marrow mesenchymal stem cells(MSCs) into cardiomyocyte in a coculture system with cardiomyocytes.Methods:Newly born Wistar Rats' cardiomyocytes were isolated and cultured and the cells were labeled with DAPI.Prepared MSCs were planted into the cardiomyocytes dishes to establish a co-culture system.Supernatant liquid of the cardiomyocytes or fibroblast was added the MSCs(non-labeled) dishes as the control. After one week of coculture,fluorescence microscope observation and immunocytochemical staining with cTnT was performed to evaluate the phenotype changes in non-labeled MSCs.Results:MSCs and cardiomyocytes were connect with each other by net in culture plate after a few days of coculture;some non-labeled cells were positively stained by cTnT antibody while the MSCs cultured with cardiomyocytes or fibroblast supernatant liquid were stained negatively.Conclusion:To obtain cardiomyocyte phenotype in vitro,direct contact of MSCs with cardiomyocytes is essential.The signaling molecules induce MSCs' transformation into cardiomyocytes are likely some membrane proteins on the surface of cardiomyocytes.

BACKGROUND:Implanted site and direction of miniscrew implants, post-implantation movement of implants, contact between tooth and implant after tooth movement al can cause root damage.
OBJECTIVE:To investigate the changes of the root surface, periodontal tissue and osteoprotegerin expression in the periodontal tissue at periods of time after miniscrew implant loading.
METHODTwo beagle dogs were selected. For each dog, miniscrews were implanted into the labial alveolar bone near the roots of the maxil ary second, third, and fourth premolars and the maxil ary second, third, and fourth premolars and first molar. Fourteen miniscrew implants were in one dog, including six in the maxil ary and eight in the mandible;two miniscrews of the maxil ary acted as control group with no loading, and the rest miniscrews were as experimental group under orthodontics force. Two weeks after miniscrew implantation, 150 g force was loaded between two miniscrews with nickel-titanium closed-coil springs. The dogs were sacrificed after miniscrews loading 4 and 8 weeks respectively, the teeth were cut completely with alveolar bone tissue and decalcified to make the histological specimen. The histological changes of the root and periodontal tissue were observed by hematoxylin-eosin staining and expression of osteoprotegrin in the periodontal ligament was also inspected by using immunohistochemistry.
RESULTS AND CONCLUSION:When the miniscrews were adjacent to the roots, absorption lacuna occurred in the alveolar bone;after horizontal force was loaded opposite to the roots, alveolar resorption became much more animate. When the miniscrews contacted with the pericementum, some areas of the cement absorbed seriously and even reached the dentin;when sustaining pressure from the miniscrews, the whole layer of the cement absorbed and the dentin was exposed and began to be absorbed. The osteoprotegrin in the periodontal ligament of control group expressed sharply and its expression was notable at 8 weeks;while the osteoprotegrin of experimental group weakly expressed at 8 weeks, and there was significant divergence between control group and experimental group at 8 weeks (P<0.01). The roots and periodontal tissue were al damaged variously after miniscrew implantation adjacent to the roots. When horizontal orthodontic force was loaded opposite to the roots, the expression of osteoprotegrin was influenced slightly;as the loading period prolonged, pressure dramatical y restrained the expression of osteoprotegrin. When discovering the miniscrew implant adjacent to roors, the miniscrew implant anchorage should not be used to load orthodontical force opposite to the roots but ought to be removed in order to leave the root repair autonomously and avoid further damage.

Objective To explore the clinical and pathological characteristics of singularity uterine leiomyoma.Methods The cause of disease and the clinical features in 15 patients with singularity uterine leiomyoma were analyzed.The patients were examined by pathology and immunohistochemistry.Results The main clinical manifestations were irregular vaginal bleeding,pelvic tumor and anemia.3 patients combined with pregnancy,4 patients had the treatment history of exogenous progesterone.The tumor appeared grayish red,and with complete envelope by macroscopic examination;The multi-and mono-nucleated giant cells were seen under surgical microscopy,the mitosis count was 0～3/10HPF.The tumors showed weak positive staining for Ki-67 and p53,negative staining for ER in 8 cases and positive staining for PR in all cases.Conclusions Singularity uterine leiomyoma belongs to benign tumor,its clinical features and treatments are similar to common leiomyoma;It has certain relation with exogenous and endogenous progesterone.The differentiation of leiomyoma with leiomyosarcoma is basically depended on HE staining observation;The immunohistochemistry of leiomyoma has certain feature.

Background and purpose:Tumor necrosis factor related apoptosis-inducing ligand (TRAIL) is a death receptor ligand that can induce apoptosis in a variety of cancer cell lines, but not all cancer cells . Past studies suggested that some chemotherapeutic drugs intensified the sensitivity of TRAIL induced apoptosis.We investigated the effect and potential mechanism of combined paclitaxel and TRAIL on apoptosis of human gastric cancer cell in vitro. Methods:SGC7901 cells were cultured with paclitaxel at different concentrations(1,5,10,20,40 ?g/ml) and different time(12,24,36,48 hr), methyl thiazolyl tetrazolium (MTT) assay was used to measure the anticancer activity of paclitaxel.The ability of TRAIL alone,paclitaxel alone and TRAIL in combination with paclitaxel to induce apoptosis was measured by a flow cytometer. Expression of DR4,DR5, DcR1,DcR2,Caspase-8 and Bcl-2 mRNA was examined by RT-PCR.Results:The apoptosis rates of control group(C group),paclitaxel group(P group),TRAIL group (T group)and combination group(T+P)in 24 hr were 2.09%,10.65%,7.79% and 24.51%, respectively. Objective To explore the characteristics of human thermal response in the natural cold environment.Methods Investigated and contrasted the skin temperature,thermal response characteristic and thermal comfort results of five women and fifteen men,aged 20 to 32 years,5 to 8 years of local living,in indoor warm environment and outdoor natural cold environment in the end of November,2007,in suburb of a city in Sinkiang(height 1 600 m,the lowest outdoor temperature in winter is-25℃ and the highest outdoor temperature in winter is 5℃,indoor average temperature is 18℃).The measurement on site and questionnaires were used.Results The mean thermal comfort vote(MTCV) mostly distributes around-3(mostly uncomfortable) when the outdoor temperature is-4.3--5.0 ℃,relative humidity is 65.2%-66.7%,The mean thermal sensation vote(MTSV) is-1.5(clod) to-3(quite cold).Human body could appear different degree of cold intensity sense and the body could not help quivering.Noses,ears and hands appeared different degree of rose color.Eyes became dryness and weep after staying long time outdoor.Conclusion Temperature,relative humidity and wind speed all around affected the mean thermal sense vote value and mean thermal comfort vote value.In the environment of low temperature,humidity and wind speed could increase the intension of cold action.Human is more sensitive to cold sense than thermal sense.

Objective:To develop an Online Game Cognitive Distortions Scale(OGCDS) and examine its applicability in Chinese adolescents.Methods:A pretest was given to 117 middle school students to determine a 17-item version of OGCDS.A sample of 495 middle school students were required to complete OGCDS,Internet Game Cognition Addiction Scale and Young's Internet Addiction Disorder scale.Reliability and validity of OGCDS were analyzed.Results:①The retest reliability,split-half and Cronbach's ? reliability coefficients of the total scale were 0.753,0.827 and 0.904,respectively.②Scores of the total scale and each subscale were both significantly correlated with scores of IGCAS and IAD scale at 0.01 levels (r=0.540～0.728;r=0.283～0.413).③Four factors were extracted by exploratory factor analysis,explaining 72.626% of the total variance;the load of each item varied from 0.490～0.886.④Confirmatory factor analysis confirmed that standard measurement model fit data well:?2=141.586,NFI=0.909,IFI=0.952,CFI=0.952,RMSEA=0.063.Conclusion:Reliability and validity of OGCDS is corresponded with psychometric standard.
