1.Effects of Qiwei Qianliening Tablets on nonbacterial chronic prostatitis and its anti-inflammatory and analgesia activity

AIM: To investigate the effects of Qiwei Qianliening Tablets(QLN)(Radix paeoniae rubra,Semen Vaccariae,Semen Citri reticulatae,etc) on nonbacterial chronic prostatitis and its anti-inflammatory and analgesia activity. METHODS: Nonbacterial chronic prostatitis model was induced by injecting 25% Xiao Zhiling Injection into the prostate of rat and then coefficent of prostate and pathologic changes were observed.Anti-inflammatory effects were tested by cotton pelle-induced granuloma in rat.Analgesia activity was tested by hot-plate method. RESULTS: QLN inhibited the increase of prostate tissue weight and the histopathology showed resolving prostatitis in QLN-treated groups compared with the controls.Anti-inflammatory activity was confirmed in cotton pelle-induced granoluma in rat and QLN-treated groups extended the licking-paw latency in hot-plate method compared with the controls. CONCLUSION: Qiwei Qianliening Tablets has prevention and treatment effect on experimental nonbacterial chronic prostatitis.
2.Significance of NF-?B Activation Induced by LPS in Rat Kupffer Cells

Objective To explore the effect of NF-?B activation induced by lipopolysaccharide(LPS) in rat Kupffer cells(KCs). Methods KCs were divided randomly into three groups. A group was control group. KCs were co-cultured with LPS in B group. KCs were co-cultured with LPS and PDTC in C group. NF-?B activity of KCs was determined with EMSA. Content of I-?B protein in KCs was detected with western blot. Expression levels of TNF-? mRNA, IL-6 mRNA of KCs were measured with RT-PCR. Results LPS induced NF-?B activation of KCs, decreased I-?B content of KCs, and increased expression levels of TNF-? mRNA and IL-6 mRNA markedly. PDTC could diminish the above effects of LPS on KCs. Conclusions NF-?B activation induced by LPS may induce cell injury by promoting expressions of TNF-? mRNA and IL-6 mRNA in KCs. PDTC could decrease mRNA expression of proinflammatory mediators by suppressing excessive NF-?B activation in KCs.
3.The Role of Endotoxin During Hepatic Ischemia Reperfusion in Rats

Objective To explore the role of endotoxin during hepatic ischemia reperfusion (HIR) in rats. Methods Wister rats were randomly divided into endotoxin, HIR and HIR plus endotoxin (HIRE) groups. In the HIR group, hepatic reperfusion was performed after 60 minutes of ischemia by interruption of the arterial and portal venous blood supply to the left lobes and middle lobes of the rat liver. In the HIRE group, LPS(1.0mg/kg) was injected via the dorsum vein of rat penis after ischemia and reperfusion. In the endotoxin group, LPS (1.0mg/kg) was injected via the dorsum vein of penis in the rats without ischemia and reperfusion. The NF-?B activities in the liver tissue of the rats were determined with EMSA. TNF-? expression levels in the liver tissue of the were measured with immunohistochemical staining. Serum ALT levels of the rats were also measured. Results NF-?B activity increased after reperfusion and remained high 12h after reperfusion in HIRE group compared with endotoxin group and HIR group. Both TNF-? expression levels and serum ALT levels increased markedly after reperfusion in HIRE group. Conclusion Endotoxin during HIR could induce hepatic injury. Endotoxin induced liver injury possibly by activating NF-?B followed by the subsequent expression of proinflammatory mediators in the liver tissue.
4.Levels of serum hypersensitive C-reaction protein and its correlation factors in cerebral infarction patients with impaired glucose tolerance

Objective To observe the levels of serum hypersensitive C-reaction protein (hs-CRP) and its correlation factors in cerebral infarction patients with impaired glucose tolerance (IGT). Methods According to the result of glucose tolevance test (GTT),the 131 patients were divided into normal glucose tolerance group (NGT, n=41),IGT group (n=47),newly diagnosed type 2 diabetes mellitus (DM) group (n=43). The serum hs-CRP levels and some biochemical parameters were measured in 72 h after the onset. The common carotid artery(CCA) and carotid plaque were examined by color B type ultrasonography. Results The hs-CRP levels were gradually increased among NGT,IGT and DM groups. 5.Influence factors of intravenous urokinase therapy for acute cerebral infarction

Objective To investigate the baseline factors that influenced prognosis after intravenous thrombolytic therapy. Methods All 82 patients received intravenous urokinase (UK) for acute ischemic stroke. The modified Rankin scale score at 90 days was dichotomized into good outcome (mRS 0-1) and poor outcome (mRS 2-6) as the primary outcome measure. Multivariable logistic regression method was used to analyze independent predictors of good outcome. Results The mRS was good in 36.6% and poor in 63.4%. Univariate analysis indicated that age, time from stroke onset to treatment, atrial fibrillation, pretreatment ESS score, baseline serum glucose, early ischemic changes on baseline CT scans and dose of UK were shown correlated significantly with mRS. However, the multivariate logistic regression did select the ESS score, time to treatment, dose of UK and serum glucose as the independent predictors of good outcome. Conclusion Pretreatment ESS score, time to treatment, dose of UK and baseline serum glucose should be independently predictive of outcome in patients with acute cerebral infarction treated by intravenous UK.
6.Study on relationship between fat-rich diet-induced nonalcoholic fatty liver disease and endoplasmic reticulum stress in rats

Objective To establish the nonalcoholic fatty liver disease(NAFLD) model in SD rats and to investigate if endoplasmic reticulum stress(ERS) plays a role in the pathogenesis of NAFLD. Methods A total of 36 male SD rats were randomly divided into 4 groups: normal control group 1(N1,n=8), normal control group 2 (N2,n=8), model group 1(M1,n=10) and model group 2(M2,n=10). The two control groups were fed with normal diets, whereas the two model groups were provided diets enriched in fat (10% lard oil and 2% cholesterol). The rats in N1 and M1 groups were sacrificed at the 12th week, and those in N2 and M2 groups were sacrificed at the 20th week. The serum alanine aminotransferase (ALT), aspartate aminotransferase(AST), alkaline phosphatase (ALP), total protein (TP), triglycerides (TG), total cholesterol (TC), low-density lipoprotein (LDL), high-density lipoprotein (HDL) were measured. The steatosis, immflamation and fibrosis of the liver were observed with HE and Masson staining. The expressions of GRP78, CHOP and procaspase-12 mRNA were tested using real-time PCR and the activation of procaspase-12 protein was detected by Western blot. Results At the 12th week, the liver index and the serum levels of ALT, AST,ALP,TP,TC,LDL in M1 group were significantly higher than those in N1 group (P<0.01), but HDL level was significantly lower in M1 group than that in N1 group (P<0.01). At the 20th week, the concentrations of TC and LDL in M2 group were significantly increased in comparison with N2 group. The histochemical study revealed that the hepatic steatosis and inflammation in M1 and M2 groups were more serious than those in N1 and N2 goups(P<0.01). The slight fibrosis was seen in M2 group. At the 12th and 20th weeks, the expressions of GRP78,CHOP and procaspase-12 mRNA and protein in M1 and M2 groups did not differ in N1 and N2 groups. Conclusions The fat-rich diet might successfully induce NAFLD in rats and there is no ERS observed in the study, which suggests that ERS may not be involved in the pathogenesis of NAFLD.
7.Analysis of glucose dysbolism in patients with ectopic ACTH syndrome

The characteristics of glucose dysbolism in ectopic ACTH syndrome (EAS) were investigated by analysing the clinical data of 117 patients with Cushing syndrome (CS). The patients with CS were divided into 3 groups : EAS group (n = 9) , Cushing's disease (CD) group (n = 67) and adrenocortical adenoma (AA) group (n =41). Retrospective analysis of the data covered the prevalence of secondary diabetes, blood glucose, HbAlc, blood potassium, and blood ACTH and 24 h urinary-free cortisol (UFC) levels. Compared with CD and AA groups, the prevalence of secondary diabetes and blood glucose level were higher in EAS group, while HbAlc level was lower. Blood glucose level in patients with ectopic ACTH syndrome decreased to normal after tumor removal.

An indexing system is constructed,based on the implementation of prevention and control technology for chronic non-communicable diseases in community health service institutions,using both literature review and Delphi method,and using the Analytic Hierarchy Process (AHP) to calculate the weight of the indexing system.The system comprises three level-1 indexes,8 level-2 indexes and 33 level-3 indexes.The research proposed a transition from emphasis of therapeutic means to prevention,and emphasis on prevention and control technologies to the high-risk population in the prevention and control technology of chronic non-communicable diseases,along with development of techniques and measures to encourage behavioral changes of the population with chronic diseases.
10.Content Determination of Aciclovir Cream by Centrifugal Partition Chromatography

OBJECTIVE:To establish a method for the content determination of Aciclovir cream. METHODS:Centrifugal parti-tion chromatography(CPC)was conducted. The solvent system was hexane-acetonitrile-water(2∶1∶1,V/V/V),the injection vehicle was an aqueous solution of 5%Tween 80 and the volume was 1.0 ml;the flow rate was 5 ml/min;the wavelength was 254 nm. RE-SULTS:There was a good linear relationship between quality concentration and peak area in the range of 0.012 7-0.126 7 mg/ml (r=0.998 7). The RSD of precision,stability and reproducibility tests was all no more than 2.0% and the average recovery was 97.34%(RSD=0.90%,n=9). CONCLUSIONS:The method is with high precision and accuracy,and can be used for the content determination of principal components of Aciclovir cream.
