Chinese Journal of Rehabilitation Theory and Practice 2007;13(5):410-411
Objective To evaluate the localization and the correlation with cardiovascular risk factors of cerebral arteries damage in patients with transient ischemic attack (TIA). Methods Digital subtraction angiography (DSA) was performed in the 97 patients with TIA. The localization, the degree of stenosis, the numbers of the arteries involved and their correlation with the cardiovascular risk factors were analyzed. Results DSA showed occlusive findings of cerebral arteries in 91.8% of the patients, and 68 cases (76.4%) among them were involved more than one artery. The proximal internal carotid arteries (35.5%) and the proximal vertebral arteries (28.2%) were the most frequently involved in this series. And the number of the arteries involved and the degree of stenosis were positively correlated with the level of risk factors. Conclusion There is a high incidence of cerebrovascular stenosis in patients with TIA. Most of them are involved more than one artery. The degree of the stenosis is positively related with the quantity of the risk factors.

Objective To investigate the course of the mandibular branch of the facial nerve and to discuss its clinical significant in the rhytidectomy. Methods The distribution of the mandibular branch was observed in 30 halves of the fifteen candaveric specimens (ten antiseptic cadaveric specimens and five fresh cadaveric specimens). Results The mandibular branch could be divided into the isolated branch type and shared branch type, after it exited from the parotid gland. 63.33 % mandibular branch was found (2.1?0.7) cm superior to the palpable edge of the mandibular bone; 23.33 % mandibular branch was along the edge of the bone; and 13.33 % was found (1.8?0.5)cm inferior to the palpable edge of the bone. Conclusion The distribution of the mandibular branch locates in the area that is a digit superior and inferior to the lower border of the mandibular bone, which arises from the angle of the mandibular bone. The dissection beneath the SMAS-platysma should be with caution of the injury of the ramification of mandibular branch in the anterior border of the masseter muscle.[

Objective To study the mechanism by which a high-intensity pulsed electromagnetic field (HIPEMF) (0.1 Hz,4 T,8 pulses) facilitates the proliferation of neural stem cells by detecting the expression of β-catenin genes and protein. Methods Neural stem cells (NSCs) were isolated from the sub-ventricular zone (SVZ) of neonatal rats and cultured in supplemented,serum-free medium for two weeks. The NSCs were then divided into an experimental group exposed to a HIPEMF for 8 pulses and a control group given sham stimulation. The gene and prorein expression of β-catenin in the NSCs were assayed by RT-PCR and Western blotting on the 1st,3rd,5th and 7th day after the stimulation. Results The NSCs' cloned spheres were round and translucent, and showed red fluorescence after staining with anti-nestin (cy3).The RT-PCR results showed β-catenin genes were highly expressed in the exposed group (significantly more than in the controls). The Western blotting showed that expression of β-catenin protein was also higher in the experimental group, especially at the 7th day after stimulation,a difference which was also statistically significant Conclusion HIPEMF at 0.1 Hz,4 T,in 8 pulse trains can promote NSC proliferation, perhaps through the Wnt/β-catenin signaling pathways.

Objective To observe expressions of mRNA for Par-4 and WT1 in bone marrow cells from acute leukemia patients and non-leukemia patients, and to approach the correlation between CR rate and Par-4, WT1 expression level. Methods To detect Par-4 and WT1 mRNA expression level in bone marrow cells from 78 acute leukemia patients and 23 non-leukemia patients by means of Real-time Fluorescent Quantitation RT-PCR. Results FQ-RT-PCR result showed that Par-4 mRNA was expressed in bone marrow cells from 78 acute leukemia patients and 23 non-leukemia patients. Compared with control groups, the expression levels of Par-4 mRNA were significantly suppressed (9.35×10-4±8.4×10-5, P <0.05). Compared with initial treatment groups and relapse groups, the expression levels of Par-4 mRNA in remission groups were significantly up-regulated (1.26×10-3±1.1×10-4) but were still significantly lower than that in control groups (3.25×10-3±2.9×10-4). There was no significance difference between initial treatment groups and relapse groups. No apparent association was found between Par-4 expression level and CR rate (P >0.05). WT1 gene was overexpressed in bone marrow cells from acute leukemia patients(2.98× 10-3±2.1×10-4), but the expression levels of WT1 mRNA were significantly lower in bone marrow cells from control groups (7.25×10-5±6.7×10-6,P <0.05). Compared with initial treatment groups and relapse groups, the expression levels of WT1 mRNA in remission groups were significantly down-regulated (6.86×10-4±5.2× 10-5) but were still significantly higher than that in control groups. There was no significant difference between initial treatment groups and relapse groups. There was significant difference between different WT1 expression levels and CR rates (P <0.05). Conclusion The result of FQ-RT-PCR testing confirmed that Par-4 mRNA expression is lower, while WT1 is higher in acute leukemia. Par-4 and WT1 gene present mutually exclusive expression patterns. There was no apparent association between Par-4 expression level and CR rate.

Objective:To investigate the expression and biological significance of MicroRNA-204 in nasopharyngeal carcinoma (NPC). Methods: qRT-PCR was applied to detect the relative expression of miR-204 in 43 paired nasopharyngeal carcinoma in comparison to the normal nasal mucosa tissues. Pearson chi-square test was used to analyze the relationship between the miR-204 expression and clinical features. The expressions of Bcl-2 and SIRT1 were measured by immunohistochemistry ( IHC ) , Spearman correlation analysis was used to analyze the relationship between miR-204 and Bcl-2,as well as SIRT1. We then transfected the miR-204 mimics into CNE-2 cells, then the Western blot was used to detect the expression of Bcl-2 and SIRT1,which were considered as the potential targets of miR-204. Results:The relative expressions of miR-204 was significantly downregulated in NPC tissues compared to those of the matched normal tumor-adjacent tissues(P<0. 05). Low expression of miR-204 was significantly associated with lymphatic metastasis(P<0. 05) and advanced TNM stage(Ⅲ+Ⅳ,P<0. 05). The expressions of Bcl-2 and Sirt1 in lower miR-204 level group were both higher than in higher miR-204 level group ( P<0. 05 ) . Both the mRNA and protin expression in CNE-2 cells were down-regulated after transfection. Conclusion: Low expression of miR-204 is related to the malignant clinicopathological features in NPC tissues, and miR-204 may through down-regulate Bcl-2 and SIRT1 to suppress NPC genesis and development.

Objective To explore the changes of Par-4 and WT1 genes expression during human leukemic K562 cells apoptosis induced by arsenic trioxide (As2O3). Methods After the K562 cells were treated with arsenic trioxide (2-10 μmol/L) for 24-72 hours, cell survival rate was evaluated by MTT assay and apoptosis was analyzed using flow cytometry. The expressions of mRNA and protein for par-4 and WT1 were tested by Real-time quantitative reverse transcriptase polymerase chain reaction (RQ-RT-PCR) method and Western blotting in K562 cells with various concentrations of arsenic trioxide at different time points. Results After the K562 cells were treated with As2O3 of different concentrations, arsenic trioxide could efficiently inhibit the growth of K562 cells and induce apoptosis with the increase of As2O3 concentration. The same time, the mRNA and protein expressions of Par-4 were up-regulated, while the mRNA and protein expression of WT1 were down-regulated. Conclusion Arsenic trioxide could inhibit the growth of K562 cells and induce apoptosis. Par-4 and WT1 genes could participate in the apoptosis of K562 cells induced by arsenic trioxide.
