Chinese Journal of Postgraduates of Medicine 2017;40(3):196-199
Objective To study the effect of gamma globulin on serum immune index in neonates with neonatal pneumonia. Methods One hundred neonates with neonatal pneumonia were selected, and they were divided into control group and observation group by random digits table method with 50 cases each. The control group was treated with conventional treatment, and the observation group was treated with conventional treatment combined with gamma globulin. The treatment efficacy, various signs disappearing times, length of stay in hospital and immune indexes were compared between 2 groups. Results The total effective rate in observation group was significantly higher than that in control group:96.00% (48/50) vs. 72.00% (36/50), and there was statistical difference (χ2 = 10.71, P<0.01). The disappearing times of cough, crackles, perioral cyanosis and fever and the length of stay in hospital in observation group were significantly shorter than those in control group:(4.39 ± 1.46) d vs. (6.32 ± 1.61) d, (4.21 ± 0.93) d vs. (5.36 ± 1.08) d, (0.83 ± 0.15) d vs. (1.16 ± 0.22) d, (2.53 ± 1.04) d vs. (4.47 ± 1.13) d and (7.68 ± 0.87) d vs. (10.03 ± 1.18) d, and there were statistical differences (P<0.01). The IgG, IgA, IgM, CD4+and CD4+/CD8+levels after treatment in observation group were significantly higher than those in control group: (10.32 ± 2.86) g/L vs. (6.88 ± 0.83) g/L, (1.14 ± 0.13) g/L vs. (0.91 ± 0.25) g/L, (0.94 ± 0.21) g/L vs. (0.85 ± 0.27) g/L, 0.516 ± 0.068 vs. 0.385 ± 0.063 and 1.89 ± 0.35 vs. 1.57 ± 0.33, but the CD8+was significantly lower than that in control group:0.303 ± 0.046 vs. 0.349 ± 0.048, and there were statistical differences (P<0.05). Conclusions The gamma globulin can not only significantly improve the curative effect in neonates with neonatal pneumonia and promote the recovery of the clinical symptoms, but also improve the efficiency of cell immunity and humoral immunity ability. It is a kind of effective treatment.

4.INDUCTION OF MYOCARDIUM CELLS FROM P19 CELLS USING DIMETHYL SULFOXIDE IN VITRO
Acta Anatomica Sinica 1954;0(02):-
Objective Observing the process of cell aggregation and differentiating into myocardium after the P19 cells treated with different methods and dimethyl sulfoxide(DMSO) inducer. Methods P19 cells were cultivated with DMSO in suspension for 7 days in Petri dishes or dishes containing a thin layer of soft agar to form cell aggregation. Then the aggregates were plated on gelatin-coated culture dishes, and cultured without DMSO to 19 days. The beating of cells was observed.?-sarcomeric actin and cardiac Troponin T(cTnT) immunocytochemical stains were used to identify cell differentiation. Results Cell aggregations were cohering-cultured with growth culture medium after DMSO induced for 7days,spontaneous and rhythmically beating cells were present within the aggregation outgrowths at day 15,which were ?-sarcomeric actin-positive and cTnT-positive.By day 19,positive rates were about 26% and 15% respectively. Conclusion By using DMSO exposure combination prior aggregation can induce P19 cells differentiate into cardiac myocytes which can spontaneously and rhythmically beat.

BACKGROUND: Although cultured technique and purified method of Schwann cells (SCs) are changed successively, purity is still limited. Whether double negative selections can culture high-purified SCs still needs a further study.OBJECTIVE: To investigate the effects of double negative selections on growth and purity of SCs.DESIGN: Controlled animal study.SETTING: Department of Otolaryngology Head and Neck Surgery, Beijing Tongren Hospital, Capital Medical University, and the Beijing Institution of Otolaryngology.MATERIALS: The experiment was carried out in the Department of Otolaryngology Head and Neck Surgery, Beijing Tongren Hospital, Capital Medical University and the Beijing Institution of Otolaryngology between May 2006 and July 2006. Twenty new-born Wistar rats of 3 days (without gender limitation) were provided by the Experimental Animal Center of the Chinese Medical Academy.METHODS: The sciatic nerves of rats were digested by 0.2% collagenase. ① Grouping: All the rats were divided into 4 groups with 5 each. The experimental group was depurated by differential adhesion and Ara-c for 30 minutes. SCs in the control group 1 and 2 were managed by differential adhesion and Ara-c for 30 minutes, respectively. Cells in the control group 3 were not purified. ② Evaluation: The cell growth was observed under invert microscope; growth curves were measured with MTT method in the experimental group and control group 3 to observe cell proliferation. In addition, the SCs were identified by immunocytochemistry; the culture purity was calculated (ratio of S100 positive cells in each sight).MAIN OUTCOME MEASURES: ① Growth of SCs; ② immunocytochemistry stain of SCs; ③ growth curve of SCs; ④ purity of SCs.RESULTS: ① Growth of SCs: At 5 days after incubation, the SCs proliferated well. SCs were spindle, bipolar and sometimes tripolar, with obvious nuclei and a little cytoplasm. There was more fibroblast cell in the group 3 than that in experimental group. ② Growth curve: The SCs in experimental group entered into the logarithm multiplication period at the third day. ③ Immunocytochemistry: The positive-stained SCs were brown in its body and process, and arranged like swirl. The FB with spread and anomaly shape was slight blue. ④ The ratio of anti-S100 positive cells was higher in experimental group than that in group 3 (P < 0.01).CONCLUSION: With the double negative selection, high-purified SCs are obtained effectively.
