Purpose To explore the accuracy of ALK fused gene expression by immunohistochemistry ( IHC) in non-small cell lung cancer ( NSCLC) patients, and to investigate the clinical and pathological features of ALK-positive NSCLC patients. Methods By u-sing rabbit monoclonal D5F3 antibody, ALK IHC was performed on 234 NSCLC patients. ALK positive cases were confirmed by reverse transcription-polymerase chain reaction ( RT-PCR) . Results The positive incidence of ALK by IHC in 234 NSCLC specimens was 8. 97% (21/234), the positive rate of ALK fused gene verificated by RT-PCR was 5. 98% (14/234). There was significant difference with histological type, age, stage (P<0. 05), but no significant difference with gender, smoking history, tumor differentiation. Of 21 cases of ALK-positive NSCLC patients, the consistency of IHC and RT-PCR was 0 when IHC was ( +) , however, when IHC was or immunohistochemical score was >120, the consistency rate was 100%. Conclusion Although immunohistochemical expres-sion of ALK fused gene may have a certain false positive, IHC or immunohistochemical score> 120 show very high value for ALK fused gene RT-PCR followed by ALK immunohistochemistry in lung cancer is a economical and feasible method for the valuation of ALK fused gene.

Objective Sharing and exploring the efficient development of scientific research management with peers in large tertiary general hospital. Methods This article took the whole process management of scientific research in our hospital during the past five years as example, analyzed and elaborated the quality control and related safeguarding measures. Results According to the comparison between 2012 and 2016,the number of SCI papers published, the number of National Natural Science Funding and other important indicators all have been increased significantly in our hospital. Conclusions The implementation of the whole process quality control management and security measures of scientific research helps hospital to achieve initial and efficient development in research management.

BACKGROUNDTo evaluate the diagnostic value of flow cytometric DNA analysis of sputum cells in lung cancer patients.
METHODSThe DNA content of sputum cells was determined by flow cytometry in 44 patients (29 patients with lung cancer, and 15 patients with benign pulmonary disease). The results of DNA content of sputum cells were compared with those of cytologic morphology of sputum cells.
RESULTSThe last pathologic diagnosis was used as a diagnostic criterion, the sensitivity of DNA heteroploidy analysis was 82.8% , which was significantly higher than that of sputum cytology (82.8% vs 27.6%, P < 0.005); and the positive rate of heteroploidy in sputum samples in stage III and IV disease was significantly higher than that in stage I and II disease (87.5% to 76.9%, P < 0.002 5). In one case, the heteroploidy was found 10 months in advance of cytometry. In the control, heteroploidy was detected in 33.3% (2/6) patients with inflammatory pseudotumor and 50.0% (2/4) patients with tuberculous nodule, who were found with positive staining of Ki67 and PCNA.
CONCLUSIONSFlow cytometry seems to be a valuable adjunctive method for the diagnosis of lung cancer, and probably for the early diagnosis. There are clonal changes in some inflammatory pseudotumor and tuberculous nodule.
