1.Analysis of emulsion explosives residues by LC/MS

Objective:To use liquid chromatography/mass spectrometry to detect single-oleic acid sorbitan ester(SP-80)of emulsion explosives residues.Methods:Samples were collected after the ultrasonic cleaning solution with hexane,then filtered,and the filtrate were dried up with the rotary evaporator,dissolved in methanol,and then centrifuged.Chromatographic conditions were as follows:column phenomenex luna of C18 3?m(100?2.0 mm)(ID),pre-column Luna of C18 3?m(10?2.0 mm)(ID),mobile phase volume ratio of 1∶200 aqueous solution of acetic acid,isopropyl alcohol of 10∶90; flow rate of 0.20 ml/min,and column temperature of 35℃.MS parameter included drying nitrogen gas temperature of 200℃,nebulizer flow of 1.5 L/min,and detection voltage of 1.40 kV.Results:The linearity range of the assay for SP-80 was 0.2~1 mg/ml with correlation coefficient of 0.997 8,and the detection limit of 1?g/ml.3 extracts detected no significant difference.SP-80 were detected in two tapes emulsion explosives.Conclusion:The method of LC/MS can be used to infer the types of explosives for its operative simplicity, small sample requirement,high sensitivity,and wide linear range.

LncRNAs( long noncoding RNAs ) can be defined as RNA transcrips longer than 200nt without protein coding capacity , which play vital roles in various aspects of gene expression regulation.Recent investigations showed aberrant expression of many lncRNAs in primary liver cancer , indicating their important roles in tumorigenesis and development of liver cancer.This review focuses on the biological functions of lncRNAs and their involvements in primary liver cancer.
3.Granzyme B, B-cell lymphoma/leukemia-2 gene and BH3-interacting domain death agonist in cell apoptais

Granzyme B is an important effect factor of cytotoxic T lymphocytes in the immune killer function,can quickly induce cell apoptosis of the target cell.It is complicated for the reasons and the ways of the occurrence of the cell apoptosis,many genes are involved in gene regulation of cell apoptosis,and the B-cell lymphoma/leukemia-2 gene family members play a crucial role in the process of cell apoptosis.This paper mainly reviews the correlation of GrB and Bcl-2,Bid with cell apoptosis.
4.Analysis to Detecting Prostaglandin E2 and Elastase Levels in the Pulp Blood of the Patients with Carious Tooth Complicated with Exposed Pulp and Its Clinical Significance

Objective To observe and analyze the changes of prostaglandin E2 (PGE-2)and elastase (EA)levels in the pulp blood of the patients with carious tooth complicated with exposed pulp and its influence on the curative effects of direct pulp capping treatment.Methods 80 cases of the patients with carious tooth complicated with exposed pulp treated with direct pulp capping treatment were selected as the observation group and 40 cases of the patients treated with normal removal of impacted third molar were selected as the control group.The levels of PGE-2 and EA in the pulp blood of the patients in the two groups before the treatments were detected and compared.A one-year follow-up was employed for the patients in the observation group and the curative effects were evaluated and compared.Results The levels of PGE-2 and EA in the pulp blood of the patients in the observation group were 132.28±63.55 ng/ml and 114.36±19.57μg/ml respectively,and the levels of PGE-2 and EA in the pulp blood of the patients in the control group were 0.61±1.26 ng/ml and 19.38±6.34μg/ml respectively.There were statistically significant differences between the two groups (t=43.129,29.675,P<0.05). There was a positive relationship between the PGE-2 level and EA level in the pulp blood of the patients in the observation group (r=0.782,P<0.05).The levels of PGE-2 and EA in the pulp blood before the treatment of the patients with suc-cessful effects were 84.63±13.17 ng/ml and 68.25±8.16μg/ml,and the levels of PGE-2 and EA in the pulp blood before the treatment of the patients with failed effects were 177.35±41.42 ng/ml and 152.36±16.28μg/ml.There were statisti-cally significant differences between the two types of patients (t=31.578,21.556,P<0.05).Conclusion The patients with carious tooth complicated with exposed pulp show the significantly increased expression levels of PGE-2 and EA in pulp blood.The higher expression levels of PGE-2 and EA before the treatment could put adverse influence on the curative effects of direct pulp capping treatment.The clinicians should give the comprehensive consideration to the specific condition of pa-tients to choose the proper treatment methods.
5.Influences of Astragalus and Hurido effective components on apoptosis of rat glomerular mesangial cells

AIM To investigate the influences of Astragalus and Hurido effective components on apoptosis of rat glomerular mesangial cells.METHODS The proliferation of rat mesangial cells was induced by LPS and then intervened with Astragalus (astragalus polysaccharide,astragaloside Ⅳ,calycosin) and Hurido (hirudin).The cells were collected after 24 h.4',6-Diamidino-2-phenylindole (DAPI) staining was used to observe cell nuclear changes.Quantitative and qualitative expressions of Bcl-2 were detected by streptavidin-biotin complex (SABC).RESULTS Astragalus and Hurido effective components could induce the apoptosis of rat glomerular mesangial cells,and decrease the expression of Bcl-2.CONCLUSION Astragalus and Hurido effective components can induce the mesangial cell apoptosis by down-regulating Bcl-2 expression,which can be used for the treatment of mesangial proliferative glomerulonephritis.
6.Comparative study of spectral CT on the hemodynamic changes of different liver lobes in cirrhotic liver

Objective To explore the clinical application of material decomposition technique on spectral CT imaging and evaluate hemodynamic changes in different liver lobes with liver cirrhosis.Methods 30 patients with liver cirrhosis diagnosed clinically in our hospital were collected and underwent enhanced scanning of abdomen with spectral CT protocol.The monochromatic energy images and iodine-based material decomposition (MD) images were reconstructed after scanning.The iodine concentration (IC) was measured in five liver lobes (the caudate, left lateral, left inner, right anterior and right posterior lobes) and the abdominal aorta of the same axial slice in both the arterial phase (AP) and portal venous phase (VP) on the iodine-water based material decomposition images.The arterial iodine fraction (AIF) and the portal venous iodine concentration (PVIC) as well as the normalized iodine concentration (NIC) during the AP and VP were calculated.The differences of IC,the NIC,the AIF,and the PVIC in five liver lobes in AP and VP were compared by using single factor analysis of variance.Results The IC,the NIC in both AP and VP and the AIF of the caudate liver lobe were higher than those of other four liver lobes, with statistically significant difference (P<0.05), while these values in the other four lobes showed no statistically significant difference (P>0.05).The PVIC of the caudate liver lobe was slightly lower than that of the other four liver lobes, however, the difference was not statistically significant (P=0.929).Conclusion The quantitative iodine concentration measurement of liver lobes on spectral CT material decomposition technique can evaluate the hemodynamic changes in liver lobes with liver cirrhosis,and provide more information about the change of blood flow in liver cirrhosis.
7.Preparation and properties evaluation of methacrylic alginate gell beads cross-linked by mixed metal-cation.

The aim of this study was to obtain the alginate gels which could have proper compressive strength and excellent permeability for cell proliferation and could have more promising potentials in the application of tissue engineering. Through the reaction of the carboxyl of the alginate and the amino of methacrylic acid, methylacrylic was generated into alginate long chain which could be enhanced by the polymerization of double bond under thermal reaction condition. And then alginate gel beads were prepared using the mixture of calcium chloride and barium chloride solution as cationic crosslinker, and the compressive modulus and permeability of the prepared alginate gel beads were investigated. When the ratio of barium ions to calcium ions was 5:5, the compression modulus was 189.7 kPa, and it showed the best permeability for trypsin with molecular weight of 24 kDa and entrapment effect for bovine serum albumin with molecular weight of 67 kDa. Compared to compositions of other ratios, the alginate gel beads made in 5:5 mixture indicated excellent compressive modulus and permeability. These results indicated that the alginate hydrogel beads with the ratio of barium ions to calcium ions being 5:5 have a potential application in tissue engineering as a support material and encapsulating materials in cell culture.

8.Determination of Content and Dissolution of Levothyroxine Sodium Tablet by RP-HPLC

【Objective】To determine the content and dissolution of two imported levothyroxine sodium tablets.【Method】The content of levothyroxine sodium was assayed on a Hypersil BDS C18 column with a mobile phase consisting of methanol-0.85% phosphoric acid (60∶40) at a flow rate of 1.5 mL/min and detected at 225 nm.The study on dissolution was conducted according to the method Ⅱ of dissolution apparatus in USP(23).【Results】The average content of two formulations was 107.74% and 99.72%,respectively.The value of T50,Td and m of two tablets differed significantly (P<0.01).【Conclusion】There was a significant dissolution difference between two levothyroxine sodium tablets.
9.Establishment of 3-dimensional finite element model of maxillary in human complete unilateral cleft lip and palate with spiral CT scan

Objective To explore a faster and more precise method to establish a 3-dimensional (3 D) finite element model of maxillary in human complete unilateral cleft lip and palate. Methods The surface of the model was created using Materialists Interactive Medical Image Control System (Mimics) software to deal with Dicom standard files obtained by scanning the cranium of the patient with multi-slice helical CT. The 3D finite element model for complete unilateral cleft lip and plate in maxillary was established by Ansys software. Results A 3D finite element model of maxillary in human complete unilateral cleft lip and palate was constructed with 27 405 units and 26 876 nodes. Conclusion The combination of Mimics software, Geomagic studio software, Ansys software, and spiral CT is able to create a 3D finite element counter model, which provides a faster and more valid method to study complete unilateral cleft lip and palate.
10.Herpes Simplex Virus Type 1 ICP27 Protein: Its Expression, Purification and Specific Antiserum Production

Herpes simplex virus type 1 (HSV-1) is the causative agent of cold sores and other more serious diseases. HSV-1 infected-cell protein 27 (ICP27) is an immediate-early regulatory phosphoprotein homologous to gene products identified in all classes of herpesviruses so far. To raise the antiserum to ICP27 for further characterization of its biological function, the ICP27 gene was cloned into the pET-28a (+) vector, then ICP27 protein was expressed in E. Coli and purified by nickel-nitrilotriacetic acid (Ni2+-NTA) affinity resin column,finally the purified protein was used to raise antiserum. Western blot analysis demonstrated that the antiserum recognized the recombinant protein, and the antiserum was able to probe the ICP27 in HSV-1 infected cells with high specificity by immunofluorescence assay (IFA). Therefore, the specific antiserum will provide a valuable tool for further studies investigating ICP27's biological function during HSV-1 infection.
