1.Analysis of paraplegia during transarterial chemoembolization in liver cancer

Objective To evaluate the pathophysiology of paraplegia during transarterial chemoembolization in liver cancer and investigate effective management and prevention for improving clinical situation and relieve major symptoms.Method 2758 patients accepted TACE procedure because of liver cancer(and/or combined with remote metastasis),demonstrated 4 cases suffering from paraplegia(3 males,1 female)since Mar.2003 to Feb.2005,with mean age of(51?14)years old,The operative records and the clinical features after chemoembolization were summarized in detail.Results The incidence of paraplegia was 0.145%,with major symptoms of dysesthesia and hypokinesise emerging within 4 hours after TACE,and most symptoms aggravating gradually within 24-48 hours untill paraplegia appeared,and then turned to stabilization about 5-7 days,with their body functions partial recovery in 2 months.Conclusion More attention should be paied to prevent ectopic embolization of spinal cord vessels originating from extrahepatic collateral arteries during TACE or TAE.

Objective To explore the expression of c-Fos protein and character of mossy fiber sprouting(MFS) in hippocampus of rat with febrile seizures(FS).Methods Thirty-six 21-day-old male Sprague-Dawley rats were randomly divided into FS group,febrile control(FG) group and normal control(NG) group.FS was established by hyperthermal bath.Immune histochemistry and(Timm′s) staining were used to examine the expression of c-Fos protein in CA1 region and MFS in CA3 region of hippocampus.Results Excessive expression of c-Fos protein presented in the hippocampal CA1 region of FS group.The surface area percentage of c-Fos protein of FS group[(2.26?0.23)%] was higher than that of FG group[(1.08?0.19)%] and NG group[(0.71?0.14)%],there were significant difference between FS group and the other two groups(?~2=10.48 P

4.Expression and significance of P-gp/mdr1 mRNA, MRP and LRP in non-Hodgkin's lymphoma.

OBJECTIVETo explore the expression and clinical significance of P-glycoprotein (P-gp)/mdr1mRNA, multidrug resistance-associated protein (MRP) and lung resistance protein (LRP) in newly diagnosed non-Hodgkin's lymphoma.
METHODSmdr1 mRNA of in 41 patients with non-Hodgkin's lymphoma was assayed by semi-quantitative RT-PCR. The expressions of P-gp, MRP and LRP proteins in lymph node viable blasts were identified by flow cytometry. The results were compared with those obtained from control cases, and the correlation of the changes with clinical outcomes was analyzed.
RESULTS(1) Among the 41 cases, the positive expression of P-gp protein was detected in 8 cases, MRP in 7 cases, LRP in 15 cases, and mdr 1 mRNA in 11 cases. (2) The P-gp and LRP levels in NHL were significantly higher than those in control group, but MRP wasn't. The P-gp over-expression was significantly associated with mdr1mRNA (r = 0.396, P = 0.01). No correlation was showed among the expressions of P-gp, MRP and LRP. (3) Patients with P-gp expression had a poorer outcome of chemotherapy than those with P-gp-negative (P = 0.005). P-gp expression was significantly associated with higher clinical stage (P = 0.046) and elevated serum lactate dehydrogenase level (P = 0.032), but not associated with malignant degree (P = 0.298). MRP had no impact on the outcome of chemotherapy (P = 0.212), and wasn't significantly associated with higher clinical stage (P = 0.369), elevated LDH (P = 0.762) and higher malignant degree (P = 0.451). Patients with LRP expression had a poorer outcome of chemotherapy than those LRP-negative (P = 0.012). LRP expression was significantly associated with higher clinical stage (P = 0.0019), elevated LDH (P = 0.02) and higher malignant degree (P = 0.01).
CONCLUSIONThe data of this study indicate that P-gp and LRP expressions but not MRP expression are important in the mechanism of drug resistance associated with a poor clinical outcome in previously untreated NHL.

5.Immunological and virological efficacy against HBV chronic infection of the therapeutic vaccine composed of HBV core plus PreS1 in HBV transgenic mice.

BACKGROUNDTo investigate the immunological and virological efficacy of the therapeutic vaccine HBV CS1, a recombinant fusion protein which is composed of HBV core aa 1-155 plus PreS1 aa 3-55,against chronic HBV infection.
METHODSHBV transgenic mice were immunized with HBV CS1(5 ug) emulsified in equal volume of complete Freund adjuvant on day 0, followed by a second vaccination with HBV CS1(5 ug) emulsified with incomplete Freund adjuvant on days 21. Mice of control group were mock-vaccinated with PBS plus complete Freund adjuvant/incomplete Freund adjuvant. The splenocytes of individual mouse were subjected to T cell proliferation assays by using 3Hg thymidine, HBsAg and HBV DNA in sera of mice were detected by ELISA and quantitative PCR, respectively.
RESULTSHBV CS1 specific T cell response were induced in mice immunized with HBV CS1, with the titer of HBsAg and the level of HBV DNA decreased significantly after twice immunization with HBV CS1, while the control group almost remained the same.
CONCLUSIONHBV CS1 has the immunological and virological efficacy against chronic HBV infection in HBV transgenic mice; HBV CS1 could represent candidate vaccine for further studies on its role as therapeutic vaccine against HBV chronic infection in human.

6.Effects of febrile seizures on motor, behavior, spatial learning and memory in rats.

OBJECTIVEFebrile seizure is a very common emergency in children. Although researchers home and abroad constantly pay close attention to studies on brain damage and lesion possibly caused by febrile seizure, studies of effects on motor, behavior, spatial learning and memory are relatively seldom. In our study, Sprague-Dawley rats were utilized for the purpose of the exploration of effects of febrile seizures on their motor, behavior, spatial learning and memory.
METHODSSixty 21-day-old male Sprague-Dawley rats, weighing (50 +/- 5) g were divided randomly and equally into febrile seizure group (FS), febrile control group (FG) and normal control group (NG). Febrile seizure animal model was induced by hyperthermal bath with 45 degrees C water. Febrile seizure was induced twice a day, thus ten times within five days in FS group. Rats of FG group were immersed in the same hyperthermal water for 2 minutes. Nothing special was performed on NG group. The abilities of motor and behavior of every rat in these 3 groups were tested in inclined plane test (IPT), overhanging test (OHT) and open field test (OFT) to show their varieties. Furthermore, Morris water maze was applied to evaluate the effects by febrile seizure on spatial learning and memory in rats during the place navigation test and spatial probe test.
RESULTSIn the present experiments, febrile seizures were altogether induced 192 times with the mean latency being (4.25 +/- 0.98) minutes and the mean duration being (1.06 +/- 0.59) minutes. The experiments confirmed that multiple febrile seizures could lead to decreases of abilities in all tests in which analysis of variance indicated that there were significant differences between febrile seizure group and the other two (P < 0.01). In inclined plane test, the turning ability of the rats was weakened. The mean turning time was (9.1 +/- 2.6) seconds for FS, (5.3 +/- 2.1) seconds for FG and (5.3 +/- 2.0) seconds for NG. In overhanging test, the overhanging time was shortened: (33.4 +/- 18.1) seconds for FS, (50.1 +/- 20.3) seconds for FG and (59.0 +/- 20.7) seconds for NG. In the open field test, the rats became less active with the scores (5.1 +/- 2.0) for FS, (10.4 +/- 3.0) for FG and (13.2 +/- 2.3) for NG. Meanwhile, the authors discovered the decreases of the abilities of spatial learning and memory in rats caused by febrile seizures many times. In the place navigation test, the mean escape latency for the rats' looking for hidden platform was prolonged; the efficiency of their search strategy decreased; the swimming time the animals spent in platform region decreased [(44.02 +/- 5.25) seconds for FS, (51.75 +/- 5.28) seconds for FG and (57.07 +/- 5.36) seconds for NG; analysis of variance, P < 0.01.]; the number of times they crossed the platform area decreased [(6.07 +/- 1.77) times for FS, (9.25 +/- 2.07) times for FG and (11.34 +/- 2.37) times for NG; analysis of variance, P < 0.01]; the percentage of their swimming time fell (36.68% for FS, 43.13% for FG and 47.56% for NG).
CONCLUSIONThe experiments confirmed that multiple febrile seizures could result in damage and lesion of motor, behavior, spatial learning and memory in rats.

7.Genetic polymorphism of dendritic cell-specific ICAM-3 grabbing nonintegrin and DC-SIGNR's exon 4 in Chinese hepatitis C patients.

OBJECTIVETo study into the genetic polymorphism of DC-SIGN and DC-SIGNR's exon 4 in Chinese hepatitis C patients and its relationship with HCV infection susceptibility.
METHODSPatients with hepatitis C (n=300, group A) and healthy subjects (n=520, group B) were genotyped and analysed for the repeat sequence of polymorphism of DC-SIGN and DC-SIGNR's exon 4 using PCR and DNA sequencing.
RESULTSThe distribution of genotypes and alleles in DC-SIGN's exon 4 in the two groups did not differ significantly (P > 0.05). The difference of allele frequency in DC-SIGNR's exon 4 between the two groups was also not significant (P > 0.05). However, 9/5 genotype distribution frequency of DC-SIGNR's exon 4 in patients with hepatitis C was significantly higher than that in the healthy subjects (P < 0.05).
CONCLUSIONThere is no significant correlation between the genetic polymorphism of DC-SIGN's exon 4 and HCV infection susceptibility. 9/5 genotype distribution frequency of DC-SIGNR's exon 4 in patients with hepatitis C is significantly higher and may be associated with HCV infection susceptibility.

8.Effect of silencing TRAF6 gene on proliferation, apoptosis and invasion of retinoblastoma Y79 cells

AIM: To investigate the effect of tumor necrosis factor receptor associated factor 6 (TRAF6) on proliferation, apoptosis and invasion of retinoblastoma Y79 cells. METHODS: The Y79 cells were divided into three groups:blank control group, negative control group and TRAF6 siRNA group. After TRAF6 siRNA transfection, the levels of TRAF6 mRNA and protein in Y79 cells were examined by RT-PCR and Western blotting. MTT assay was used to detect cell proliferation. Flow cytometry was employed to detect changes in cell cycle and apoptosis. Cell invasiveness was detected by the Transwell method. RESULTS: Expression of TRAF6 mRNA and protein in the TRAF6 siRNA group significantly decreased compared with the negative and blank control groups. Following the silencing of TRAF6,cell proliferation was inhibited and the apoptosis rate increased; the cell cycle was arrested at G0/G1 phase; the number of cells in S phase was reduced, while the invasion ability of cancer cells decreased. CONCLUSION: Silencing TRAF6 may inhibit the proliferation of Y79 cells, promote cell apoptosis, arrest the cell cycle at G0/G1 phase and decrease the invasive ability. Thus,TRAF6 may be a potential target in therapy for retinoblastoma.
9.Expansion of CD4~+ CD25~+ FoxP3~+ regulatory T cell pool in patients with active tuberculosis

Objective To investigate the frequency of CD4~+CD25~+FoxP3~+regulatory T cells (Treg)and the expression of the functional protein,FoxP3,in patients with active tuberculosis and the relationship between Treg and the pathogenesis of tuberculosis.Methods Forty-five patients with active tuberculosis(including 25 cases of pulmonary tuberculosis and 20 tuberculous lymphadenitis), 20 healthy controls,20 recovered tuberculosis patients and 6 patients with reactive hyperplasia in cer- vical lymph node were enrolled.The frequency of CD4~+ CD25~+ FoxP3~+ Treg in the peripheral blood was measured by flow cytometry.FoxP3 mRNA expression was determined by real-time reverse transcriptase-polymerase chain reaction(RT-PCR)and the expression of FoxP3 protein in lymphoid tissues was measured by immunohistochemistry.Results The frequency of natural Treg in the peripheral blood from the patients with active tuberculosis was 2.91%?0.23%,which was signifi- cantly higher than that of healthy control group(1.22%?0.18%)and recovered tuberculosis patients(1.50%?0.17%,P
10.Genetic polymorphism analysis of 3 short tandem repeat loci on chromosome 21 in Chinese Han population in Shaanxi Province.

OBJECTIVETo investigate the polymorphism of 3 short tandem repeat (STR) loci D21S1442, D21S1413 and D21S1809 on human chromosome 21 in Shaanxi Han nationality in China.
METHODSBlood samples anti-coagulated with EDTA were collected from 160 unrelated Chinese Han individuals in Shaanxi Province. The genomic DNA was extracted with Chelex-100, amplified by PCR and analyzed using PAGE with silver staining.
RESULTSNine, eight and seven alleles were detected at the loci of D21S1442, D21S1413 and D21S1809, respectively. The genotype distributions of all the 3 loci were in accordance with Hardy-Weinberg equilibrium. The heterozygosities of the 3 loci were 0.750, 0.742 and 0.694, respectively. No linkage disequilibrium was found between the loci.
CONCLUSIONThe 3 loci on chromosome 21 have high heterozygosity and polymorphic information content in Shaanxi Han population, suggesting their value as potentially useful genetic markers.
