1. Chemical constituents from plant of Stelleropsis tianschanica (II)

Objective: To further study the chemical constituents of Stelleropsis tianschanica. Methods: The constituents were isolated and purified by silica gel chromatography repeatedly, and the structures were identified by spectra analysis and chemical methods. Results: Twelve compounds including five lignans and three diphenylpentane derivatives were obtained, and named as kusunokinin (1), pluviatolide (2), dibutyl phthalate (3), dipropyl phtalate (4), p-dihydroxybenzene (5), daphneolon (6), 2(S)- hydroxy-1-(4-hydroxy phenyl)-5-phenyl-1-pentanone (7), (2R,3R)-1,5-diphenylpentane-2,3-diol (8). lariciresinol (9), isolariciresinol (10), matairesinol (11), and 4α,5βH-guai-9,7(11)-dien-12,8-olide-12,8α-diol (12). Conclusion: Compounds 1-12 are all obtained from the plant of S. tianschanica and the genus Stelleropsis Pobed. for the first time.

Objective: To study the chemical constituents from the arerial parts of Stelleropsis tianschanica. Methods: The constituents were isolated and purified by silica gel chromatography repeatedly, and the structures were identified by spectra analysis and chemical methods. Results: Thirteen compounds were isolated from S. tianschanica and the structures were identified as (+)-pinoresinol (1), (-)-pinoresinol (2), 3'-desmethylarctigenin (3), arctigenin (4), pluviatolide (5), umbelliferone (6), 4-(3,4- dimethoxybenzyl)-3-(4-hydroxy-3-methoxybenzyl)-tetrahydrofuran-2-ol (7), daphnogitin (8), daphnetone (9), blumenol B (10), loliolide (11), 4'-hydroxyacetophenone (12), and 4'-hydroxybenzoic acid (13). Conclusion: Compounds 1-13 were all obtained for the first time from the plant of S. tianschanica and the genus Stelleropsis Pobed.
3.The correlation analysis of the physical and mental status of organ transplanted patients' family members during the patients' rehabilitation period and the long term survival of the transplanted organ

ObjectiveTo know about the correlation analysis of the physical and mental status of organ transplanted patients' family members during the patients' rehabilitation period and the long term survival of the transplanted organs.MethodsA total of 600 organ transplanted patients' family members were investigated by Zung self-rating anxiety scale (SAS) and simple coping style questionnaire.The results underwent subsequent analysis.ResultsAnxious state of organ transplanted patients' family members was more severe than that of domestic norm (P<0.01).There were some differences in terms of anxious state of family members with different genders,education backgrounds,income and the resource of medical expenditure (P<0.05).Family members most took positive coping styles,whereas fewer adopted negative coping styles (P<0.01).Positive coping styles were negatively correlated with the anxiety of family members (P<0.01) and positively correlated with long term survival of transplanted organ(P<0.05).However,negative coping styles of family members were significantly positively correlated with their anxiety (P<0.05) and were negatively correlated with long term survival of transplanted organ.ConclusionsThe anxiety generally exists in organ transplanted patients' family members.There are differences in terms of anxious state among family members of different genders,different education backgrounds,income or with the resource of medical expenditure.The more they adopt positive coping styles,the lower anxiety level they show and the longer the transplanted organ survive.Conversely,the more they adopt negative coping styles,the higher anxious level they show and the shorter the transplanted organ survive.

5.hsa-miR-1908 target genes prediction and bioinformatics analysis

Objective To predict the biological process and signaling pathways in which hsa-miR-1908 might be in-volved by a series of bioinformatics analysis, so as to lay foundations and provide theoretical basis for the further studies of hsa-miR-1908 biological function in human preadipocytes. Methods The sequence of hsa-miR-1908 was acquired from miR-Base database, and target genes of hsa-miR-1908 were predicted by miRanda, and then the intersection of the results and the results of gene-chip as gene set were further analyzed by gene ontology and pathway enrichment. Results The hsa-miR-1908 had some conserved property among different species. The functions of the target genes were enriched in Wnt receptor signal-ing pathway through beta-catenin, cell cycle, cell apeptosis and other biological processes. The GnRH signaling, MAPK sig-naling, insulin signaling, cell cycle signal transduction pathways and signaling pathway in pancreatic cancer were signiifcantly enriched. Conclusions The target genes set of hsa-miR-1908 were enriched in multiple biological process which are related with the obesity. This study provides guidance for the further study in human preadipocytes.
6.Application of micro course in health education for prevention shoulder-hand syndrome in patients with stoke

Objective To explore the clinic effect of micro course in health education for prevention shoulder-hand syndrome in patients with stoke.Method The 42 stoke patients treated in our sector from October 2013 to September 2014 were observed (observation group) under health education by micro course,taking the 45 stoke patients treated in our sector from October 2012 to September 2013 as control group under traditional health education.Heahh education evaluation,occurrence rate of shoulder-hand syndrome,shoulder joint passive range of motion (ROM) and simple test for evaluating hand function (STEF) of the two groups of patients were compared after one and three months,respectively.Results Compared to the control group,the observation group made higher evaluation of health education method,showed lower occurrence rate of shoulder-hand syndrome,better shoulder joint passive ROM and upper limb movement function.In the time point of 1 and 3 months after the intervention,the incidence rate of shoulder-hand syndrome in the observation group was 2.5%(1/40) and 7.5%(3/40) respectively,which was significant lower than that of in the control group (19.05%,8/42 and 47.63%,20/40 respectively),thex2 value was 4.173 and 16.340 respectively,P<0.05.In the time point of 1 and 3 months after the intervention,the passive ROM in the observation group was (172.80 ± 15.18)° and (174.76 ± 7.45)°,which was significant higher than that of in the control group,(143.81 ± 22.76)° and (132.12 ± 16.67)° respectively,the t value was 6.816 and 15.074respectively,P<0.01.Conclusion Micro course used in health education of stroke patients could stimulate their study interests and activity,which played an important role in shoulder-hand syndrome occurrence prevention,shoulder injury reduction and upper limb function restoration acceleration.
7.Inductive effect of temozolomide on small cell lung cancer H446 cell apoptosis

Objective To explore the effect of temozolomide on apoptosis and molecular mechanism in small cell lung cancer (SCLC) cell H446. Methods The effect of temozolomide on the viability of H446 cell was measured by MTT assay.The effect of temozolomide on cell cycle was detected by flow cytometry.The activation of phosphatidyl inositol 3-kinase (PI3K)/AKT signaling pathway and expression level of downstream target genes (Cyclin B1), cell division cycle 2 (Cdc2), Bax, Bcl-2 and Survivin were detected by western blot.Results Temozolomide (50, 100, 200 μmol/L) could inhibit H446 cell viability, and the inhibitory rate was highest at 48 h.Moreover, temozolomide made H446 cell cycle arrested in G2 phase.Western blott showed the expression of PI3K, Cyclin B1, Cdc2, Bcl-2, Survivin and the phosphorylation of AKT were reduced, but the expression of Bax were increased by temozolomide.Conclusion Temozolomide could induce SCLC cell H446 apoptosis via blocking PI3K/AKT signal pathway.
8.Secretion of adipokines in miR-26b over-expressed human adipocytes differentiation process

Objective To investigate the inlfuence of overexpression of miR-26b on the secretion of adipokines dur-ing human adipocyte differentiation. Methods Human preadipocytes were infected with the hsa-miR-26b over-expressing lentivirus and were induced to differentiate, and then the levels of adipokines (IL-6, leptin, resistin, TNF-α) at different time points during differentiation were measured by ELISA. Results Compared with control group, decreased secretions of both IL-6 and leptin, and increased secretion of resistin were found during the differentiation of human adipocytes in miR-26b overexpressed group. However, the secretion of TNF-αwas not measured in both groups. Conclusion The miR-26b can improve the inlfammation and insulin resistance of human adipocytes, which will provide potential targets for obesity treat-ment.
9.Detection on Antibiotics-resistant Genes in Enterococci

OBJECTIVE To investigate the antibiotics-resistant genes in enterococci isolated from the 98th Hospital of PLA,Huzhou,Zhejiang Province,China.METHODS The antibiotics-resistant genes of TEM,aac(6′)/aph(2″),aph(3′)-Ⅲ,ant(2″)-Ⅰ,ant(4′,4″),ant(6)-Ⅰ,ermB,mefA,tetM,vanA,and vanB were analyzed by polymerase chain reaction(PCR) and verified by DNA sequencing in the 15 isolates of Enterococcus faecalis and 9 isolates of E.faecium.RESULTS The positive rate of the resistance genes of TEM,aac(6′)/aph(2″),aph(3′)-Ⅲ,ant(2″)-Ⅰ,ant(4′,4″),ant(6)-Ⅰ,(ermB,) mefA,tetM,vanA,and vanB in the 24 strains of enterococci tested were 37.5%,70.8%,25.0%,0.0%,0.0%,41.7%,75.0%,0.0%,41.7%,4.2%,and 4.2%,(respectively.) CONCLUSIONS The multidrug resistance of enterococci was a serious issue,and harbored antibiotics-resistance genes were the very important reasons of resistance to antibiotics in enterococci.
10.Effects of nitric oxide,endothelin-1, and liver sieve on biliary fibrosis in rats

Objective To study the effects of nitric oxide (NO) and endothelin-1(ET-1) on cholestasis liver fibrosis. Methods Forty-eight Wistar rats were randomily divided into 3 groups:the control group,the common bile dute ligation(CBDL)group,and the CBDL plus lactulose group.The animals were killed on the 3rd,7th,14th, and 21th days respectively to determine the contents of plasma endotoxin,NO,ET-1, and transaminases(ALT,AST).The degree of hepatic fibrosis was observed by microscopy and the ultrastructural changes of liver were observed by electron microscope. Results After common bile dute ligation,the contents of plasma ET-1 and NO increased significahtly,the plasma endotoxin level also increased and there were positive correlation between the levels of ET-1,and NO and the level of endotoxin.Plasma transaminases also increased greatly compared with those of the control group.The hepatic sinusoidal steonsis and widen Disse space were found.The diameter and number of fenestrate on hepatic sinusoid greatly decreased.Treatment with lactulose could partially alleviate all the pathological changes noted above. Conclusions NO and ET-1 may damage the "liver sieve"to cause the cholestasis liver fibrosis.
